New steroidal oxazolines, benzoxazoles and benzimidazoles related to abiraterone and galeterone

Seven new oxazoline, benzoxazole and benzimidazole derivatives were synthesized from 3 beta-acetoxyandrosta-5,16-dien-17-carboxylic, 3 beta-acetoxyandrost-5-en-17 beta-carboxylic and 3 beta-acetoxypregn-5-en-21-oic acids. Docking to active site of human 17 alpha-hydroxylase/17,20-lyase revealed that all oxazolines, as well as benzoxazoles and benzimidazoles comprising Delta(16) could form stable complexes with enzyme, in which steroid moiety is positioned similarly to that of abiraterone and galeterone, and nitrogen atom coordinates heme iron, while 16,17-saturated benzoxazoles and benzimidazoles could only bind in a position where heterocycle is located nearly parallel to heme plane. Modeling of the interaction of new benzoxazole and benzimidazole derivatives with androgen receptor revealed the destabilization of helix 12, constituting activation function 2 (AF2) site, by mentioned compounds, similar to one induced by known antagonist galeterone. The synthesized compounds inhibited growth of prostate carcinoma LNCaP and PC-3 cells at 96 h incubation; the potency of 2′-(3-hydroxyandrosta-5,16-dien-17-yl)-4′,5′-dihydro-1′,3′-oxazole and 2′-(3 beta-hydroxyandrosta-5,16-dien-17-yl)-benzimidazole was superior and could inspire further investigations of these compounds as potential anti-cancer agents.

Chromogenic properties of 2-(2-carbomethoxy-3,4-dichloro-6-hydroxyphenyl)benzoxazole and its Zn (II) and Cd(II) complexes

The chromogenic properties of 2-(2-carbomethoxy-3,4-dichloro-6-hydroxyphenyl)benzoxazole (1) were studied. Solvatochromism of 1 was observed at keto-enol equilibrium due to the ground state intramolecular proton transfer (GSIPT). It was found that solvents with a good hydrogen bond acceptor (beta) ability were able to stabilise the keto form of 1. The excited-state intra molecular proton transfer (ESIPT) in molecules of 1 resulted in intense fluorescence with an anomalous Stokes shift of up to 8493 cm 1 and quantum yields of 0.08-0.19. The keto-form of 1 exhibits negative T-type photochromism with a thermally reversible photoconversion to the enol form. The complexes of ligand 1 with Zn(II) and Cd(II) were synthesised. According to X-ray structural analysis, the zinc complex is the neutral complex, which is formed by two oxazole-N and two phenol O donors of ligand 1 which crystallise in a centrosymmetric group with the zinc ion located on the inversion centres. The synthesised complexes possess effective blue fluorescence and resistance to photodegradation.

Design and synthesis of new benzoxazole/benzothiazole-phthalimide hybrids as antitumor-apoptotic agents

Herein, we synthesized a series of twelve benzoxazole and benzothiazole derivatives incorporated with phthalimide core as anticancer agents. The most active compounds were 5a and 5g against HepG2 and MCF7 cell lines with IC50 = 0.011 and 0.006 mu M, respectively. They evaluated against EGFR and HER2 enzymes. From cell cycle analysis, it was observed that test compounds exerted pre G1 apoptosis and cell cycle arrest at G2/M phase. The achieved results suggested that apoptosis was due to activation of caspase-7 and caspase-9. EGFR was chosen as a biological target for carrying molecular modeling study for the newly synthesized compounds.

The discovery and optimization of a series of 2-aminobenzoxazole derivatives as ChemR23 inhibitors

A structural class of 2-aminobenzoxazole derivatives possessing biphenyltetrazole was discovered to be potent human ChemR23 inhibitors. We initially tried to improve the potency of compound 1, which was found through in-house screening using the human plasmacytoid dendritic cell (pDC)-like cell line CAL-1. The introduction of a chiral methyl moiety at a benzylic position in a center of compound 1 showed a large impact on the inhibitory activity against calcium signaling of ChemR23 induced by the natural ligand chemerin. As a result of further investigations at the benzylic position, (R)-isomer 6b was found to show a 30-fold increased potency over desmethyl compound 1. In addition, an extensive structure-activity relationship study on the benzoxazole moiety successfully led to a further increase in the potency. The antagonistic effect of the compounds was based on the induction of ChemR23 internalization. In addition, we observed that compound 31, which contained an amide moiety on benzoxazole, inhibited chemotaxis of CAL-1 cells induced by chemerin in vitro. These results suggest that our ChemR23 inhibitors are attractive compounds for the treatment of pDC-related autoimmune diseases, such as systemic lupus erythematosus and psoriasis.

The effects of the heteroatom and position on excited-state intramolecular proton transfer of new hydroxyphenyl benzoxazole derivatives: a time-dependent density functional theory study

The effects of the heteroatom and position on excited-state intramolecular proton transfer (ESIPT) of 2-[4 ‘-(N-4,6-dichloro-1,3,5-triazi-n-2-yl)2 ‘ hydroxyphenyl]benzoxazole (4THBO) have been investigated via time-dependent density functional theory studies. The heteroatoms refer to O and S atoms, and the position effect refers to the N-4,6-trichloro-1,3,5-triazin-2-yl (TCT) substituents in the para and meta positions. The configuration of the four compounds (4THBO, 4THBT, 5THBO and 5THBT) was optimized and the bond lengths, bond angles and infrared spectra of the atoms participating in the proton transfer in the S0 and S1 states were studied. The occurrence of ultrafast ESIPT in the four compounds was demonstrated. Moreover, the potential energy curves of the S0 and S1 states were constructed, and the effects of the heteroatom substitution and substituent position changes on the ESIPT mechanism of the four 4THBO derivatives were analyzed. The results show that the ESIPT barrier of the S atom substitution in the excited state is lower than that of the O atom-substituted molecule, and the energy barrier of the substituent (TCT) in the meta-position is significantly smaller than that in the para-position. These results indicate that the substitution of the S heteroatom promotes the ESIPT of the 4THBO compound and that the substituent (TCT) in the para position is more prone to proton transfer than that in the meta position. Our work could provide a theoretical basis for further experiments.

Facile synthesis of 2-benzoxazoles via CuI/2,2′-bipyridine catalyzed intramolecular C-O coupling of 2-haloanilides

Development of newer methods for the synthesis of Benzoxazoles has of greater interest due to their wide range of biological activities and pharmaceutical importance. We herein report a facile and general method for the synthesis of 2-substituted Benzoxazoles via copper catalyzed intramolecular C-O cross-coupling of 2-haloanilides. A combination of CuI (5 mol%), 2,2′-bipyridine (10 mol%), Cs2CO3 (2 equiv.) in DMF solvent with 4 angstrom molecular sieves at 140 degrees C, illustrated the scope for tuning the reactivity of 2-haloanilides toward the selective formation of a series of 2-alkyl benzoxazole derivatives in moderate to good yields. This is the first systematic study using CuI/2,2′-Bipyridine as the catalytic system for the synthesis of 2-substituted Benzoxazoles. The outcome of the reaction was found to be significantly influenced by the aromatic and amide substituents of 2-haloanilides. [GRAPHICS]

Tunable Luminescent Properties of Tricyanoosmium Nitrido Complexes Bearing a Chelating O boolean AND N Ligand

We have recently reported a strongly luminescent osmium(VI) nitrido complex [Os-VI(N)(NO2-L)(CN)(3)](-) [HNO2-L = 2-(2-hydroxy-5-nitrophenyl)benzoxazole]. The excited state of this complex readily activates the strong C-H bonds of alkanes and arenes (Commun. Chem. 2019, 2, 40). In this work, we attempted to tune the excited-state properties of this complex by introducing various substituents on the bidentate L ligand. The series of nitrido complexes were characterized by IR, UV/vis, H-1 NMR, and electrospray ionization mass spectrometry. The molecular structures of five of the nitrido compounds have been determined by X-ray crystallography. The photophysical and electrochemical properties of these complexes have been investigated. The luminescence of these nitrido complexes in the solid state, in a CH2Cl2 solution, and in a CH2Cl2 solid matrix at 77 K glassy medium clearly shows that these emissions are due to (LML)-L-3’CT [L ligand to Os (math)N] phosphorescence. The presence of strongly electron-withdrawing substituents in these complexes enhances the LML’CT emission. Our result demonstrates that the excited-state properties of this novel class of luminescent osmium(VI) nitrido complexes can be fine-tuned by introducing various substituents on the bidentate L ligand.
