A novel series of benzoxazole linked combretastatin derivatives (11a-11n) have been synthesized and confirmed by H-1 NMR, C-13 NMR, and Mass spectral analysis. The synthesized compounds (11a-11n) were screened for anticancer activity against three human cancer cell lines, Breast (MCF-7), Lung (A549), and Melanoma (A375). Most of the compounds exhibit moderate to potent anticancer activity. Among the compounds, 11g, 11h, 11l, 11m, and 11n showed more potent activity than the positive control Doxorubicin. In addition, compounds 11g, 11l, 11m, and 11n were carried out their molecular docking studies on EGFR receptor (PDB ID: 4hjo) and results indicated that 11g and 11l have strong binding interactions with the receptor. It was found that the binding energy calculations were in good agreement with the observed IC50 values.

A tridentate benzoxazole-containing aminophenol ligand NNOH2 was coordinated to Co and V metal centers and the electronic structure of the resultant complexes characterized by both experimental and theoretical methods. The solid state structure of the Co complex exhibits a distorted octahedral geometry with two tridentate ligands bound in meridional fashion, and coordination-sphere bond lengths consistent with a Co(iii) oxidation state. However, the metrical parameters are similar for both ligands in the solid state, and DFT calculations support delocalization of the ligand radical over both ligands, affording an intermediate ligand redox level Co(iii)(NNO1.5-)(NNO1.5-). The vanadyl complex exhibits a distorted octahedral geometry in the solid state consistent with a V(v) metal center and amidophenoxide (NNOAP), acetylacetonate and oxo ligands. The ligand metrical parameters are consistent with significant amidophenoxide to V(v) pi donation. Overall, our results highlight the roles of electron transfer, delocalization, and pi bonding in the metal complexes under study, and thus the complexity in assignment of the electronic structure in these systems.

To develop novel CNS penetrant HDAC inhibitors, a new series of HDAC inhibitors having benzoheterocycle were designed, synthesized, and biologically evaluated. Among the synthesized compounds, benzothiazole derivative 9b exhibited a remarkable anti-proliferative activity (GI(50) = 2.01 mu M) against SH-SY5Y cancer cell line in a dose and time-dependent manner, better than the reference drug SAHA (GI(50) = 2.90 mu M). Moreover, compound 9b effectively promoted the accumulation of acetylated Histone H3 and alpha-tubulin through inhibition of HDAC1 and HDAC6 enzymes, respectively. HDAC enzyme assay also confirmed that compound 9b efficiently inhibited HDAC1 and HDAC6 isoforms with IC50 values of 84.9 nM and 95.9 nM. Furthermore, compound 9b inhibited colony formation capacity of SH-SY5Y cells, which is considered a hallmark of cell carcinogenesis and metastatic potential. The theoretical prediction, in vitro PAMPA-BBB assay, and in vivo brain pharmacokinetic studies confirmed that compound 9b had much higher BBB permeability than SAHA. In silico docking study demonstrated that compound 9b fitted in the substrate binding pocket of HDAC1 and HDAC6. Taken together, compound 9b provided a novel scaffold for developing CNS penetrant HDAC inhibitors and therapeutic potential for CNS-related diseases.

Herein, ten approved cysteine covalent drugs were selected for exploring FBPase inhibitory activities, and the alcohol deterrent disulfiram displayed superior inhibitory efficacy among those drugs. Based on the structure of lead compound disulfiram, 58 disulfide-derived compounds were designed and synthesized for investigating FBPase inhibitory activities. Optimal compound 3a exhibited significant FBPase inhibition and glucose-lowering efficacy in vitro and in vivo. Furthermore, 3a covalently modified the C128 site, and then regulated the N125-S124-S123 allosteric pathway of FBPase in mechanism. In this article, new fluorescent lipophilic based benzazoles were synthesized from the reaction between photoactive formyl derivatives and aliphatic amines followed by NaBH4 reduction with good yields. The photophysics of the benzazoles was investigated experimentally and theoretically. These compounds present absorption maxima in the UV region (similar to 339 nm) and fluorescence emission maxima in the cyan to green region with a large Stokes shift (similar to 175 nm) due to a proton transfer process in the excited state. Two fluorophores were successfully used as a proof of concept to produce stable photoactive liposomes prepared from phosphatidylcholine (PC) and were characterized by zeta potential, small angle X-ray scattering (SAXS), FTIR and UV-Vis experiments (turbidity). The scattering data indicate that the presence of compounds 20 and 23 reduces the overall surface charge of the PC vesicles, possibly due to the partial neutralization of phosphatidic acid and/or phosphatidylinositol phosphate by the amine groups, and they also modify the structural features of the assemblies, leading, in particular, to a reduction in the thickness of the hydrophobic inner segment (t(t)) of the liposomes. DFT and TD-DFT calculations were performed with the omega B97XD functional. Geometric analyses show that the 2-(2′-hydroxyphenyl) benzazolic planar portion allows an effective pi pi* electronic transition. Additionally, the calculations indicate a small energy barrier to proton transfer. The results of the absorption and emission maxima show a slight solvent influence on the wavelengths.

In this study, a novel series of 4-(2-(alkylthio)benzo[d]oxazol-5-yl)-2,4-dihydro-3H-1,2,4-triazol-3-ones (4a-m) was designed and synthesized. The anticonvulsant activities of these compounds were evaluated by using the maximal electroshock seizure (MES) and subcutaneous pentylenetetrazole (scPTZ) seizure models in mice. In an ELISA test, compound 4g significantly increased the gamma-aminobutyric acid (GABA) content in mouse brain. In addition, pretreatment with thiosemicarbazide (an inhibitor of the GABA synthesizing enzyme) significantly decreased the activity of 4g in the MES model, which suggests that the mechanism through which compound 4g elicits its anticonvulsive action is at least in part through increasing the GABA level in the brain.

A one-pot direct transformation to remotely C-H alkene functionalized 2-aryl benzoxazoles from the reaction of amidophenol and electronically deficient olefin was reported.
