Prion diseases are a group of neurodegenerative and fatal central nervous system disorders. The pathogenic mechanism involves the conversion of cellular priori protein (PrPC) to an altered scrapie isoform (PrPSc), which accumulates in amyloid deposits in the brain. However, no therapeutic drugs have demonstrated efficacy in clinical trials. We previously reported that BMD42-29, a synthetic compound discovered in silico, is a novel anti-prion compound that inhibits the conversion of PrPC to protease K (PK)-resistant PrPSc fragments (PrPres). In the present study. 14 derivatives of BMD42-29 were obtained from BMD42-29 by modifying in the side chain by in silico feedback, with the aim to determine whether they improve anti-priori activity. These derivatives were assessed in a PrPSc-infected cell model and some derivatives were further tested using real time-quaking induced conversion (RT-QuIC). Among them, BMD42-2910 showed high anti-prion activity at low concentrations in vitro and also no toxic effects in a mouse model. Interestingly; abundant PrPres was reduced in brains of mice infected with prion strain when treated with BMD42-2910, and the mice survived longer than control mice and even that treated with BMD42-29. Finally, high binding affinity was predicted in the virtual binding sites (Asn159, Gln 160, Lys194, and Glu196) when PIPC was combined with BMD-42-2910. Our findings showed that BMD42-2910 sufficiently reduces PrPres generation in vitro and in vivo and may be a promising novel anti-prion compound.

We have developed herein a new approach to the diverse synthesis of novel derivatives of 3-aryl benzo[d]thiazole-2(3H)-imines (3a-g), by a two-component reaction between diazonium salt (2) and various synthesized N-acyl-N’-aryl thioureas (1a-g), in the presence of sodium tert-butoxide as strong base. Finally, it resulted in the production of the desired products with a moderate yield. The chemical structures of these synthesized compounds were confirmed by various physico-chemical methods viz. Herein, we report the fabrication of multiwalled carbon nanotube (MWCNT) conjugates of cobalt phthalocyanines (CoPcs) with peripherally substituted Flavone (flay) or benzoxazole (bo) groups. The CoPc-X-f-MWCNT (X = flay or bo) conjugates were characterized by Transmission Electron Microscopy (TEM) and Infrared (IR) spectroscopy. The respective nanocomposite materials were drop-casted onto the surfaces of glassy carbon electrodes (GCEs) and the resultant chemically modified electrodes (CMEs) were used as electrocatalytic detectors for acetaminophen. Under optimized conditions, the peak currents for CoPc-flay-f-MWCNTs- and CoPc-bo-f-MWCNTs- GCEs were linear in wide acetaminophen concentration ranges of 1-1000 mu M and 15-1000 mu M, respectively. Electron transfer kinetics of the CMEs were superior than the bare electrode where diffusion- and convection-controlled electrocatalytic constants in the order of 10(2) and 10(4) M-1 s(-1) were obtained for both the CMEs. The CMEs showed good selectivity for APAP when tested in the presence of other emerging water pollutants.
