Finally, the phenol derivative, produced following protonation of the aryloxide, is proposed to act as a BrOnsted acid, which promotes an isothiourea-catalysed kinetic resolution of benzoxazole-derived dihydropyranones.

Tuberculosis (TB) is one of the major contagious diseases with high mortality which is caused by Mycobacterium tuberculosis (Mtb) pathogen. Due to the existing antibiotic resistance (MDR-TB) to tuberculosis, the demand for the development of new potential chemotherapy drugs is increasing. Herein, we report synthesis of two novel benzoxazole-based series, namely 2-phenyl benzoxazole sulfonamide and 2-piperidine-benzoxazole sulfonamides. These compounds were evaluated for their antimycobacterial activity against Mycobacterium tuberculosis H37Rv strain, using the microplate alamarBlue assay. Molecular docking studies were carried out to comprehend the binding mode of the compounds. It is evident from molecular docking studies and minimum inhibitory concentration assay (MIC) that 2-phenyl benzoxazole sulfonamide scaffold has a greater potential of antitubercular activity possibly by ENR inhibition (inhA inhibitors). Herein, a series of novel triptycene-containing thermally rearranged poly(benzoxazole-co-imide) membranes (TPI-PBOs) with enhanced mechanical properties and gas separation performance are reported. The effects of chemical structures and poly(benzoxazole-co-imide) composition on the fractional free volume, polymer chain packing, microcavity size and size distribution, mechanical properties, and gas transport properties have been comprehensively investigated. Due to the incorporation of triptycene-containing non-TR-able polyimide segments, the resulting poly(benzoxazole-co-imide) films exhibited good mechanical properties even after being treated at 450 degrees C. The incorporation of bulky and rigid triptycene units led to significantly improved fractional free volume and gas transport properties compared to previously reported poly(benzoxazole-co-imide)s. Several triptycene-containing poly(benzoxazole-co-imide) films displayed excellent gas separation performance for H-2/CH4, H-2/N-2, and CO2/CH4 gas pairs that exceeded the 2008 upper bounds.

Ulcerogenic activity with histopathological studies was performed, and the screened compounds demonstrated significant gastric tolerance than ibuprofen. Molecular Docking study was also performed with resolved crystal structure of COX-2 to understand the interacting mechanisms of newly synthesized inhibitors with the active site of COX-2 enzyme and the results were found to be in line with the biological evaluation studies of the compounds. Especially, compound 9 is more active than standard drugs ciprofloxacin and cefotaxime against E. coli isolate with a MIC value of 8 mu g/mL. Also new compounds are less active than fluconazole with a MIC value of 256 mu g/mL against C. albicans and its isolate except for compound 9 that shows better activity other compounds with a MIC value of >4 mu g/mL for their antifungal activity.

Background: A new series of benzoxazole analogues was synthesized and checked for their in vitro antibacterial, antifungal and anticancer activities. Results and discussion: The synthesized benzoxazole compounds were confirmed by IR, H-1/C-13-NMR, mass and screened for their in vitro antimicrobial activity against Gram-positive bacterium: Bacillus subtilis, four Gram-negative bacteria: Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, Salmonella typhi and two fungal strains: Candida albicans and Aspergillus niger using tube dilution technique and minimum inhibitory concentration (MIC) was noted in mu M and compared to ofloxacin and fluconazole. Human colorectal carcinoma (HCT116) cancer cell line was used for the determination of in vitro anticancer activity (IC50 value) by Sulforhodamine B assay using 5-fluorouracil as standard drug. Conclusion: The performed study indicated that the compounds 1, 10, 13, 16, 19, 20 and 24 had highest antimicrobial activity with MIC values comparable to ofloxacin and fluconazole and compounds 4, 6, 25 and 26 had best anticancer activity in comparison to 5-fluorouracil.

The reaction mechanism and the substituent effects of a base-free and catalyst-free synthesis of functionalized dihydrobenzoxazole have been investigated in detail by using the density functional theory (DFT) method. The possibility to afford crossover reaction products has been carefully excluded based on the extremely high barrier, which is well-consistent with the experimental results that the crossover products cannot be observed.
