In this paper, new liquid crystal compounds with negative dielectric anisotropy were synthesised by regulating the substituents on the benzofuran ring. Their structures were confirmed by(1)H-nuclear magnetic resonance,(CNMR)-C-13 and mass spectra. The effects of substituent on the properties of liquid crystals were investigated. Compared with the reference compounds, the compounds with 5-ethoxy-6,7-difluorobenzofuran ring exhibit higher clearing points as well as the tendency of expansion of SmA phase range. The density functional theory calculation results reveal that the 6-ethoxy-7-fluorobenzofuran-based compound has smaller aspect ratio than that of the 5-ethoxy-6,7-difluorobenzofuran-based compound, which results in the diminish of the mesophase. Although the dielectric anisotropy of the 5-ethoxy-6,7-difluorobenzofuran-based compound is slightly lower than that of the reference compound, it has both the largest perpendicular and parallel dielectric constant.

The detection of Hg2+ in biological systems and its imaging is of highly importance. In this work, a novel ratiometric fluorescence probe is developed based on through-bond energy transfer (TBET) with a 2-(2-hydroxyphenyl)benzoxazole (HBO) as donor and a Rhodamine derivative-Hg conjugate (RDM-Hg) as acceptor. Hg2+ weakens the fluorescence of HBO at 430 nm and meanwhile interacts with Rhodamine B derivative to form a fluorescent conjugate (RDM-Hg) giving rise to emission at 597 nm with a 167 nm red-shift. Further, the difference 282 nm between the donor absorption (315 nm) and the accepter emission (597 nm) for 1 + Hg2+ is comparable to the highest value of the Stokes shift (282 nm) reported earlier for other reported TBET-based cassette. Through-bond energy transfer from HBO to RDM-Hg is triggered by Hg2+ resulting in concentration dependent variation of fluorescence ratio I-597/I-430. A linear calibration of I-597/I-430 versus Hg2+ concentration is obtained within 0-5 mu M, along with the lowest detection limit being found to be as low as 1.31 x 10(-9) mol center dot l(-1) (similar to 0.26 ppb) for Hg2+. This feature is further demonstrated by colorimetric imaging of test strip and intracellular Hg2+. On the other hand, the HBO/RDM TBET sensing system is characterized by a combination of high sensitivity and selectivity. The present study provides an approach for further development of ratiometric probes dedicated to selective in vitro or in vivo sensing some species of biologically interest.

Conclusion: These compiled data in the article will surely update the scientific community with the recent development in this area and will provide direction for further research in this area.

Unassymetric bis[2-(2′-hydroxyphenylbenzoxole)] bis(HBO) derivatives with a DPA functionality for zinc binding have been developed with an efficient synthetic route, using the retrosynthetic analysis. The probes are readily excitable by a 488 nm laser, making this series of compounds a suitable imaging tool for in vitro and in vivo study on a confocal microscope. The application of zinc binding-induced fluorescence turn-on is successfully demonstrated in cellular environments and thrombus imaging.

There is current interest in developing human hypoxia-inducible factor (HIF) prolyl hydroxylase domain (PHD) inhibitors for the treatment of anemia and other hypoxia-related diseases. We describe the synthesis of 3-carbamoylpropanoic acid derivatives and their evaluation as human PHD-2 inhibitors. MS assays indicated that derivatives with a 3-carbamoylpropanoic acids-containing benzoxazole moiety are inhibitors of PHD-2 with IC50 values of 2.24 mu M and 1.32 mu M, respectively. However, neither the acids nor their respective ethyl esters were observed to upregulate HIF-1 alpha levels in cells.

Simple benzoxazines were mixed and reacted with various phenolics such as phenol, p-nitrophenol, p-cresol, 1,3-dihyroxybenzene (resorcinol), 1,3,5-trihydroxybenzene (phloroglucinol), and N-(2-hydroxyphenyl)benzamide. The influence of these phenolic compounds on ring-opening polymerization temperature of benzoxazines was examined by DSC analysis. The cresol-based benzoxazine (C-a) and phenolics yielded polymers with molecular weight of around 1500 Da. Interestingly, for C-a/resorcinol-and C-a/phloroglucinol-based polymers, a second GPC trace was observed that corresponds to a few million daltons for mixing ratios of 4:1 and 5:1. Moreover, the mixtures of C-a and N-(2-hydroxyphenyl)benzamide gave poly(benzoxazine-benzoxazole)s through a new methodology eliminating the need of synthesis of ortho-amide benzoxazines. The obtained polymers were soluble and characterized in detail by H-1 NMR, C-13 NMR, GPC, DSC, and TGA analyses.

A new highly luminescent iridium complex, bis(2-phenylpyridine-N,C-2′)[2-(2′-tosylaminophenyl)benzoxazole-N, N’] iridium(III) [Ir(ppy)(2)(TAPBO)] (TAPBO-2-(2′-tosylaminophenyl)benzoxazole) has been synthesized and its molecular structure determined using single crystal X-ray diffraction analysis. The Ir(III) complex displayed intense yellow photophosphorescence emission that manifested its potential for organic electroluminescence. Photo- and electroluminescent properties of the OLEDs fabricated on the basis of [Ir(ppy)(2)(TAPBO)] with doping concentration of Ir(ppy)(2)(TAPBO) varied from 3 to 20 wt% have been investigated. The OLED with 9 wt% Ir (ppy)(2)(TAPBO) exhibited maximum luminance of 9000 cdm(-2) at 180 mA/cm(-2) and had sufficiently low turn-on voltage of ca. 6 V.

We have developed herein a new approach to the diverse synthesis of novel derivatives of 3-aryl benzo[d]thiazole-2(3H)-imines (3a-g), by a two-component reaction between diazonium salt (2) and various synthesized N-acyl-N’-aryl thioureas (1a-g), in the presence of sodium tert-butoxide as strong base. Finally, it resulted in the production of the desired products with a moderate yield. The chemical structures of these synthesized compounds were confirmed by various physico-chemical methods viz. FT-IR, 1H-NMR, 13C-NMR, and elemental analysis.

Thymoquinone (TQ), a natural compound with antimicrobial and antitumor activity, was used as the starting molecule for the preparation of 3-aminothymoquinone (ATQ) from which ten novel benzoxazole derivatives were prepared and characterized by elemental analysis, IR spectroscopy, mass spectrometry and NMR (H-1, C-13) spectroscopy in solution. The crystal structure of 4-methyl-2-phenyl-7-isopropyl-1,3-benzoxazole-5-ol (1a) has been determined by X-ray diffraction. All compounds were tested for their antibacterial, antifungal and antitumor activities. TQ and ATQ showed better antibacterial activity against tested Gram-positive and Gram-negative bacterial strains than benzoxazoles. ATQ had the most potent antifungal effect against Candida albicans, Saccharomyces cerevisiae and Aspergillus brasiliensis. Three benzoxazole derivatives and ATQ showed the highest antitumor activities. The most potent was 2-(4-fluorophenyl)-4-methyl-7-isopropyl-1,3-benzoxazole-5-ol (1f). Western blot analyses have shown that this compound inhibited phosphorylation of protein kinase B (Akt) and Insulin-like Growth Factor-1 Receptor (IGF1R ) in HeLa and HepG2 cells. The least toxic compound against normal fibroblast cells, which maintains similar antitumor activities as TQ, was 2-(4-chlorophenyl)-4-methyl-7-isopropyl-1,3-benzoxazole-5-ol (1e). Docking studies indicated that 1e and 1f have significant effects against selected receptors playing important roles in tumour survival.
