A methacrylamide containing the n-dodecyl group was selected as a suitable monomer to obtain beneficial thermally conductive polymeric materials because of its sufficient solubility in toluene in the subsequent radical polymerization and because of its liquid crystallinity. This methacrylamide led to a highly soluble polymer with film-forming ability. The BO-containing polymethacrylamide also exhibited liquid-crystalline (LC) behavior during the heating and cooling processes. The cast film was kept at a slightly lower temperature than the clearing temperature and quenched to room temperature. This procedure afforded a film maintaining optical anisotropy at room temperature, implying that a frozen LC structure is maintained at room temperature. The quenched film also resulted in a significantly enhanced thermal conductivity (lambda = 0.60 W m(-1) K-1) without the aid of fillers and external fields, unlike the as-cast counterpart without optical anisotropy. A polyacrylate containing a flexible long alkylene (C-6) spacer between the main chains and a BO-containing mesogenic side group was also investigated. The polyacrylate exhibited LC behavior during the heating and cooling processes over wide ranges. The polyacrylate film quenched from an established temperature, which contains a frozen LC structure, also exhibited a significantly enhanced lambda value (0.63 W m(-1) K-1) without fillers and external fields. The polyhydroxyamide copolymers (F-PHAs) were synthesized by low-temperature solution polycondensation of 2,2-bis(3-amino-4-hydroxyphenyl) hexafluoropropane (BAHHFP) and/or 3,3 ‘-dihydroxybenzidine (DHB) with terephthaloyl chloride (TPC) in N,N-dimethylacetamide (DMAc) with the aid of lithium chloride (LiCl). A series of polybenzoxazole copolymer (F-PBO) films were fabricated via an efficient solution-casting and thermal-treatment technique, using their precursors (F-PHA films) with different BAHHFP fractions (25-100 mol%). The solubility results show that F-PHAs with higher BAHHFP content (>= 75 mol%) are readily soluble in anhydrous DMAc and N-methyl-2-pyrrolidinone (NMP) without LiCl at room temperature. Differential scanning calorimetry (DSC) indicates that the thermal cyclization temperature of the F-PHA films decreases with increasing BAHHFP content. Thermogravimetric analysis (TGA) reveals that the F-PHA and F-PBO films are well prepared and completely converted. The UV-visible spectra indicate that the F-75-PBO film (BAHHFP content: 75 mol%) has reasonable transparency, with 84.9 % transmittance at 600 nm and 397-nm UV cut-off wavelength. It also has lower coefficient of thermal expansion (CTE) (48 ppm/degrees C) and water absorption (WA) values (0.14%). The BAHHFP as diamine component provides the final F-PBO film with desirable properties, e.g., flexibility, thermal stability, transparency, and WA values.

Mutations in RAS/RAF occur in large portion of malignancies and are associated with aggressive clinical behaviors and poor prognosis. Therefore, we developed a novel benzoxazole compound (KZ-001) as a highly potent and selective MEK 1/2 inhibitor. Our efforts were focused on enhancing the activity of the known MEK inhibitor AZD6244 and overcoming the shortcomings existing in current MEK inhibitors. Here we show that compound KZ-001 exhibits approximately 30-fold greater inhibition against BRAF- and KRAS-mutant tumor cells than that of AZD6244. These results were also demonstrated using in vivo xenograft models. Furthermore, pharmacokinetics (PK) analysis was performed for KZ-001, and this compound showed good orally bioavailability (28%) and exposure (AUC(0-infinity) = 337 +/- 169 ng h/mL). To determine its potential clinical application, the synergistic effect of KZ-001 with other agents was investigated both in vitro and in vivo (xenograft models). KZ-001 exhibited synergistic anti-cancer effect in combination with BRAF inhibitor vemurafenib and a microtubule-stabilizing chemotherapeutic agent docetaxel. In addition, KZ-001 inhibited the MAPK pathway like known MEK inhibitors. In summary, KZ-001, a structurally novel benzoxazole compound, was developed as a MEK inhibitor that has potential for cancer treatment.

We report a new mononuclear molybdenum(iv) complex, (MoOLLSQ)-L-BIS, in which L-SQ (2,4-di-tert-butyl o-semibenzoquinone ligand) has been prepared from the reaction of the o-iminosemibenzoquinone form of a tridentate non-innocent benzoxazole ligand, L-BIS, and MoO2(acac)(2).
