Thymoquinone (TQ), a natural compound with antimicrobial and antitumor activity, was used as the starting molecule for the preparation of 3-aminothymoquinone (ATQ) from which ten novel benzoxazole derivatives were prepared and characterized by elemental analysis, IR spectroscopy, mass spectrometry and NMR (H-1, C-13) spectroscopy in solution. The crystal structure of 4-methyl-2-phenyl-7-isopropyl-1,3-benzoxazole-5-ol (1a) has been determined by X-ray diffraction. All compounds were tested for their antibacterial, antifungal and antitumor activities. TQ and ATQ showed better antibacterial activity against tested Gram-positive and Gram-negative bacterial strains than benzoxazoles. ATQ had the most potent antifungal effect against Candida albicans, Saccharomyces cerevisiae and Aspergillus brasiliensis. Three benzoxazole derivatives and ATQ showed the highest antitumor activities. The most potent was 2-(4-fluorophenyl)-4-methyl-7-isopropyl-1,3-benzoxazole-5-ol (1f). Western blot analyses have shown that this compound inhibited phosphorylation of protein kinase B (Akt) and Insulin-like Growth Factor-1 Receptor (IGF1R ) in HeLa and HepG2 cells. The least toxic compound against normal fibroblast cells, which maintains similar antitumor activities as TQ, was 2-(4-chlorophenyl)-4-methyl-7-isopropyl-1,3-benzoxazole-5-ol (1e). Docking studies indicated that 1e and 1f have significant effects against selected receptors playing important roles in tumour survival.

Metabolic pathways mediated by human gut bacteria have emerged as potential therapeutic targets because of their association with the pathophysiology of various human diseases. The anaerobic transformation of choline into trimethylamine (TMA) by gut microbiota is directly linked to type 2 diabetes, fatty liver disease, and cardiovascular diseases. Structural analogs of choline have been developed as competitive inhibitors of choline TMA-lyase (CutC), a key enzyme for the conversion of choline to TMA. However, weak to moderate CutC inhibitory profiles of the choline analogs limit their further advancement into clinical translation. In this study, we introduce a glycomimetic-based approach for the identification of CutC inhibitors with intestinal metabolic stability. Our workflow started with screening of a small library of glycomimetics for metabolic stability in the presence of human intestinal S9 fraction. Further screening using an in vitro CutC inhibitory assay identified a benzoxazole ligand (BO-I) as a CutC inhibitor with an IC50 value of 2.4 +/- 0.3 mu M. Kinetic analysis revealed that BO-I functions as a non-competitive inhibitor of CutC. Interestingly, BO-I reduced the production of TMA in whole cell assays of multiple bacterial strains as well as in complex biological environments. Therefore, structural optimization of BO-I holds promise for the development of efficient gut microbiota-targeted small molecules.

Development of newer methods for the synthesis of Benzoxazoles has of greater interest due to their wide range of biological activities and pharmaceutical importance. We herein report a facile and general method for the synthesis of 2-substituted Benzoxazoles via copper catalyzed intramolecular C-O cross-coupling of 2-haloanilides. A combination of CuI (5 mol%), 2,2′-bipyridine (10 mol%), Cs2CO3 (2 equiv.) in DMF solvent with 4 angstrom molecular sieves at 140 degrees C, illustrated the scope for tuning the reactivity of 2-haloanilides toward the selective formation of a series of 2-alkyl benzoxazole derivatives in moderate to good yields. This is the first systematic study using CuI/2,2′-Bipyridine as the catalytic system for the synthesis of 2-substituted Benzoxazoles. Here, we have demonstrated rice husk derived chemically activated carbon catalyzed synthesis of benzothiazole, benzoxazole and N-benzimidazole derivatives at room temperature in aqueous ethanol by the condensation of 2-aminothiophenol, 2-amino phenol and ortho-phenylene diamine with aromatic aldehydes respectively. The activated carbon showed excellent catalytic activity and furnished superior to excellent yields of desired products (95-98%). Flexible poly(benzoxazole-co-amide) (PBOA) gas separation hollow fiber membranes were successfully prepared deriving from ether-containing polyamide precursors by thermal densification with or without a supplementary coating treatment. Compared with polyimide precursors, thermally rearrangement in this work required lower treatment temperature but generated better mechanical properties. Both spinning process and thermal treatment conditions were systemically manipulated to intensify the membrane densification aiming at achieving greatly improved selectivity of the obtained TR-PBOA hollow fiber membranes. It was found that draft ratio had little contribution to selectivity enhancement. Separation performance was substantially improved when precursor fibers were dried using ethanol-hexane exchange method and then thermally treated at 350-390 degrees C TR hollow fibers obtained by combined thermal treatment with coating method exhibited the highest selectivity values in this study.
