Carbazoles and hydrazone-bridged thiazole-pyrrole derivatives are known to exhibit a wide range of biological activities including antimicrobial activity. This work is a further extension of their biological activities that identifies a total of 13 of these derivatives as new alpha-glucosidase inhibitors. The carbazole derivatives exhibited noncompetitive inhibition of the enzyme with the inhibitor possessing the 2-benzoimidazole substitution being the most potent in the series. Compounds possessing the 2-benzothiazole, 2-benzoxazole and quinoline groups were also found to be promising for enzyme inhibition. The hydrazone-bridged thiazole-pyrrole derivatives showed competitive enzyme inhibition with a number of groups responsible for potent enzyme inhibition including 4-nitrophenyl, 4-bromophenyl, and 4-methoxyphenyl groups. Moreover, the hydrazone derivatives with unsubstituted pyrrole ring were found to be more favorable to alpha-glucosidase inhibition than the ones possessing the methyl-substituted ring. The current work may provide new structural and functional diversity to drug discovery of promising alpha-glucosidase inhibitors as anti-diabetic drugs.

A new series of benzimidazole/benzoxazole linked beta-carbolines (9a-j) were rationally designed and synthesized by combining two different anti-cancer fragments. The new hybrid beta-carbolines are subjected to anti-cancer screening against four different human cancer cell lines such as MCF-7 (breast), A549 (lung), Colo-205 (colon) and A2780 (ovarian) by using standard MIT assay. These hybrid beta-carbolines exhibited significant and high fold anti-cancer activity against MCF-7 cell lines than reference standard. They are also proved to be effective against A549 and Colo-205 cell lines. Further, compound 9b, 9c from benzimidazole and 9i from benzoxazole series have exhibited maximum anti-cancer activity among these hybrid beta-carbolines. Later, all of the hybrid beta-carbolines were subjected to molecular interaction analysis against a few selected kinase targets such as cdc-like kinase (CLK-1 to CLK-4), epidermal growth factor reductase (EGFR) kinase, protein (ATR) kinase along with APC-Asef interface. The violin plot of binding energies of 9a-9j have suggested them as good kinase binders. Result interpretation suggested hybrid beta-carbolines as promising CLK binders. The gas transport properties of the mixed matrix membranes (MMM) prepared from the synthesized, structurally different poly(hydroxyimide)s (HPI) and mesoporous MCM-41 silica particles were investigated. With 7 wt% of MCM-41 silica, gas permeability of the filled HPI precursors and that of the respective filled PBOs increased 1.5-2.2 and 4.4-15.1 times, respectively, comparing to the neat HPIs. The permeability increase along with relatively low loss of gas selectivity for filled membranes shift them towards the upper bound demonstrating that incorporation of porous fillers into polybenzoxazoles precursors can be a new approach to improve membrane performance.

We previously reported 2-aminobenzoxazole analogue 1 as a potent ChemR23 inhibitor. The compound showed inhibitory activity against chemerin-induced calcium signaling through ChemR23 internalization in CAL-1 cells, which are cell lines of plasmacytoid dendric cells (pDCs). To address this issue, we performed optimization of ChemR23 inhibitors using CAL-1 cells endogenously expressing human ChemR23 and conducted a pharmacokinetics study in cynomolgus monkeys. Various substituents at the 4-position of the benzoxazole ring exhibited potent in vitro bioactivity, while those at the 6-position were not tolerated. Among substituents, a carboxyl group was identified as key for improving the oral bioavailability in cynomolgus monkeys. Compound 38a with the acidic part changed from a tetrazole group to a 1,2,4-oxadiazol-5-one group to improve bioactivity and pharmacokinetic parameters exhibited inhibitory activity against chemerin-induced chemotaxis in vitro. In addition, we confirmed the ChemR23 internalization of pDCs by compound 38a orally administered to cynomolgus monkeys. These 2-aminobenzoxazole-based ChemR23 inhibitors may be useful as novel immunotherapeutic agents capable of suppressing the migration of pDCs, which are known to be major producers of type I interferons in the lesion area of certain autoimmune diseases, such as systemic lupus erythematosus and psoriasis.

As the orexin signaling system is crucial for the regulation of the sleep/wake cycle, inhibitors of orexin-1 and orexin-2 receptors are of significant interest in the treatment of insomnia. Herein, a series of novel azacycloheptane sulfonamide derivatives were designed and synthesized, and all the compounds were evaluated as potential orexin receptor inhibitors by FLIPR Tetra calcium assay. A majority of the tested azacycloheptane sulfonamide derivatives showed OX1R and OX2R inhibitory activity. Chloro-substituted derivatives functionalized at the C5 or C6 position of the benzoxazole group exhibited better inhibitory activity for OX1R and OX2R than unsubstituted derivatives functionalized at C5 or C6. In addition, phenyl group modification had positive effects on the inhibitory activities, and an electron-withdrawing fluorine group at theorthoormetaposition of the phenyl ring improved the OX2R inhibitory activity of the derivatives. This suggests that azacycloheptane sulfonamide derivatives are promising scaffolds for the development of OX1R and OX2R antagonists.
