Mixed-matrix membrane (MMM) is an effective way to overcome trade-off limitations of conventional polymeric membranes. However, the existence of defect voids at the polymer/filler interface often limits their performance improvement. Similar issues are also present in thermally rearranged polybenzoxazole (TR-PBO)-derived MMMs. To address this challenge, the selection of fillers is of great importance. Herein, a novel organic porous nanosphere (TC-cPSB), which is prepared by the polycondensation of 9,9-bis(4-aminophenyl) fluorene (BAFL) and terephthalaldehyde (TPAL) followed by thermal crosslinking, is chosen to engineer the polymer/filler interface. Benefiting from strong intermolecular interaction (pi-pi stacking and hydrogen bonding), the TC-cPSB nanosphere can well disperse in TR-PBO matrix with a defect-free interface. With an increase in TC-cPSB loading, well-designed MMMs exhibit a significant anti-trade-off’ phenomenon whereby gas permeability and selectivity increase simultaneously, following the trend predicted by the Maxwell model. Compared with TR-PBO membrane, the MMM containing 15 wt% of nanosphere shows an increase of 282% and 217.6% in H-2/CO2 selectivity and H-2 permeability, respectively, which is far beyond 2008 Robeson upper bound.

New kinds of heterocyclic mesogenic compounds containing benzoxazole, ethynyl, and laterally fluorinated phenyl groups, namely 2-[4-[2-[4-alkoxyphenyl]ethynyl]-2,3-difluorophenyl]benzoxazole derivatives (nPEDFPBx), are synthesized and evaluated. The compounds nPEDFPBx except nitro-substituted ones have enantiotropic nematic mesophases with mesophase ranges of 16-86 degrees C and 29-108 degrees C on heating and cooling, respectively. The results show that introduction of two lateral fluorine atoms into molecule results in enhanced nematic mesophase stability. Meanwhile, nPEDFPBx displays much higher birefringence (0.507-0.624) than common tolanebased liquid crystals, which is ascribed to its large pi-conjugated molecule composed of benzene, ethynyl, and benzoxazole mesogenic unit. The nPEDFPBx has a high potential to serve as a dopant for liquid crystal mixture. [GRAPHICS] .

In this study, a total of 17 piece 2,5-disubstituted benzoxazole derivatives were synthesized, 2 of which were not original, their antimicrobial activities were determined using microdilution method and theirin vitrocytotoxic activities were investigated on MCF-7 and A549 cells by MTT test. When the activity results are examined, although the antibacterial effects of benzoxazole derivatives are weaker than standard drugs;3N13and3N19againstCandida albicansisolate showed the closest activity to fluconazole with MIC: 16 mu g/ml. The cytotoxicity test was measured at a concentration of 100 mu M and a 24-h incubation period. The results showed that the compounds had weak activities against two cell lines. Molecular docking studies of synthesized compounds were performed on sterol 14 alpha-demethylase protein (CYP51) and protein-ligand interactions of3N13, the most effective derivative againstC. albicansisolate, were showed (PDB: 5TZ1). Estimated ADME profiles of compounds were calculated and also3N13’s were calculated HUMO-LUMO energies, molecular electrostatic potential analysis, and geometric optimization parameters with 6-311 G+ (d,p) base set using DFT/B3LYP theory, and the results were displayed.

In the present study, we attempted to develop novel class of Mycobacterium tuberculosis (Mtb) inhibitors by exploring the pharmaceutically underexploited enzyme targets which are majorly involved in cell wall biosynthesis of mycobacteria. For this purpose glutamate racemase was selected which racemizes D-glutamate from L-glutamate, a key step in peptidoglycan synthesis. Furthermore, enzyme is neither expressed nor its product, n-glutamate is produced in mammals, and hence inhibiting this enzyme will have no vulnerable effect in host organism. A library of our in-house compounds were screened against glutamate racemase using a biophysical technique; thermal shift assay and further by enzyme inhibition assay to identify Lead 1 molecule. Lead 1 optimization and expansion resulted in twenty four compounds. Among the synthesized compounds twelve compounds shown good enzyme inhibition than Lead 1 (IC50 20.07 +/- 0.29 mu M). Among all the compounds; compound 22 (IC50 1.1 +/- 0.52 mu M) showed potent non-competitive mode of inhibition in enzyme assay. Further showed good susceptibility (in replicating bacteria) of MIC 8.72 mu M and bactericidal time dependant kill on dormant culture. It also exhibited significant activity in Mtb nutrient starvation model (2.5) and Mtb biofilm model (2.4) and in vivo M. marinum infected Zebra fish model studies (3.6) reduction at logarithmic scale. Macrophage migration inhibitory factor (MIF) is a key pleiotropic mediator and a promising therapeutic target in cancer as well as in several inflammatory and cardiovascular diseases including pulmonary arterial hypertension (PAH). Here, a novel series of N-(phenylmethyl)-benzoxazol-2-thiones 5-32 designed to target the MIF tautomerase active site was synthesized and evaluated for its effects on cell survival. Investigation of structure-activity relationship (SAR) particularly at the 5-position of the benzoxazole core led to the identification of 31 that potently inhibits cell survival in DU-145 prostate cancer cells and pulmonary endothelial cells derived from patients with idiopathic PAH (iPAH-ECs), two cell lines for which survival is MIF-dependent. Molecular docking studies helped to interpret initial SAR related to MIF tautomerase inhibition and propose preferred binding mode for 31 within the MIF tautomerase active site. Interestingly, daily treatment with 31 started 2 weeks after a subcutaneous monocrotaline injection regressed established pulmonary hypertension in rats.

In this work, the 4-methyl-5-thiazoleethanol (C6H9NSO) molecule was studied by using the experimental spectroscopic techniques (UV-vis in three different solvents and the Fourier transform infrared spectroscopies) and density functional theory calculations. The molecular geometric parameters, vibrational wavenumbers, high-occupied-low-occupied molecular orbitals energies, H-1 and C-13 NMR chemical shift values, molecular electrostatic potential, natural bond orbitals, and nonlinear optical properties of the 4-methyl-5-thiazoleethanol were performed by using the B3LYP, B3LYP-GD3 and HSEH1PBE levels of density functional theory with 6-311++ G(d, p) basis set. The spectral results obtained from the quantum chemical calculations of 4-methyl-5-thiazoleethanol are in a good agreement with the experimental results.

Two fluorescent heterocyclic ligands were obtained by reduction of imidazo[4,5-e][2,1]benzoxazole derivatives with Fe/HCl. New fluorescent nickel(II) complexes were synthesized by coordination of Ni(II) cation with the heterocyclic ligands. The structure of the complexes was established by spectral and analytical data, Job plot, and DFT calculations. Photophysical properties of Ni(II) complexes were characterized by UVVis and fluorescence spectroscopy. The optimized geometry, spectral properties, and energy difference between the HOMO and LUMO frontier orbitals of the ligands and Ni(II) complexes were obtained by DFT calculations at the B3LYP/6-311++G(d, p) level. The calculated spectral properties are in good agreement with the experimental values. The complexes were also tested as homogeneous catalysts for the transesterification of corn oil with methanol. The structure of the obtained product was confirmed by 1H NMR analysis. The catalytic results showed that the new Ni(II) complexes can be considered as potential candidates for the development of new catalytic systems for biodiesel production.

Synthesis and isolation of highly unstable azirinobenzoxazole and benzoxazines in a chemodivergent fashion from aryl azido vinylogous carbonates by simple change in transition metal acetate is described. Their nickel(II) acetate-promoted reaction gave 4-dihydro-2H-benzoxazines, whereas copper(II) acetate led to the corresponding oxidized imine derivatives. Benzaoxazine derivative could be kinetically resolved using a proline-catalyzed Mannich reaction. The benzoxazines were rapidly elaborated to angularly fused tetracyclic systems and coumarin-fused derivatives in a one pot fashion.

Each component of a gas mixture affects the permeation of the other gases. This mutual influence was systematically investigated for binary gas mixtures that included CO2, N-2, CH4, H-2, and CO permeating through thermally rearranged poly(benzoxazole-co-imide) (TR-PBOI) membranes. The mixed gas permeation was measured at 35 degrees C at up to 5 bar of feed pressure. Sorption isotherms of binary mixtures were calculated using the Grand Canonical Monte Carlo approach for quantifying the sorption contribution to the permeation. The diffusivity contribution was estimated by using the permeability value. How and how much CO2 (the most soluble gas) and H-2 (the fastest diffusing gas among the gas species tested) affect each other’s permeation as well as the permeation of the other gases were specifically addressed, including a discussion based on sorption and diffusion contributions. The permeance of CO2 and H-2 remained unchanged, whereas the permeance of the other gases was reduced by the presence of CO2 or increased by the presence of H-2.
