Synthesis and mesomorphic properties of benzoxazole derivatives with lateral multifluoro substituents

Fluorinated aromatics is generally chosen as mesogenic cores to design novel liquid crystal compounds. Here, a series of benzoxazole derivatives with laterally multifluorinated biphenyl units, 2-(3 ‘,3-difluoro -4 ‘-alkoxy-1,1 ‘-biphenyl-4-yl)-benzoxazole derivatives (coded as nPF(3)PF(3)Bx), are synthesized and characterized, where methyl and nitro moieties are selected as terminal groups to investigate the effects of different polar substituents on the liquid crystal properties. The compounds nPF(3)PF(3)Bx show enantiotropic mesophases with mesophase ranges of 0-40 degrees C and 0-63 degrees C on heating and cooling for hydrogen-terminated derivatives (nPF(3)PF(3)BH), 43-93 degrees C and 54-123 degrees C for methyl-terminated ones (nPF(3)PF(3)BM), 60-108 degrees C and 74-152 degrees C for nitro terminated ones (nPF(3)PF(3)BN), respectively. They exhibit photoluminescence emission peaks at 390-392 nm and UV-vis absorption bands with maxima at 327-330 nm, respectively. The results reveal that lateral multifluoro substituents lead to a decrease in melting/clearing points, while electron-withdrawing terminal nitro moiety results in increases in both melting point and mesophase range. 2,5-bis(2 ‘-benzoxazolyl)hydroquinone (BBHQ), a dually fluorescent ESIPT system revisited: XRD analysis and supersonic jet studies of deuterated species

The new experimental results of the dually emitting ESIPT molecule, the 2,5-bis(2’-benzoxazolyl)hydroquinone (BBHQ) are presented. The results of the XRD analysis are the following: the unit cell of the monoclinic symmetry contains the perfectly planar BBHQ molecules, with two O-H center dot center dot center dot N intramolecular hydrogen bonds. There are two molecules in the asymmetric part of the cell, one the single BBHQ structure and the other being superposition of the main component with the site occupation factor (sof) 77.4(6)% and the minor one: sof = 22.6(6)%- the same structure rotated by 180 degrees along the long axis of the molecule. The remarkable agreement of the experimentally determined molecular structure with theoretical results reported in the literature, was observed. The dispersed fluorescence spectra of jet cooled molecule, covering two fluorescence bands were measured. It was observed that the intensity ratio of both emissions was dependent on excitation by subsequent quanta of the leading vibration of 114 cm(-1). The height of the barrier separating the two emitting tautomers was estimated as similar to 340 cm(-1). Moreover, the two independent experimental tools, the double-resonance fluorescence depletion (DFD) and mass selected two photon ionization (R2PI) techniques were applied to the partially deuterated jet cooled BBHQ enabling the separation and identification of four different deuterated species, some not known until now. Utilization of cyanoacetohydrazide and 2-(1,3-dioxoisoindolin-2-yl) acetyl chloride in the synthesis of some novel anti-proliferative heterocyclic compounds

Owing to its high reactivity and commercial availability, 2-cyanoacetohydrazide can be utilized as a versatile and appropriate intermediate for synthesis of a broad variety of heterocyclic compounds. Thus, 2-cyanoacetohydrazide and 2-(1,3-dioxoisoindolin-2-yl) acetyl chloride were used as starting materials for construction of new heterocyclic compounds bearing 1,3-dioxoisoindoline moiety. The newly synthesized compounds were recognized by elemental analyses and spectral data (IR,H-1-NMR, and(13)C-NMR spectra). The synthesized compounds were screened for their anti-proliferative activity against two human epithelial cell lines; breast (MCF-7) and liver (HepG2) as well as to normal fibroblasts (WI-38). The data showed distinctly that compounds1and12presented promisingin-vitroanti-proliferative activity against two cell lines (MCF-7 and HepG2) without harming normal fibroblasts.

Benzoxazole/benzothiazole-derived VEGFR-2 inhibitors: Design, synthesis, molecular docking, and anticancer evaluations

A novel series of benzoxazole/benzothiazole derivatives 4a-c-11a-e were designed, synthesized, and evaluated for anticancer activity against HepG2, HCT-116, and MCF-7 cells. HCT-116 was the most sensitive cell line to the influence of the new derivatives. In particular, compound 4c was found to be the most potent derivative against HepG2, HCT-116, and MCF-7 cells, with IC50 values = 9.45 +/- 0.8, 5.76 +/- 0.4, and 7.36 +/- 0.5 mu M, respectively. Compounds 4b, 9f, and 9c showed the highest anticancer activities against HepG2 cells with IC50 values of 9.97 +/- 0.8, 9.99 +/- 0.8, and 11.02 +/- 1.0 mu M, respectively, HCT-116 cells with IC50 values of 6.99 +/- 0.5, 7.44 +/- 0.4, and 8.15 +/- 0.8 mu M, respectively, and MCF-7 cells with IC50 values of 7.89 +/- 0.7, 8.24 +/- 0.7, and 9.32 +/- 0.7 mu M, respectively, in comparison with sorafenib as reference drug with IC50 values of 9.18 +/- 0.6, 5.47 +/- 0.3, and 7.26 +/- 0.3 mu M, respectively. The most active compounds 4a-c, 9b,c,e,f,h, and 11c,e were further evaluated for their VEGFR-2 inhibition. Compounds 4c and 4b potently inhibited VEGFR-2 at IC50 values of 0.12 +/- 0.01 and 0.13 +/- 0.02 mu M, respectively, which are nearly equipotent to the sorafenib IC50 value (0.10 +/- 0.02 mu M). Furthermore, molecular docking studies were performed for all synthesized compounds to assess their binding pattern and affinity toward the VEGFR-2 active site.

Design, synthesis and biological evaluation of a series of CNS penetrant HDAC inhibitors structurally derived from amyloid-beta probes

To develop novel CNS penetrant HDAC inhibitors, a new series of HDAC inhibitors having benzoheterocycle were designed, synthesized, and biologically evaluated. Among the synthesized compounds, benzothiazole derivative 9b exhibited a remarkable anti-proliferative activity (GI(50) = 2.01 mu M) against SH-SY5Y cancer cell line in a dose and time-dependent manner, better than the reference drug SAHA (GI(50) = 2.90 mu M). Moreover, compound 9b effectively promoted the accumulation of acetylated Histone H3 and alpha-tubulin through inhibition of HDAC1 and HDAC6 enzymes, respectively. HDAC enzyme assay also confirmed that compound 9b efficiently inhibited HDAC1 and HDAC6 isoforms with IC50 values of 84.9 nM and 95.9 nM. Furthermore, compound 9b inhibited colony formation capacity of SH-SY5Y cells, which is considered a hallmark of cell carcinogenesis and metastatic potential. The theoretical prediction, in vitro PAMPA-BBB assay, and in vivo brain pharmacokinetic studies confirmed that compound 9b had much higher BBB permeability than SAHA. In silico docking study demonstrated that compound 9b fitted in the substrate binding pocket of HDAC1 and HDAC6. Taken together, compound 9b provided a novel scaffold for developing CNS penetrant HDAC inhibitors and therapeutic potential for CNS-related diseases.

Synthesis and thermally induced structural transformation of phthalimide and nitrile-functionalized benzoxazine: toward smart ortho-benzoxazine chemistry for low flammability thermosets

A novel ortho-phthalimide-functionalized benzoxazine monomer containing an ortho-nitrile group has been synthesized in order to further systematically evaluate the thermally induced structural transformation from benzoxazine resin to cross-linked polybenzoxazole. The chemical structure of the synthesized monomer has been confirmed by H-1 and C-13 nuclear magnetic resonance (NMR) spectroscopy and Fourier transform infrared (FT-IR) spectroscopy. Also supporting the detailed structure is H-1-C-13 heteronuclear multiple quantum coherence (HMQC), which identifies the local proton-carbon proximities. The polymerization behaviors, including the ring-opening polymerization of the oxazine rings and the cyclotrimerization of the nitrile functionalities, are studied by differential scanning calorimetry (DSC) and in situ FT-IR. In addition, the subsequent benzoxazole formation after polymerization has also been analyzed using thermogravimetric analysis (TGA) and magic-angle spinning (MAS) solid-state C-13 NMR. The resulting cross-linked polybenzoxazole derived from the benzoxazine monomer exhibits exceptionally high thermal stability and low flammability, with an extremely high T-d5 temperature (550 degrees C), a high char yield value (70%) and an extraordinarily low total heat release (THR of 7.6 kJ g(-1)).

Development of benzoxazole deoxybenzoin oxime and acyloxylamine derivatives targeting innate immune sensors and xanthine oxidase for treatment of gout

Both the inhibition of inflammatory flares and the treatment of hyperuricemia itself are included in the management of gout. Extending our efforts to development of gout therapy, two series of benzoxazole deoxybenzoin oxime derivatives as inhibitors of innate immune sensors and xanthine oxidase (XOD) were discovered in improving hyperuricemia and acute gouty arthritis. In vitro studies revealed that most compounds not only suppressed XOD activity, but blocked activations of NOD-like receptor (NLRP3) inflammasome and Toll-like receptor 4 (TLR4) signaling pathway. More importantly, (E)-1-(6-methoxybenzo[d]oxazol-2-yl)-2-(4-methoxyphenyl)ethanone oxime (5d) exhibited anti-hyperuricemic and antiacute gouty arthritis activities through regulating XOD, NLRP3 and TLR4.
