Synthesis and Properties of Polybenzoxazole Copolymers Having Non Linear Units

A series of poly(hyroxyamide)s (PHAs) was prepared by direct polycondensation reaction of 4,4′-(2,3-pyridinedioxy)dibenzoic acid and/or isophthalic acid with 3,3′-dihydroxybenzidine. The identification and pharmacological evaluation of potent, selective and orally available ACC1 inhibitor

In our effort to explore the potential of ACC1-selective inhibitor as in vivo probe molecule, a series of 1,3-benzoxazole derivatives was synthesized. Previously, we reported a series of novel bicyclic and monocyclic ACC1-selective inhibitors. Among them, compound 1a exhibited highly potent cellular activity (acetate uptake IC50 = 0.76 nM) as well as promising in vivo PD efficacy. However, compound 1a caused severe body weight reduction in repeated dose administration in the mouse model. Since 1a showed potent inhibitory activity against mouse ACC1 as well as strong inhibition of mouse ACC2, we further examined a series of 1a analogues in order to reduce undesirable body weight change. The replacement of acetamide moiety with ureido moiety dramatically improved selectivity of mouse ACC1 against ACC2. In addition, analogue 1b displayed favorable bioavailability in mouse cassette dosing PK study, hence in vivo PD studies were also carried out. Oral administration of 1b significantly reduced the concentration of malonyl-CoA in HCT-116 xenograft tumors at doses of more than 30 mg/kg. Furthermore, compound 1b showed significant antitumor efficacy in 786-O xenograft mice at an oral dose of 30 mg/kg (T/C = 0.5%). Accordingly, our novel potent ACC1-selective inhibitor represents a set of useful orally-available research tools, as well as potential therapeutic agents particularly in terms of new cancer therapies.

Cardo-type porous organic nanospheres: Tailoring interfacial compatibility in thermally rearranged mixed matrix membranes for improved hydrogen purification

Mixed-matrix membrane (MMM) is an effective way to overcome trade-off limitations of conventional polymeric membranes. However, the existence of defect voids at the polymer/filler interface often limits their performance improvement. Similar issues are also present in thermally rearranged polybenzoxazole (TR-PBO)-derived MMMs. To address this challenge, the selection of fillers is of great importance. Herein, a novel organic porous nanosphere (TC-cPSB), which is prepared by the polycondensation of 9,9-bis(4-aminophenyl) fluorene (BAFL) and terephthalaldehyde (TPAL) followed by thermal crosslinking, is chosen to engineer the polymer/filler interface. Benefiting from strong intermolecular interaction (pi-pi stacking and hydrogen bonding), the TC-cPSB nanosphere can well disperse in TR-PBO matrix with a defect-free interface. With an increase in TC-cPSB loading, well-designed MMMs exhibit a significant anti-trade-off’ phenomenon whereby gas permeability and selectivity increase simultaneously, following the trend predicted by the Maxwell model. Compared with TR-PBO membrane, the MMM containing 15 wt% of nanosphere shows an increase of 282% and 217.6% in H-2/CO2 selectivity and H-2 permeability, respectively, which is far beyond 2008 Robeson upper bound.

Identification and development of benzoxazole derivatives as novel bacterial glutamate racemase inhibitors

In the present study, we attempted to develop novel class of Mycobacterium tuberculosis (Mtb) inhibitors by exploring the pharmaceutically underexploited enzyme targets which are majorly involved in cell wall biosynthesis of mycobacteria. For this purpose glutamate racemase was selected which racemizes D-glutamate from L-glutamate, a key step in peptidoglycan synthesis. Furthermore, enzyme is neither expressed nor its product, n-glutamate is produced in mammals, and hence inhibiting this enzyme will have no vulnerable effect in host organism. A library of our in-house compounds were screened against glutamate racemase using a biophysical technique; thermal shift assay and further by enzyme inhibition assay to identify Lead 1 molecule. Lead 1 optimization and expansion resulted in twenty four compounds. Among the synthesized compounds twelve compounds shown good enzyme inhibition than Lead 1 (IC50 20.07 +/- 0.29 mu M). Among all the compounds; compound 22 (IC50 1.1 +/- 0.52 mu M) showed potent non-competitive mode of inhibition in enzyme assay. Further showed good susceptibility (in replicating bacteria) of MIC 8.72 mu M and bactericidal time dependant kill on dormant culture. It also exhibited significant activity in Mtb nutrient starvation model (2.5) and Mtb biofilm model (2.4) and in vivo M. marinum infected Zebra fish model studies (3.6) reduction at logarithmic scale. Benzoxazole and dioxolane substituted benzimidazole-based N-heterocyclic carbene-silver( I) complexes: Synthesis, structural characterization and in vitro antimicrobial activity

A novel series of 1,3-benzoxazole and 1,3-dioxolane substituted benzimidazole-based N-heterocyclic carbene (NHC) precursors (6a-b and 11a-b) and their corresponding NHC-silver(I) acetate (12a-b and 14a-b) and bis-NHC-silver(I) hexafluorophosphate complexes (13a-b and 15a-b) were synthesized and characterized by H-1 NMR, C-13 NMR, ATR-IR spectroscopic methods and elemental analysis techniques. Also, the molecular structure of benzimidazolium salt (6a) and bis-NHC-silver(I) hexafluorophosphate complex (13b) were unambiguously established by single crystal X-ray diffraction analysis. Fascinatingly, bis-NHC-silver(I) hexafluorophosphate complex (13b) was revealed to be dinuclear in nature. All the azolium salts (6a-b and 11a-b) as well as the corresponding silver(I) complexes (12a-b, 13a-b, 14a-b and 15a-b) were evaluated for their antimicrobial potential against Gram positive (Staphylococcus aureus), and a set of Gram negative (Escherichia coli, Klebsiella pneumoniae, Acinetobacter baumannii and Pseudomonas aeruginosa) bacterial strains and two fungi, Candida albicans and Cryptococcus neoformans. Antimicrobial studies revealed the comparable activities of benzimidazolium salts (6a-b and 11a-b) and the silver(I) complexes (12a-b, 13a-b and 15a-b). Whereas, the 1,3-dioxolane substituted benzimidazole-based silver(I) acetate complexes (14a-b) demonstrated excellent potentials with both bacterial and fungal growth inhibition above 95%. Design, synthesis and biological potential of heterocyclic benzoxazole scaffolds as promising antimicrobial and anticancer agents

Background: Benzoxazole is the most important class of heterocyclic compound in medicinal chemistry. It has been incorporated in many medicinal compounds making it a versatile heterocyclic compound that possess a wide spectrum of biological activities. Results: The molecular structures of synthesized benzoxazole derivatives were confirmed by physicochemical and spectral means. The synthesized compounds were further evaluated for their in vitro biological potentials i.e. antimicrobial activity against selected microbial species using tube dilution method and antiproliferative activity against human colorectal carcinoma (HCT 116) cancer cell line by Sulforhodamine B assay. Conclusion: In vitro antimicrobial results demonstrated that compounds 4, 5, 7 and 16 showed promising antimicrobial potential. The in vitro anticancer activity indicated that compounds 4 and 16 showed promising anticancer activity against human colorectal cancer cell line (HCT 116) when compared to standard drug and these compounds may serve as lead compound for further development of novel antimicrobial and anticancer agents.

Synthesis and biological activity evaluation of azacycloheptane sulfonamide derivatives as potential orexin receptor antagonists

As the orexin signaling system is crucial for the regulation of the sleep/wake cycle, inhibitors of orexin-1 and orexin-2 receptors are of significant interest in the treatment of insomnia. Herein, a series of novel azacycloheptane sulfonamide derivatives were designed and synthesized, and all the compounds were evaluated as potential orexin receptor inhibitors by FLIPR Tetra calcium assay. A majority of the tested azacycloheptane sulfonamide derivatives showed OX1R and OX2R inhibitory activity. Chloro-substituted derivatives functionalized at the C5 or C6 position of the benzoxazole group exhibited better inhibitory activity for OX1R and OX2R than unsubstituted derivatives functionalized at C5 or C6. In addition, phenyl group modification had positive effects on the inhibitory activities, and an electron-withdrawing fluorine group at theorthoormetaposition of the phenyl ring improved the OX2R inhibitory activity of the derivatives. This suggests that azacycloheptane sulfonamide derivatives are promising scaffolds for the development of OX1R and OX2R antagonists.

Encapsulation of Risperidone by Methylated beta-Cyclodextrins: Physicochemical and Molecular Modeling Studies

Risperidone (RSP) is an atypical antipsychotic drug which acts as a potent antagonist of serotonin-2 (5TH2) and dopamine-2 (D2) receptors in the brain; it is used to treat schizophrenia, behavioral and psychological symptoms of dementia and irritability associated with autism. It is a poorly water soluble benzoxazole derivative with high lipophilicity. Supramolecular adducts between drug substance and two methylated beta-cyclodextrins, namely heptakis(2,6-di-O-methyl)-beta-cyclodextrin (DM-beta-CD) and heptakis(2,3,6-tri-O-methyl)-beta-cyclodextrin (TM-beta-CD) were obtained in order to enhance RSP solubility and improve its biopharmaceutical profile. The inclusion complexes were evaluated by means of thermoanalytical methods (TG-thermogravimetry/DTG-derivative thermogravimetry/HF-heat flow), powder X-ray diffractometry (PXRD), universal-attenuated total reflectance Fourier transform infrared (UATR-FTIR), UV spectroscopy and saturation solubility studies. Job’s method was employed for the determination of the stoichiometry of the inclusion complexes, which was found to be 2:1 for both guest-host systems. Molecular modeling studies were carried out for an in-depth characterization of the interaction between drug substance and cyclodextrins (CDs). The physicochemical properties of the supramolecular systems differ from those of RSP, demonstrating the inclusion complex formation between drug and CDs. The RSP solubility was enhanced as a result of drug encapsulation in the CDs cavity, the higher increase being obtained with DM-beta-CD as host; the guest-host system RSP/DM-beta-CD can thus be a starting point for further research in developing new formulations containing RSP, with enhanced bioavailability.
