Thermal induced structural changes of polyhydroxyamide by two-dimensional (2D) infrared correlation study

Two-dimensional (2D) correlation analysis of in situ MR spectra was used to probe the thermally induced structural changes in a poly(hydroxyamide) (PHA) precursor prepared from the reaction of 2,2-bis(3-amino-4-hydroxyphenyl)hexafluoropropane (Bis-AP-AF) with terephthaloyl chloride. Large spectral changes in the in situ FTIR spectra of the PHA precursor film were observed in the range of 200-300 degrees C. The thermal cyclodehydration reaction of the PHA precursor film strongly affects the spectral changes corresponding to the amide group and the adjacent phenyl ring in the Bis-AP-AF unit. The thermal cyclodehydration reaction of the PHA precursor film in the range of 240-300 degrees C induced the spectral changes in amide linkage before the formation of the benzoxazole ring. Regulatory Dendritic Cells Induced by K313 Display Anti-Inflammatory Properties and Ameliorate Experimental Autoimmune Encephalitis in Mice

As a GSK-3 beta inhibitor reported by our group, K313 is a novel benzoxazole derivative and displays anti-inflammatory properties in RAW264.7 macrophages without cytotoxicity. The activity of GSK-3 beta affects the differentiation and maturation of bone marrow-derived dendritic cells (DCs). This study aims to investigate whether K313 can be used to induce regulatory/tolerogenic dendritic cells (DCregs), and the therapeutic effects of DCregs induced by K313 in the autoimmune model of experimental autoimmune encephalitis (EAE). The results show that compared with LPS stimulated mature DCs, K313-treated bone marrow-derived DCs display obvious tolerogenic characteristics with decreased expression of co-stimulatory molecules, downregulated secretions of pro-inflammatory cytokines and unregulated secretion of anti-inflammatory cytokine IL-10. The above characteristics conform to the typical phenotypes of DCregs. Moreover, K313-modified DCregs inhibit antigen-specific T cell responses in vitro. Furthermore, by adoptive transfer, K313 modified DCregs to the EAE mice, and the development of disease was ameliorated to some extent. In addition, treatment with K313-modified DCregs also significantly reduced the percentages of splenetic Th1 and Th17 cells and increased the percentage of regulatory T cells in EAE mice. In conclusion, K313-modified DCregs show anti-inflammatory properties in vitro and have a significant positive effect on the EAE disease in vivo. Our data indicate that K313-induced DCregs pulsed with auto-antigen might have potential use as a therapeutic approach for autoimmune inflammation of the central nervous system.

Heterogeneous palladium (II)-complexed dendronized polymer: A rare palladium catalyst for the one-pot synthesis of 2-arylbenzoxazoles

The palladium complex of dendronized amine polymer (EG-Gn-Pd, n = 0, 1 and 2) having ethylene glycol-initiated polyepichlorohydrin as core was synthesized on a Merrifield resin support and was well characterized. Generally, palladium catalysts are known for carbon-carbon coupling reactions. Here, a developed catalyst was found to be good for benzoxazole synthesis. Higher generation dendronized polymer (EG-G2-Pd) was found to be better catalyst over lower generation dendronized polymers. Moreover, dendronized polymers were found to be a better catalyst over dendrigraft polymers. The catalyst reusability was checked and good yield was obtained for five cycles.

Mechanofluorochromism of difluoroboron beta-ketoiminate boron complexes functionalized with benzoxazole and benzothiazole

New difluoroboron beta-ketoiminate boron complexes bearing benzoxazole (CBO) and benzothiazole (CBS) have been synthesized. It was found that CBO was almost non-emissive in THF, and the emission could be intensified significantly when great amount of H2O was added, illustrating AIE (aggregation-induced emission) property. CBS could not show AIE property in THF/H2O system, but its emission in solid state was also strong. The single crystal structure of CBS suggested that pi-pi interactions and the hydrogen bonds of C(Ar) – F center dot center dot center dot H, C(Ar) – H center dot center dot center dot S and C(Ar) – H center dot center dot center dot Cl would suppress the rotation of single bonds, resulting into obvious emission enhancement. It is interesting that the as-synthesized crystals of CBO and CBS both emitted azure light, and the grinding made their emitting colors to change into cyan and green, which could be recovered under fuming with DCM or heating. The reversible mechanofluorochromism was due to the transformation between crystalline and amorphous states, which could be confirmed from the results of absorption spectra, XRD patterns and DSC curves in different solid states. Additionally, the high-contrast mechanofluorochromism of CBS compared with CBO might be originated from the loose packing in crystalline state and the better pi-electron delocalization.

Stabilization of different redox levels of a tridentate benzoxazole amidophenoxide ligand when bound to Co(iii) or V(v)

A tridentate benzoxazole-containing aminophenol ligand NNOH2 was coordinated to Co and V metal centers and the electronic structure of the resultant complexes characterized by both experimental and theoretical methods. The solid state structure of the Co complex exhibits a distorted octahedral geometry with two tridentate ligands bound in meridional fashion, and coordination-sphere bond lengths consistent with a Co(iii) oxidation state. EPR and magnetic data support a S = 1/2 ground state, and a formal electronic description of Co(iii)(NNOAP)(NNOISQ) where NNOAP corresponds to an amidophenoxide and NNOISQ to the iminosemiquinone redox level. However, the metrical parameters are similar for both ligands in the solid state, and DFT calculations support delocalization of the ligand radical over both ligands, affording an intermediate ligand redox level Co(iii)(NNO1.5-)(NNO1.5-). The vanadyl complex exhibits a distorted octahedral geometry in the solid state consistent with a V(v) metal center and amidophenoxide (NNOAP), acetylacetonate and oxo ligands. The ligand metrical parameters are consistent with significant amidophenoxide to V(v) pi donation. Overall, our results highlight the roles of electron transfer, delocalization, and pi bonding in the metal complexes under study, and thus the complexity in assignment of the electronic structure in these systems.

Virtual screening as a tool to discover new beta-haematin inhibitors with activity against malaria parasites

Malaria remains a major public health problem. With the loss of antimalarials to resistance, the malaria burden will likely continue for decades. New antimalarial scaffolds are crucial to avoid cross-resistance. Here, we present the first structure based virtual screening using the beta-haematin crystal as a target for new inhibitor scaffolds by applying a docking method. The ZINC15 database was searched for compounds with high binding affinity with the surface of the beta-haematin crystal using the PyRx Virtual Screening Tool. Top-ranked compounds predicted to interact with beta-haematin were submitted to a second screen applying in silico toxicity and drug-likeness predictions using Osiris DataWarrior. Fifteen compounds were purchased for experimental testing. An NP-40 mediated beta-haematin inhibition assay and parasite growth inhibition activity assay were performed. The benzoxazole moiety was found to be a promising scaffold for further development, showing intraparasitic haemozoin inhibition using a cellular haem fractionation assay causing a decrease in haemozoin in a dose dependent manner with a corresponding increase in exchangeable haem. A beta-haematin inhibition hit rate of 73% was found, a large enrichment over random screening, demonstrating that virtual screening can be a useful and cost-effective approach in the search for new haemozoin inhibiting antimalarials.

Preparation and characterization of novel polyimide/functionalized ZnO bionanocomposite for gas separation and study of their antibacterial activity

In the present investigation novel Polyimide/functionalized ZnO (PI/ZnO) bionanocomposites containing amino acid (Methionine) and benzimidazole pendent groups with different amounts of modified ZnO nanoparticles (ZnO NPs) were successfully prepared through ultrasonic irradiation technique. Due to the high surface energy and tendency for agglomeration, the surface ZnO NPs was modified by a coupling agent as 3- methacryloxypropyl-trimethoxysilane (MPS) to form MPS-ZnO nanoparticles. The ultrasonic irradiation effectively changes the rheology and the glass transition temperature and the crystallinity of the composite polymer. PI/ZnO nanocomposites were characterized by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), scanning electron microscopy (SEM) and transmission electron microscope (TEM). TEM analysis showed that the modified ZnO nanoparticles were homogeneously dispersed in polymer matrix. The TGA results of PI/ZnO nanocomposites showed that the thermal stability is obviously improved the presence of MPS-ZnO NPs in comparison with the pure PI and that this increase is higher when the NP content increases. The permeabilities of pure H-2, CH4, O-2, and N-2 gases through prepared membranes were determined at room temperature (25 degrees C) and 20 bar feed pressure. The membranes having 20% ZnO showed higher values of H-2 permeability, and H-2/CH4 and H-2/N-2 ideal selectivities (the ratio of pair gas permeabilities) compared with other membranes. The antibacterial activity of bionanocomposite films was tested against gram-positive bacteria (Staphylococcus aureus and Bacillus subtilis) and gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa). Further, it was observed that antibacterial activity of the resulting hybrid biofilms showed somewhat higher for gram-positive bacteria compared to gram-negative bacteria.
