Developing a large database of polymers structures and properties, for which suitable polymer structural models are a prerequisite, is critical for polymer informatics. We present a simple strategy, referred to as polymer structure predictor (PSP), for predicting the crystal structural models of linear polymers, given their chain-level atomic connectivity information. The PSP, which was designed specifically for polymers, relies on properly defining and sampling the configuration space. Using this approach, we have successfully recovered eight known crystal structures of six common linear polymers, including polyethylene, polyvinylidene fluoride, poly(vinyl chloride), poly(p-phenylene sulfide), polyacrylonitrile, and poly-2,5-benzoxazole, while discovering some new stable structures of three of them, i.e., polyvinylidene fluoride, polyacrylonitrile, and poly(p-phenylene sulfide). The PSP is very simple, highly scalable, suitable for automatic workflows, and comparable to the best major structure prediction method in terms of efficiency in polymer crystal structure prediction. Although challenges in comprehensively accounting for possible chain-level conformations remain, the PSP will be very useful in efficiently generating polymer data and strengthening the emerging polymer informatics ecosystems.

Four new heteroleptic [Cu(NN)P-2](+)-type cuprous complexes-1-TPP, 2-POP, 3-Xantphos, and 4-DPPF-were designed and synthesized using a diimine ligand 2-(2 ‘-pyridyl)benzoxazole (2-PBO) and different phosphine ligands (TPP, triphenylphosphine; POP, bis[2-(diphenylphosphino)phenyl]ether; Xantphos, 4,5-bis(diphenylphosphino)-9,9-dimethylxanthene; DPPF, 1,1 ‘-bis(diphenylphosphino)-ferrocene). All complexes were characterized using single-crystal X-ray diffraction, spectroscopic analysis (infrared, UV-Vis.), elemental analysis, and photoluminescence (PL). Single-crystal X-ray diffraction revealed complexes 1-4 as isolated cation complex structures with a tetrahedral CuN2P2 coordination geometry and diverse P-Cu-P angles. Their UV-Vis. absorption spectra exhibited a blue-shift sequence in wavelength with an enlarged P-Cu-P angle from 4 to 2 then to 3 and then to 1. The PL emission peaks of 1-3 also exhibited a similar blue-shift sequence (2 -> 3 -> 1). Their PL lifetime in microseconds (7.5, 5.1, and 4.7 mu s for 1, 2, and 3, respectively) indicated that their PL behavior represents phosphorescence. Time-dependent density functional theory (TD-DFT) calculation and wavefunction analysis revealed that S-1 and T-1 states of 1-3 should be assigned as metal-ligand and ligand-ligand charge-transfer (ML + L’L)CT states. Their UV-Vis. absorption and phosphorescence should be attributed to the charge transfer from the P-Cu-P segment to the 2-PBO ligand. Therefore, as the P-Cu-P angle increased (lower HOMO), the energy of S-1 and T-1 states also increased, following the change of PL color.

The experimental results showed that all three complexes could bind to DNA via an intercalative mode. Moreover, the cytotoxic effects were evaluated using the MTT method, and the antimicrobial activity of these complexes was tested against Bacillus subtilis, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. The results showed that the activities are consistent with their DNA binding abilities, following the order of 1 > 2 > 3.

In this study we present a novel and simple approach to improve the gas separation performance of the thermally rearranged membranes, which involves doping the polyimide precursors (HPI) with ionic liquid (IL), and carrying out its degradation along with the conversion process to polybenzoxazole (PBO) in order to facilitate the formation of larger and /or more numerous free volumes. A series of aromatic (co)poly(hydroxyimide)s based on 6FDA and HAB/4MPD diamines in different molar ratios, as well as BPADA-HAB poly(hydroxyimide) were synthesized as the precursors to be doped with IL and thermally rearranged to PBO. The structural modifications of the precursor backbone were applied to study the impact of IL on the physical properties, thermal conversion process, as well as gas transport properties of the doped polymers with different chain rigidities. The pure and IL doped (co)polyimides and their thermally rearranged counterparts were characterized by WAXD, DSC, TGA, tensile tests, and PALS, and examined in pure gas permeation experiments. TR conversion temperature was considerably reduced by IL doping (e.g. by 126 degrees C). This effect depended on the precursor chemical structure and the IL content. After thermal rearrangement of the IL doped HPIs, the membrane permeability to gases increased significantly compared to the un-doped analogues (e.g. 2 fold increase for O-2 permeability). The permeability increase was larger for the higher IL content and the precursor chain rigidity. This was accompanied by a relatively small loss in selectivity leading to the performance shift towards the 2008 upper bound. The differences in permeability among the samples correlated with the free volume size from PALS. In particular, a very good correlation was obtained (r(2) = 0.958), when the data fitted to the Cohen-Turnbull model concerned only PBO samples with low cohesive energy density. Further studies on HPIs doped with IL with lower degradation temperatures are suggested to mitigate polymer degradation and explore this new method for the design of improved gas separation membrane materials.

JMJD3 is a member of the KDM6 subfamily and catalyzes the demethylation of lysine 27 on histone H3 (H3K27). This protein was identified as a useful tool in understanding the role of epigenetics in inflammatory conditions and in cancer as well. Guided by a virtual fragment screening approach, we identified the benzoxazole scaffold as a new hit suitable for the development of tighter JMJD3 inhibitors. The compound 3b (79.54%), 31 (75.00%), 3m (72.72%) and 3d (68.18%) exhibited significant anti-inflammatory activity than standard drug ibuprofen (65.90%). Ulcerogenic activity with histopathological studies was performed, and the screened compounds demonstrated significant gastric tolerance than ibuprofen. Molecular Docking study was also performed with resolved crystal structure of COX-2 to understand the interacting mechanisms of newly synthesized inhibitors with the active site of COX-2 enzyme and the results were found to be in line with the biological evaluation studies of the compounds. Novel series of benzoxazoles 4(a-f)-16 were designed, synthesized, and evaluated for anticancer activity against HepG2, HCT-116, and MCF-7 cells. HCT-116 was the most sensitive cell line to the influence of the new derivatives. Compounds 5(c), 5(f), 6(b), 5(d), and 6(c) showed the highest anticancer activities against HepG2 cells with IC50 of 5.93 +/- 0.2, 6.58 +/- 0.4, 8.10 +/- 0.7, 8.75 +/- 0.7, and 9.95 +/- 0.9 mu M, respectively; HCT-116 cells with IC50 of 7.14 +/- 0.4, 9.10 +/- 0.8, 7.91 +/- 0.6, 9.52 +/- 0.5, and 12.48 +/- 1.1 mu M, respectively; and MCF-7 cells with IC50 of 8.93 +/- 0.6, 10.11 +/- 0.9, 12.31 +/- 1.0, 9.95 +/- 0.8, and 15.70 +/- 1.4 mu M, respectively, compared with sorafenib as a reference drug with IC50 of 9.18 +/- 0.6, 5.47 +/- 0.3, and 7.26 +/- 0.3 mu M, respectively. The most active compounds 5(c-f) and 6(b,c) were further evaluated for their vascular endothelial growth factor receptor-2 (VEGFR-2) inhibition. Compounds 5(e) and 5(c) potently inhibited VEGFR-2 at lower IC50 values of 0.07 +/- 0.01 and 0.08 +/- 0.01 mu M, respectively, compared with sorafenib (IC50 = 0.1 +/- 0.02 mu M). Compound 5(f) potently inhibited VEGFR-2 at low IC50 value (0.10 +/- 0.02 mu M) equipotent to sorafenib. Our design was based on the essential pharmacophoric features of the VEGFR-2 inhibitor sorafenib. Molecular docking was performed for all compounds to assess their binding pattern and affinity toward the VEGFR-2 active site.
