Unexpected Rearrangement Reaction and Synthesis of Benzoxazoles

Novel series of rearrangement reactions were herein reported that enable access to a variety of unique 2-aryl-3-(3′-oxobutenyl)-benzoxazole compounds 6a similar to 6f from 2-aminophenol, aromatic aldehyde and 3-butyn-2-one as materials by nucleophilic conjugate addition, dehydration and rearrangement reactions and intramolecular cyclization in the presence of a catalytic amount of CH3COOH in CH2Cl2 at ambient temperature. On the basis of products and intermediate products, a series of possible mechanism was presented and theoretically verified by density functional theory (DFT) method at B3LYP/6-31G (d,p) level from both molecular energy and atomic charge in Gaussian 03 package. The results show that the theory and experiment consistently explain the rationality of the reaction mechanism. The mechanism of rearrangement provides a basis for further study of this type of reaction. The advantage of this method is that a novel structure of benzoxazole derivative was synthesized successfully via a series of rearrangement reactions. Therefore, this method can be used as an attractive strategy for practical synthesis of nitrogen heterocyclic compounds.

New Approach for Controllable Synthesis of N-MnOx Microflowers and Their Superior Catalytic Performance for Benzoxazole Synthesis

Condensation of 2-amino-phenols with carboxylic acid derivatives has been practically used for the production of benzoxazoles, wherein the homogeneous catalytic system including strong acids and dangerous peroxides is generally employed. Herein, we report the novel approach for controlled synthesis of nitrogen-doped MnOx (denoted as N-MnO2, N-Mn5O8, and N-Mn3O4), by adjusting the heteroatom nitrogen amount, and uncover that the N-MnO2 catalyst as a sustainable and cost-effective heterogeneous catalyst exhibits high catalytic performance for condensation of 2-amino-phenols and o-phenylenediamine with alcohols into the corresponding benzoxazoles and benzimidazole, respectively. The N-MnO2 catalyst displays >99.9% yield for benzoxazole synthesis under room temperature and no decay (10 cycles), compared with the neglect activity (similar to 0%) for MnO2 catalysts. X-ray absorption spectroscopies and experimental studies uncovered that oxygen vacancies generated by heteroatom N doping play a key role for promoting intramolecular oxidative dehydrogenation of alcohol and 2-aminophenol derivatives to directly yield desired products. N-MnO2 catalyst rapidly oxidative dehydrogenated the reactants into corresponding benzoxazoles for 10 examples with >87.6% yields.

Design, synthesis and in vitro evaluation of 6-amide-2-aryl benzoxazole/benzimidazole derivatives against tumor cells by inhibiting VEGFR-2 kinase

Herein, we have carried out a structural optimization campaign to discover the novel anti-tumor agents with our previously screened YQY-26 as the hit compound. A library of thirty-seven 6-amide-2-aryl benzoxazole/benzimidazole derivatives has been designed and synthesized based on the highly conserved active site of VEGFR-2. Several title compounds exhibited selective inhibitory activities against VEGFR-2 than EGFR kinases, which also displayed selective anti-proliferation potency against the HUVEC and HepG2 than the A549 and MDA-MB-231 cancer cell lines. The newly synthesized compounds were evaluated for anti-angiogenesis capability by chick chorioallantoic membrane (CAM) assay. Among them, compounds 9d showed the most potent anti-angiogenesis ability (79% inhibition at 10 nM/eggs), the efficient cytotoxic activities (in vitro against the HUVEC and HepG2 cell lines with IC50 values of 1.47 and 2.57 mu M, respectively), and excellent VEGFR-2 kinase inhibition (IC50 = 0.051 mu M). The molecular docking analysis revealed that compound 9d is a Type 11 inhibitor of VEGFR-2 kinase. These results indicated that the 6-amide-2-arylbenzoxazole and 6-amide-2-aryl benzimidazole derivatives are promising inhibitors of VEGFR-2 kinase for the potential treatment of anti-angiogenesis. In-vitro Anti-cancer assay and apoptotic cell pathway of newly synthesized benzoxazole-N-heterocyclic hybrids as potent tyrosine kinase inhibitors

A series of benzoxazole-N-heterocyclic hybrids have been synthesized by a one-pot strategy. Molecular docking study revealed that such compounds have the ability to inhibit enzyme protein tyrosine kinase. The findings of this work have been the successful synthesis of benzoxazole scaffolds, featuring hybrids of benzoxazole with quinoline and quinoxaline respectively. The molecular docking studies have showed these compounds to be inhibitors of tyrosine kinase enzyme which triggers growth of cancer cells. The cytotoxicity study of compounds 4a-f showed better potency against breast cancer cell lines MCF-7 and MDA-MB-231 in contrast to oral and lung cancer cell lines KB and A549. The tyrosine kinase activity was measured using Universal Tyrosine Kinase Assay kit using horseradish peroxide (HRP)-conjugated anti-phosphotyrosine kinase solution as a substrate. The compounds 4c exhibited maximum inhibition in the activity of enzyme tyrosine kinase with IC50 value 0.10 +/- 0.16 mu M, than other compounds which were studied and thus proved to be inhibitors of enzyme tyrosine kinase. The selective index of all four compounds was found out to be greater than two, indicating the non-toxic behaviour, i.e. good anti-cancer activity. Further, fluorescence microscopic study helped to characterize the mode of cell death, which was found to be late apoptosis as indicated by the orange fluorescence. The SAR analysis has also been carried out.

New Coumarin-benzoxazole derivatives: Synthesis, photophysical and NLO properties

In this work, a series of novel D-pi-A structure coumarin-benzoxazole derivatives have been synthesized. The third-order NLO properties of the coumarin derivatives in DMSO are investigated by Z-scan measurement, and the photophysical properties of the compounds are studied through experimental and theoretical calculation methods. The results showed that the introducing specific functional groups at multiple active sites of coumarin reduced the pi-pi intermolecular stacking, while the formation of push-pull structure decreased E-gap of entire molecule. The excellent third-order NLO responses based on chromophore 4c with maximum nonlinear absorptive coefficients is 50.64 x 10(-13) esu and the largest second-order NLO hyperpolarizability is 7.12 x 10(-30) esu. Experimental results have also been confirmed through DFT, TDDFT and ZINDO calculations. Our results indicate that the coumarin fluorescent dyes are promising candidate materials for NLO applications.

Thermal induced structural changes of polyhydroxyamide by two-dimensional (2D) infrared correlation study

Two-dimensional (2D) correlation analysis of in situ MR spectra was used to probe the thermally induced structural changes in a poly(hydroxyamide) (PHA) precursor prepared from the reaction of 2,2-bis(3-amino-4-hydroxyphenyl)hexafluoropropane (Bis-AP-AF) with terephthaloyl chloride. Large spectral changes in the in situ FTIR spectra of the PHA precursor film were observed in the range of 200-300 degrees C. The thermal cyclodehydration reaction of the PHA precursor film strongly affects the spectral changes corresponding to the amide group and the adjacent phenyl ring in the Bis-AP-AF unit. The thermal cyclodehydration reaction of the PHA precursor film in the range of 240-300 degrees C induced the spectral changes in amide linkage before the formation of the benzoxazole ring. 2-Aminobenzimidazole and -benzoxazole as N-nucleophile in palladium-catalysed aminocarbonylation

Palladium-catalysed aminocarbonylation of aryl iodides in the presence of 2-aminobenzimidazole and 2-aminobenzoxazole as N-nucleophile was carried out. Antiinflammatory and Antiproliferative Activity of Naphthoxazole, Fused Hetero-benzoxazole and Bridged Benzobicyclic Photoproducts

Biological activity of naphthoxazoles, fused hetero-benzoxazoles and benzobicyclo[3.2.1]-derivatives was investigated in proliferation and inflammation based assays. The tested compounds were prepared by photocylization or photocycloaddition reactions. Effect of compounds on proliferation of several cancer cell lines was determined by measuring cell metabolic activity through time. Lipopolysaccharide (LPS) stimulation of peripheral blood mononuclear cells (PBMC) was used to investigate antiinflammatory properties of the compounds. Several naphthoxazoles and fused hetero-benzoxazoles inhibited TNF alpha protein expression in LPS stimulated PBMC, indicating possible antiinflammatory role which would be interesting to further investigate. Physico-chemical properties of tested compounds have been also studied using chromatographic lipophilicity measure, chrom logD and logP was calculated as the importance of physico-chemical properties of compounds at early stage of discovery of new drugs is well established. The similarities in structure and activity of some representative compounds affirm the need to further address their antiinflammatory properties.

New Fluorescent Nickel(II) Complexes as a Catalyst for Biodiesel Formation: Synthesis, Structure, Spectral Properties, and DFT Calculations

Two fluorescent heterocyclic ligands were obtained by reduction of imidazo[4,5-e][2,1]benzoxazole derivatives with Fe/HCl. New fluorescent nickel(II) complexes were synthesized by coordination of Ni(II) cation with the heterocyclic ligands. The structure of the complexes was established by spectral and analytical data, Job plot, and DFT calculations. Photophysical properties of Ni(II) complexes were characterized by UVVis and fluorescence spectroscopy. The optimized geometry, spectral properties, and energy difference between the HOMO and LUMO frontier orbitals of the ligands and Ni(II) complexes were obtained by DFT calculations at the B3LYP/6-311++G(d, p) level. The calculated spectral properties are in good agreement with the experimental values. The complexes were also tested as homogeneous catalysts for the transesterification of corn oil with methanol. The structure of the obtained product was confirmed by 1H NMR analysis. The catalytic results showed that the new Ni(II) complexes can be considered as potential candidates for the development of new catalytic systems for biodiesel production.

SYNTHESIS, CHARACTERIZATION AND BIOLOGICAL EVALUATION OF SOME 2-ARYLBENZOXAZOLE ACETIC ACID DERIVATIVES AS PROMISING ANTICANCER AGENTS

The synthesis of a series of 2-arylbenzoxazole compounds possessing a cytotoxic activity as potential anticancer agents was achieved. Oxidative coupling of benzaldehyde with o-aminophenol utilizing lead tetraacelate approach was used to realize the synthesis of compounds 1-11. The cytotoxicity of 1-11 was screened against breast cancer cell line MCF-7 and human colon cancer cell line HCT-116 utilizing doxorubicin as a reference drug. Among these compounds, 2-(3-benzyloxyphenyl)benzoxazole-5-acetic acid (5) and 2-(4-methoxyphenyl)benzoxazol-5-acetic acid (10), were found to be promising cytotoxic compounds against the MCF-7 cell line. In addition, this study showed that the presence of an acetic acid group at position 5 of the benzoxazole nucleus enhances the activity. Moreover, we noticed that the presence of an oxygen atom directly linked to the phenyl substituent improves activity as well. These results offer a new benzoxazole based template to design and develop novel antineoplastic agents.
