Studies on New Schiff bases of Benzoxazole: Synthesis, Anticonvulsant and Neurotoxicity Evaluation

Twelve Schiff bases of benzoxazole were prepared by usage of methyl-3-amino-4-hydroxybenzoate. The chemistry of the prepared molecules was established based on the spectral data and tested for anticonvulsant activity using maximal electroshock (MES) induced seizure and subcutaneous pentylenetetrazole (scPTZ). In addition, a rotarod method to detect minimal neurological impairment in mice. In anti-MES test molecules 3d, 3e, 3i, 3j and 3k presented effective action corresponding to hydrophobicity. Other compounds of the series like 3b, 3c, 3g and 3l were remarkably less lipophilic and have some potencies. Compounds 3d and 3j effectively passed the rotarod trial without any mark of CNS deficit. In conclusion, the synthesized compounds with distal aryl groups showed higher hydrophobicity and resulted in better pharmacological action, which can be the future of new promising anticonvulsant drugs.

Zr and Hf-metal-organic frameworks: Efficient and recyclable heterogeneous catalysts for the synthesis of 2-arylbenzoxazole via ring open pathway acylation reaction

Zirconium- and hafnium-based metal-organic frameworks which constructed by 12-coordinated clusters and 6-coodinated clusters were shown to be highly effective heterogeneous catalysts for the ring opening acylation of benzoxazole to 2-arylbenzoxazole under solvent free conditions. Owning the wide opening spaces structures and inherent formate sites, MOFs based on 6-connected Zr-6/Hf-6 node were able to identify a significantly enhanced yield in Bronsted acid catalyzed reactions under conventional heating and microwave irradiation. Efficient synthesis of NIR emitting bis [2-(2 ‘-hydroxylphenyl)benzoxazole] derivative and its potential for imaging applications

Unassymetric bis[2-(2’-hydroxyphenylbenzoxole)] bis(HBO) derivatives with a DPA functionality for zinc binding have been developed with an efficient synthetic route, using the retrosynthetic analysis. Comparison of bis(HBO) derivatives with different substitution patterns allows us to verify and optimize their unique fluorescence properties. Upon binding zinc cation, bis(HBO) derivatives give a large fluorescence turn-on in both visible (lambda(em) approximate to 536 nm) and near-infrared (NIR) window (lambda(em) approximate to 746 nm). The probes are readily excitable by a 488 nm laser, making this series of compounds a suitable imaging tool for in vitro and in vivo study on a confocal microscope. The application of zinc binding-induced fluorescence turn-on is successfully demonstrated in cellular environments and thrombus imaging.

Synthesis, anticancer evaluation, and molecular docking studies of benzoxazole linked combretastatin analogues

A novel series of benzoxazole linked combretastatin derivatives (11a-11n) have been synthesized and confirmed by H-1 NMR, C-13 NMR, and Mass spectral analysis. The synthesized compounds (11a-11n) were screened for anticancer activity against three human cancer cell lines, Breast (MCF-7), Lung (A549), and Melanoma (A375). Most of the compounds exhibit moderate to potent anticancer activity. Among the compounds, 11g, 11h, 11l, 11m, and 11n showed more potent activity than the positive control Doxorubicin. In addition, compounds 11g, 11l, 11m, and 11n were carried out their molecular docking studies on EGFR receptor (PDB ID: 4hjo) and results indicated that 11g and 11l have strong binding interactions with the receptor. It was found that the binding energy calculations were in good agreement with the observed IC50 values.

Novel alanines bearing a heteroaromatic side chain: synthesis and studies on fluorescent chemosensing of metal cations with biological relevance

A family of novel thienylbenzoxazol-5-yl-L-alanines, consisting of an alanine core bearing a benzoxazole at the side chain with a thiophene ring at position 2, substituted with different (hetero)aryl substituents, was synthesised to study the tuning of the photophysical and chemosensory properties of the resulting compounds. These novel heterocyclic alanines 3a-f and a series of structurally related bis-thienylbenzoxazolyl-alanines 3g-j were evaluated for the first time in the recognition of selected metal cations with environmental, medicinal and analytical interest such as Co2+, Cu2+, Zn2+ and Ni2+, in acetonitrile solution, with the heterocycles at the side chain acting simultaneously as the coordinating and reporting units, via fluorescence changes. This behaviour can be explained by the involvement of the electron donor heteroatoms in the recognition event, through complexation of the metal cations. The spectrofluorimetric titrations showed that thienylbenzoxazolyl-alanines 3a-j and 4a,b were non-selective fluorimetric chemosensors for the above-mentioned cations, with the best results being obtained for the interaction of Cu2+ with bis-alanine 3j and deprotected alanines 4a,b. The encouraging photophysical and metal ion sensing properties of these thienylbenzoxazolyl-alanines suggest that they can be used to obtain bioinspired fluorescent reporters for metal ion such as peptides/proteins with chemosensory/probing ability.

Synthesis, characterization, and gas permeation properties of thermally rearranged poly(hydroxyimide)s filled with mesoporous MCM-41 silica

The gas transport properties of the mixed matrix membranes (MMM) prepared from the synthesized, structurally different poly(hydroxyimide)s (HPI) and mesoporous MCM-41 silica particles were investigated. The both series of filled and unfilled precursors were used to obtain and examine thermally rearranged (TR) polybenzoxazoles (PBO). The effect of a number and position of hydroxyl groups in HPI chains as well as that of filler presence on TR conversion was studied using FTIR, DSC, TGA, WAXD, SEM, mechanical testing, and density measurements. With 7 wt% of MCM-41 silica, gas permeability of the filled HPI precursors and that of the respective filled PBOs increased 1.5-2.2 and 4.4-15.1 times, respectively, comparing to the neat HPIs. The permeability increase along with relatively low loss of gas selectivity for filled membranes shift them towards the upper bound demonstrating that incorporation of porous fillers into polybenzoxazoles precursors can be a new approach to improve membrane performance.

Synthesis and biological activity evaluation of azacycloheptane sulfonamide derivatives as potential orexin receptor antagonists

As the orexin signaling system is crucial for the regulation of the sleep/wake cycle, inhibitors of orexin-1 and orexin-2 receptors are of significant interest in the treatment of insomnia. Herein, a series of novel azacycloheptane sulfonamide derivatives were designed and synthesized, and all the compounds were evaluated as potential orexin receptor inhibitors by FLIPR Tetra calcium assay. A majority of the tested azacycloheptane sulfonamide derivatives showed OX1R and OX2R inhibitory activity. Chloro-substituted derivatives functionalized at the C5 or C6 position of the benzoxazole group exhibited better inhibitory activity for OX1R and OX2R than unsubstituted derivatives functionalized at C5 or C6. In addition, phenyl group modification had positive effects on the inhibitory activities, and an electron-withdrawing fluorine group at theorthoormetaposition of the phenyl ring improved the OX2R inhibitory activity of the derivatives. This suggests that azacycloheptane sulfonamide derivatives are promising scaffolds for the development of OX1R and OX2R antagonists.

Laser-induced linear and non-linear optical features in novel benzoxazole-based on donor-acceptor chromophores

The photophysical properties of donor-acceptor based benzoxazoles were investigated by means of steady-state and time-resolved fluorescence techniques. Additionally, laser stimulated second-order optical effects (preliminary second harmonic generation (SHG) were studied. The studies show that the nitro-benzoxazole derivatives decorated with different alkylo/arylo amino groups dissolved in medium polar aprotic solvents: tetrahydrofuran and butyl chloride, exhibit dual fluorescence from the locally excited state ((LE)-L-1) and a highly polar charge transfer (CT) state. Ambient-temperature time resolved fluorescence investigations prove the bimodal kinetics of the excited-state electron transfer reaction (LE)-L-1 ->(CT)-C-1. The lack of any CT emissions in cyclohexane and highly polar acetonitrile is presumably due to an efficient inter-system crossing process or fast deactivation of a low-lying CT state. In addition, strong CT properties inspired us to check a possibility to vary the absorption of the first UV-Vis maxima under influence of the external bicolour coherent laser light and the related optical second harmonic generation (SHG). The two coherent beams of the infrared Er:glass laser at 1540 nm/770 nm formed gratings in the photopolymer solidified chromophore. The performed experiments are compared with the theoretically calculated electronic parameters within the B3LYP DFT approach. The presented data open novel type of materials, allowing to operate its transparency using external IR laser light in the near IR spectral range. (C) 2018 Elsevier B.V. All rights reserved.
