General and efficient synthesis of benzoxazol-2(3H)-ones: Evolution of their anti-cancer and anti-mycobacterial activities

A novel class of benzo[d]oxazol-2(3H)-one derivatives has been synthesized and their in vitro cytotoxicity against human pancreatic adenocarcinoma and human non-small cell lung carcinoma cancer cell lines was evaluated. Many of these compounds were found to display excellent to moderate activity. Among them, 6b, 6l, 6n and 6x were identified as lead molecules. In particular, 6l and 6n were found to be potent against the pancreatic adenocarcinoma cell line whereas the 6x was found to be effective against the human non-small cell lung carcinoma cell line. Conversely, the compounds 6l-x were found to be ineffective against Mycobacterium tuberculosis. Of the various molecules, 6h showed promising anti-mycobacterial activity, with an IC50 value equal to that of ciprofloxacin.

By acid hydrolysis, these compounds yielded 3-(2-phenylcarbazoyl)-2(3H)benzoxazolone derivatives 4 which were not isolated and were transformed via an intramolecular reaction into 4-(2-hydroxyphenyl)-1-phenyl-1,2,4-triazolidine-3,5-dione derivatives 5 in a good yield. Attempts to cyclize these compounds by intramolecular elimination of water into tricyclic compounds 6 with various dehydrating agents were unsuccessful.

General procedure: To a stirred suspension of benzo [d]oxazol-2(3H)-ones, anhydrousK2CO3 (1.2 mmol) was added in the dry DMF solvent for20 min at 60 C. Then chloroacetylated 3a-3e (1.3 mmol) and TBAI(0.1 mmol) were added simultaneously. The reaction was stirredand heated for 5 h. After completion, the mixture was extractedwith DCM/H2O three times. The combined organic layers werewashed with brine, dried over anhydrous Na2SO4, filtered and concentrated under reduced pressure. The crude product was purifiedby silica gel column chromatography eluting with DCM/Methanol (20:1e10:1) to afford compounds D01-D08.

A solution of 28A (0.69 g, 3.83 mmol) in 2-ethoxyethanol (10 mL) was treated with KOH (0.22 g, 3.83 mmol) and stirred at RT for 1 h. Step 2. A mixture of 5-nitrobenzo[d]oxazol-2(3H)-one (3.60 g, 20.0 mmol), benzyl bromide (2.61 mL, 22.0 mmol) and cesium carbonate (7.81 g, 23.98 mmol) in DMF (50 mL) was sealed and heated in a microwave system at 85 C for 2 h. The reaction mixture was poured into water (500 mL), heated with a heat gun to near boiling and allowed to cool to rt with stirring. The resulting solids were collected by filtration, washed with water and hexanes and dried in vacuo to afford 3-benzyl-5-nitrobenzo[d]oxazol-2(3H)-one (5.20 g) as a gray solid. NMR (400 MHZ, DMSO-de) delta ppm 8.22 – 8.06 (m, 2H), 7.63 (d, J=8.8 Hz, 1H), 7.50 – 7.20 (m, 5H), 5.17 (s, 2H).
