An operationally simple method for synthesis of benzimidazole and 3H-imidazo[4,5-c]pyridine from o-phenylenediamine or pyridine-3, 4-diamine and N,N-dimethylformamide (DMF) in the presence of hexamethyldisilazane (HMDS) as a reagent is described. To evaluate the scope of application of this reagent, it was also used to prepare benzothiazole, 1H-perimidine, and benzoxazole, which was successful for benzothiazole and 1H-perimidine but benzoxazole was not formed. This reaction complies with the principles of green chemistry as it does not use toxic solvents, transition metals, or strong acids. The products are obtained in moderate to excellent yields.

This article describes the multi-step synthesis of 2-(2-hydroxybenzofuran)benzoxazole (HBBO) derivatives functionalised with one to three oligo(ethylene glycol) (OEG) chains with the goal to allow a good vectorization in aqueous media. Benzoxazole dyes are well-known organic fluorophores exhibiting excited-state intramolecular proton transfer (ESIPT) emission. The insertion of these highly hydrophilic OEG chains allows a good solubilization of these dyes in a wide range of solvents of different polarity including PBS buffer/DMSO (95/5) and water, for one of them. The photophysical properties of these ESIPT emitters in solution, and also in the solid-state, as doped as 1 wt% in poly(methylmethacrylate) (PMMA) or polystyrene (PS) films, are discussed. Specifically, the presence of an OEG chain at the 3-position of the benzofuran ring was found to sizeably enhance the fluorescence intensity of the ESIPT emission in the solution state.

A series of four water-soluble benzazole dyes that emit fluorescence by the excited state intramolecular proton transfer (ESIPT) mechanism were structurally characterized by Fourier transform infrared spectroscopy (FTIR), H-1 and C-13 nuclear magnetic resonance (NMR) attached proton test (APT) and mass spectrometry. Their photophysical properties were systematically studied by UV-absorption and fluorescence emission. Some photophysical parameters were obtained by semi-empirical PM3 and ZINDO methods, and related to experimental photophysical data. Changes in the absorption and fluorescence emission spectra as well as conformational equilibrium between different species were investigated in solvents of different polarities and under different pH conditions. These benzazole dyes emit dual fluorescence emission in the blue-orange region with a Stokes shift between 2617-12337 cm(-1). In general, these dyes are potentially interesting for studies of biological systems in an aqueous environment due to the presence of groups that increase aqueous solubility and reactivity with biomolecules. The present dyes were successfully used as new probes by means of direct staining of larvae. The obtained results indicate that these fluorescent dyes permit a quick, easy and selective visualization of larvae.

A new highly luminescent iridium complex, bis(2-phenylpyridine-N,C-2′)[2-(2′-tosylaminophenyl)benzoxazole-N, N’] iridium(III) [Ir(ppy)(2)(TAPBO)] (TAPBO-2-(2′-tosylaminophenyl)benzoxazole) has been synthesized and its molecular structure determined using single crystal X-ray diffraction analysis. The Ir(III) complex displayed intense yellow photophosphorescence emission that manifested its potential for organic electroluminescence. Photo- and electroluminescent properties of the OLEDs fabricated on the basis of [Ir(ppy)(2)(TAPBO)] with doping concentration of Ir(ppy)(2)(TAPBO) varied from 3 to 20 wt% have been investigated. The OLED with 9 wt% Ir (ppy)(2)(TAPBO) exhibited maximum luminance of 9000 cdm(-2) at 180 mA/cm(-2) and had sufficiently low turn-on voltage of ca. 6 V.

A new series of benzimidazole/benzoxazole linked beta-carbolines (9a-j) were rationally designed and synthesized by combining two different anti-cancer fragments. The new hybrid beta-carbolines are subjected to anti-cancer screening against four different human cancer cell lines such as MCF-7 (breast), A549 (lung), Colo-205 (colon) and A2780 (ovarian) by using standard MIT assay. These hybrid beta-carbolines exhibited significant and high fold anti-cancer activity against MCF-7 cell lines than reference standard. They are also proved to be effective against A549 and Colo-205 cell lines. Further, compound 9b, 9c from benzimidazole and 9i from benzoxazole series have exhibited maximum anti-cancer activity among these hybrid beta-carbolines. Later, all of the hybrid beta-carbolines were subjected to molecular interaction analysis against a few selected kinase targets such as cdc-like kinase (CLK-1 to CLK-4), epidermal growth factor reductase (EGFR) kinase, protein (ATR) kinase along with APC-Asef interface. The violin plot of binding energies of 9a-9j have suggested them as good kinase binders. Result interpretation suggested hybrid beta-carbolines as promising CLK binders. The anti-cancer data of new hybrid beta-carbolines against MCF-7 cell lines are in agreement with parent beta-carboline skeleton. (C) 2020 Elsevier B.V. All rights reserved.

Karanja (Pongamia pinnata) is a medicinal tree used in the Indian traditional ayurvedic system for treating several ailments. The seeds contain a unique furano-flavonoid karanjin, which has shown to possess many medicinal properties. Its usage at the clinical level is affected due to poor solubility and absorption. In the present investigation, molecular modifications of karanjin were attempted and evaluated their effect on anti-inflammatory activity. Firstly, Karanja ketone was obtained from karanjin by hydrolysis, and it was converted into karanja ketone oxime. The oxime undergoes Beckmann rearrangement and cyclized to yield furano benzoxazole (karanja oxazole). The new derivatives were purified with >95% purity (HPLC) and spectrally characterized (HR-MS, FTIR, and NMR). Among the test compounds, karanja ketone oxime exhibited higher antioxidant activity with an IC50 value of 360 mu g/ml (DPPH). Soy lipoxygenase-1 (LOX-1) inhibitory activity of oxime was higher (IC(50 )65.4 mu m) than other compounds. Fluorescence studies showed that oxime had higher quenching capacity with a Qmax of 76.3% and a binding constant of 0.9 x 10(5) M-1 for soy LOX-1. In-silico interaction studies showed that karanja ketone oxime had the least binding energy of -5.76 kcal/mol with LOX-1 by forming two hydrogen bonds with hydrophobic amino acids Leu 390 and Gly 392. The compounds were evaluated for their acute antiinflammatory activity by the paw and ear edema in the rat model. Karanjin inhibits paw edema and ear edema by 34.13% and 51.13%, respectively, whereas the derivatives inhibited by 45-57 % and 70-76.8%. This study reports a rational approach to synthesize karanjin derivatives with considerable anti-inflammatory properties, both in-vitro and in-vivo.

A series of N-(2-(3,5-dimethoxyphenyl)benzoxazole-5-yl)benzamide derivatives (3am) was synthesized and evaluated for their in vitro inhibitory activity against COX-1 and COX-2. The compounds with considerable in vitro activity (IC50<1M) were evaluated in vivo for their anti-inflammatory potential by the carrageenan-induced rat paw edema method. Out of 13 newly synthesized compounds, 3a, 3b, 3d, 3g, 3j, and 3k were found to be the most potent COX-2 inhibitors in the in vitro enzymatic assay, with IC50 values in the range of 0.06-0.71M. The in vivo anti-inflammatory activity of these six compounds (3a, 3b, 3d, 3g, 3j, and 3k) was assessed by the carrageenan-induced rat paw edema method. Compounds 3d (84.09%), 3g (79.54%), and 3a (70.45%) demonstrated significant anti-inflammatory activity compared to the standard drug ibuprofen (65.90%) and were also found to be safer than ibuprofen, by ulcerogenic studies. A docking study was done using the crystal structure of human COX-2, to understand the binding mechanism of these inhibitors to the active site of COX-2. We previously reported 2-aminobenzoxazole analogue 1 as a potent ChemR23 inhibitor. Various substituents at the 4-position of the benzoxazole ring exhibited potent in vitro bioactivity, while those at the 6-position were not tolerated. Among substituents, a carboxyl group was identified as key for improving the oral bioavailability in cynomolgus monkeys. Compound 38a with the acidic part changed from a tetrazole group to a 1,2,4-oxadiazol-5-one group to improve bioactivity and pharmacokinetic parameters exhibited inhibitory activity against chemerin-induced chemotaxis in vitro. In addition, we confirmed the ChemR23 internalization of pDCs by compound 38a orally administered to cynomolgus monkeys. These 2-aminobenzoxazole-based ChemR23 inhibitors may be useful as novel immunotherapeutic agents capable of suppressing the migration of pDCs, which are known to be major producers of type I interferons in the lesion area of certain autoimmune diseases, such as systemic lupus erythematosus and psoriasis.
