SYNTHESIS AND ANTIFUNGAL ACTIVITY OF BENZOXAZOLE DERIVATIVES WITH THEIR SAR ANALYSIS BY SAS-MAP

A simple and efficient method has been developed for the synthesis of benzoxazole derivatives. In the first step, 2-aminophenols reacted with various aldehydes in the presence of titanium supported nano-silica to produce an imine intermediate. Then the imine was oxidized to get the final azole compounds. Eleven derivatives were synthesized (c)-(m) via this simple and environmentally friendly procedure with high yields. The simple methodology (only 2 steps) with high yields for the reactions and easy procedure are the advantages of the newly developed method. The identification and characterization of all the synthesized compounds were confirmed by melting point, thin layer chromatography, FT-IR, H-1 NMR and C-13 NMR spectral data. Also elemental analysis was applied for five compounds. All the compounds were screened for antimicrobial activity by broth microdilution methods as recommended by CLSI. Of the tested compounds 2-(2,4-dichlorophenyl)-1,3-benzoxazole (g), and 2-( 4-chlorophenyl)-1,3-benzoxazole (f) inhibited the growth of all examined fungi, while 2-(4-nitrophenyl)-1,3-benzoxazole (c) exhibited inhibitory activities only against the tested yeasts.

Synthesis and antimicrobial evaluation of novel 5-substituted-2-(p-tert-butylphenyl)benzoxazoles

In the present study, a series of nine novel 5-substituted-2-(p-tert-butylphenyl)benzoxazole derivatives have been synthesized and their structures confirmed by spectral techniques and also tested for their antimicrobial activities. The minimum inhibitory concentrations (MIC) of the new benzoxazoles have been determined against standard bacterial and fungal strains and drug-resistant isolates and compared to those of several reference drugs. The new benzoxazole derivatives are found to possess a broad spectrum of antibacterial activity with MIC values of 8-256 mu g/mL. Especially, compound 9 is more active than standard drugs ciprofloxacin and cefotaxime against E. coli isolate with a MIC value of 8 mu g/mL. Also new compounds are less active than fluconazole with a MIC value of 256 mu g/mL against C. albicans and its isolate except for compound 9 that shows better activity other compounds with a MIC value of >4 mu g/mL for their antifungal activity.

We previously reported 2-aminobenzoxazole analogue 1 as a potent ChemR23 inhibitor. The compound showed inhibitory activity against chemerin-induced calcium signaling through ChemR23 internalization in CAL-1 cells, which are cell lines of plasmacytoid dendric cells (pDCs). Furthermore, compound 2 inhibited chemotaxis of CAL-1 triggered by chemerin in vitro. However, we noted a difference in the ChemR23 response to our inhibitor between rodents and non-rodents in a previous study. To address this issue, we performed optimization of ChemR23 inhibitors using CAL-1 cells endogenously expressing human ChemR23 and conducted a pharmacokinetics study in cynomolgus monkeys. Various substituents at the 4-position of the benzoxazole ring exhibited potent in vitro bioactivity, while those at the 6-position were not tolerated. Among substituents, a carboxyl group was identified as key for improving the oral bioavailability in cynomolgus monkeys. Compound 38a with the acidic part changed from a tetrazole group to a 1,2,4-oxadiazol-5-one group to improve bioactivity and pharmacokinetic parameters exhibited inhibitory activity against chemerin-induced chemotaxis in vitro. In addition, we confirmed the ChemR23 internalization of pDCs by compound 38a orally administered to cynomolgus monkeys. These 2-aminobenzoxazole-based ChemR23 inhibitors may be useful as novel immunotherapeutic agents capable of suppressing the migration of pDCs, which are known to be major producers of type I interferons in the lesion area of certain autoimmune diseases, such as systemic lupus erythematosus and psoriasis.

Cu(I) complexes regulated by N-heterocyclic ligands: Syntheses, structures, fluorescence and electrochemical properties

New pyrimidine-benzoxazole/benzimidazole hybrids: Synthesis, antioxidant, cytotoxic activity, in vitro cyclooxygenase and phospholipase A2-V inhibition

Furthermore, the antioxidant activity, phospholipase A2-V and cyclooxygenases inhibitory activities of the target compounds 7 aRrb were evaluated and the new compounds showed potent activity (cytotoxicity IC50 range from 4.3 to 9.2 mu m, antioxidant activity from 40% to 80%, COXs or LOX inhibitory activity from 1.92 mu M to 8.21 mu M). Light-Induced Bending of Needle-Like Crystals of Naphthylvinylbenzoxazole Triggered by trans-cis Isomerization

New diarylethene derivatives containing benzoxazole (NBO) and benzothiazole (NBT) have been synthesized. Light-induced trans-cis isomerization of NBO and NBT took place in crystals, and only induced the needle-like crystals of NBO to bend backwards away from the UV light source. The movement of the atoms was deemed to take place during the isomerization of NBO; hence, strain would be produced and accumulated rapidly in the surface of crystals exposed to UV light. The uniform release of strain led to the bending of needle-like crystals. The light-induced trans-cis isomerization efficiency of NBT was too low to drive the motion of crystals, which might have originated from the large repulsion between naphthyl and benzothiazole. These results provide a new platform for the transformation of light energy into mechanical energy in molecular crystals through the unimolecular photochemical reaction of diarylethene derivatives.
