General procedure: Commercially available 5-chloro benzoxazoles or benzothiazoles(5 mmol) and K2CO3 (15 mmol) were added to 50 mL of DMF.The mixture was heated at 60 C and 1-(2-chloroethyl)hexahydro-1H-azepine hydrochloride (6.5 mmol) was added. The mixture was maintained at 60 C under stirring for 3 h, then was cooled to room temperature, poured into water, and extracted with diethyl ether(3 50 mL). The organic layer was washed with brine, dried overan hydrous Na2SO4 and evaporated under reduced pressure to give are sidue, which was purified by flash column chromatography oversilica gel using ethyl acetate/methanol 8:2 v/v as eluent. Immediately after solvent evaporation, free bases were converted into hydrochlorides by dissolving the corresponding amine in diethyl ether and treating with a 1.25MHCl ethanol solution (pH 1e2). The precipitate was filtered off, washed with diethyl ether, and dried under nitrogen. The following compounds were obtained using this procedure:

Step 2 2.97 g (15.9 mmol) of the compound obtained in Step 1 and 4.43 g (23.8 mmol) of 1-tert-butyloxycarbonylpiperazine were placed in a 250 mL flask, /j-xylene was added thereto, and the mixtue was incubated at 138 C for 15 hrs. Thionyl Chloride (540 mL) and Dimethyl formamide (270 mL) were added to a stirred mixture of 5-Chloro-1,3-benzoxazole-2-thiol (540 g, 2.903 moles) and dichloromethane (5 liter) at 5-10 C. and stirred till clear solution was observed. The reaction mixture was then stirred at 10 C. to room temperature for 4 hour. After completion of the reaction, the mixture was poured into cold water (4 liter), neutralized with solid sodium bicarbonate (1440 g) portion wise over a period of 1 hour and extracted with dichloromethane (2*2.5 liter). The combined organic extracts were washed with brine solution, dried over anhydrous sodium sulphate (300 g) and concentrated under reduced pressure to give a crude compound. The crude material was triturated with Hexane (2*2 liter) at -20 C., filtered and and dried under reduced pressure to obtain 2,5-dichloro-1,3-benzoxazole (475 g, 88.9%) as a yellow liquid.

Subsequently, 5-chloro-2-mercaptobenzoxazole (3.1 g, 16.7 mmol) was dissolved in thionyl chloride (30 mL, 413 mmol). DMF (1.5 mL) was added and the reaction mixture was heated at 65 C. for 45 min. The solvent was removed under reduced pressure and to the residue was added toluene (2*60 mL) followed by evaporation each time to remove the excess SOCl2 (azetrope). The resultant crude product was dissolved in ethyl acetate (100 mL), washed with water (100 mL) and dried over Na2SO4. Evaporation of ethyl acetate gave 2,5-dichlorobenzoxazole, compound 19, as a red oil (3.2 g).

General procedure: Phosphorus tribromide (89 muL, 0.11 mmol) was dropwisely added to a solution of commercial 2-phenethylbenzylalcohol (75 mg, 0.35 mmol) in dry THF (5 mL) at 0 C. After stirring at room temperature for 2h, the reaction mixture was extracted with EtOAc after quenching with water. The organic extract was washed with brine and dried over MgSO4. Evaporation of the filtrate provided crude 2-phenethylbenzyl bromide. Then, commercial 2-mercaptoimidazole (70 mg, 0.47 mmol), sodium hydride (22 mg, 0.55 mmol) and potassium iodide (78 mg, 0.47 mmol) was added to a solution of crude 2-phenethylbenzyl bromide in DMF (3 mL). After stirring at 100C overnight, the reaction mixture was quenched with water, extracted with EtOAc, washed with brine and dried over MgSO4 and evaporated. Purification of the residue by column chromatography on silica gel (hexane/EtOAc = 1/1) provided the title compound (44 mg, 0.15 mmol) as a colorless solid in 43% yield.
