In this report, we present a new benzoxazole derivative endowed with inhibitory activity against the HIV-1 nucleocapsid protein (NC). NC is a 55-residue basic protein with nucleic acid chaperone properties, which has emerged as a novel and potential pharmacological target against HIV-1. In the pursuit of novel NC-inhibitor chemotypes, we performed virtual screening and in vitro biological evaluation of a large library of chemical entities. We found that compounds sharing a benzoxazolinone moiety displayed putative inhibitory properties, which we further investigated by considering a series of chemical analogues. This approach provided valuable information on the structure-activity relationships of these compounds and, in the process, demonstrated that their anti-NC activity could be finely tuned by the addition of specific substituents to the initial benzoxazolinone scaffold. A novel series of benzylpyridinium-based benzoheterocycles (benzimidazole, benzoxazole or benzothiazole) were designed as potent acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) inhibitors. The title compounds 4a-q were conveniently synthesized via condensation reaction of 1,2-phenylenediamine, 2-amino-phenol or 2-aminothiophenol with pyridin-4-carbalehyde, followed by N-benzylation using various benzyl halides. The results of in vitro biological assays revealed that most of them, especially 4c and 4g, had potent anticholinesterase activity comparable or more potent than reference drug, donepezil. The kinetic study demonstrated that the representative compound 4c inhibits AChE in competitive manner. According to the ligand-enzyme docking simulation, compound 4c occupied the active site at the vicinity of catalytic triad. The compounds 4c and 4g were found to be inhibitors of A beta self-aggregation as well as AChE-induced A beta aggregation. Meanwhile, these compounds could significantly protect PC12 cells against H2O2-induced injury and showed no toxicity against HepG2 cells. As multi-targeted structures, compounds 4c and 4g could be considered as promising candidate for further lead developments to treat Alzheimer’s disease.

The new experimental results of the dually emitting ESIPT molecule, the 2,5-bis(2′-benzoxazolyl)hydroquinone (BBHQ) are presented. The results of the XRD analysis are the following: the unit cell of the monoclinic symmetry contains the perfectly planar BBHQ molecules, with two O-H center dot center dot center dot N intramolecular hydrogen bonds. There are two molecules in the asymmetric part of the cell, one the single BBHQ structure and the other being superposition of the main component with the site occupation factor (sof) 77.4(6)% and the minor one: sof = 22.6(6)%- the same structure rotated by 180 degrees along the long axis of the molecule. The remarkable agreement of the experimentally determined molecular structure with theoretical results reported in the literature, was observed. The dispersed fluorescence spectra of jet cooled molecule, covering two fluorescence bands were measured. It was observed that the intensity ratio of both emissions was dependent on excitation by subsequent quanta of the leading vibration of 114 cm(-1). The height of the barrier separating the two emitting tautomers was estimated as similar to 340 cm(-1). Moreover, the two independent experimental tools, the double-resonance fluorescence depletion (DFD) and mass selected two photon ionization (R2PI) techniques were applied to the partially deuterated jet cooled BBHQ enabling the separation and identification of four different deuterated species, some not known until now. Three novel copper(II) complexes, [Cu(Gly-l-Val)(HPBM)(H2O)]ClO4H2O (1), [Cu(Gly-l-Val)(TBZ)(H2O)]ClO4 (2) and [Cu(Gly-l-Val)(PBO)(H2O)]ClO4 (3) (Gly-l-Val=glycyl-l-valine anion, HPBM=5-methyl-2-(2-pyridyl)benzimidazole, TBZ=2-(4-thiazolyl)benzimidazole, PBO=2-(2-pyridyl)benzoxazole), have been prepared and characterized with elemental analyses, conductivity measurements as well as various spectroscopic techniques. The interactions of these copper complexes with calf thymus DNA were explored using UV-visible, fluorescence, circular dichroism, thermal denaturation, viscosity and docking analyses methods. The experimental results showed that all three complexes could bind to DNA via an intercalative mode. Moreover, the cytotoxic effects were evaluated using the MTT method, and the antimicrobial activity of these complexes was tested against Bacillus subtilis, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. The results showed that the activities are consistent with their DNA binding abilities, following the order of 1 > 2 > 3.

Since thermally rearranged (TR) polymers were known as high gas permeable and processable materials, fabricating high performance hollow fiber (HF) membranes have been tried using them. However, an unexpected drawback emerged which is the gas productivity loss by thermal densification of skin layers during thermal treatment above their glass transition temperature (T-g). In this work, we used a recently reported crosslinked-TR (XTR) polybenzoxazole to develop a new class of high-flux TR hollow fibers by directly exploiting the thermal densification phenomenon. The TR temperature range (320-460 degrees C) and T-g (394 degrees C) were determined by thermal gravimetric analysis (TGA) and dynamic mechanical analysis (DMA). The chain rigidity of the XTR polymer increased during an isotherm treatment at its T-g, suggesting a restricted densification. Surprisingly, the undesired pinhole-defects (pore diameter < 5 nm) on precursor fibers were perfectly healed after thermal treatment (> 400 degrees C), forming an ultrathin defect-free skin layer on thermally-densified XTR hollow fiber membranes. The pore-healed XTR hollow fibers exhibited an outstanding CO2 permeance of similar to 2300 GPU and a CO2/N-2 selectivity of 17.4 with a skin thickness of 103 nm.
