Synthesis, structure, electrochemical properties and superoxide radical scavenging activities of two thiocyanate copper(II) complexes with different pyridyl-benzoxazole ligands

Reaction of 2-(2′-pyridyl)benzoxazole (2-PBO) or 2-(4′-pyridyl)benzoxazole (4-PBO) ligands with CuSCN afforded two thiocyanate copper (II) complexes, Cu(2-PBO) (SCN)(2) (1) and Cu(4-PBO)(2)(SCN)(2) (2), have been characterized by elemental analysis, UV-Vis, IR spectra and single-crystal X-ray diffraction. The structural analysis reveals that although the structures of complexes 1-2 are both four coordinated and show plane quadrilateral structure, the distorted of complex 1 is greater than 2. The cyclic voltammogram of complexes 1-2 represent quasi-reversible Cu2+/Cu+ pairs. The superoxide radical scavenging test in vitro showed that complex 1-2 had significant antioxidant activity on superoxide radicals, and the activity of complex 2 was higher than that of 1. The crystal structure of AjiA1 reveals a novel structural motion mechanism in the adenylate-forming enzyme family

Adenylate-forming enzymes (AFEs) are a mechanistic superfamily of proteins that are involved in many cellular roles. In the biosynthesis of benzoxazole antibiotics, an AFE has been reported to play a key role in the condensation of cyclic molecules. In the biosynthetic gene cluster for the benzoxazole AJI9561, AjiA1 catalyzes the condensation of two 3-hydroxyanthranilic acid (3-HAA) molecules using ATP as a co-substrate. Here, the enzymatic activity of AjiA1 is reported together with a structural analysis of its apo form. The structure of AjiA1 was solved at 2.0 angstrom resolution and shows a conserved fold with other AFE family members. AjiA1 exhibits activity in the presence of 3-HAA (K-m = 77.86 +/- 28.36, k(cat) = 0.04 +/- 0.004) and also with the alternative substrate 3-hydroxybenzoic acid (3-HBA; K-m = 22.12 +/- 31.35, k(cat) = 0.08 +/- 0.005). The structure of AjiA1 in the apo form also reveals crucial conformational changes that occur during the catalytic cycle of this enzyme which have not been described for any other AFE member. Consequently, the results shown here provide insights into this protein family and a new subgroup is proposed for enzymes that are involved in benzoxazole-ring formation.

TBET-based ratiometric fluorescent probe for Hg2+ with large pseudo-Stokes shift and emission shift in aqueous media and intracellular colorimetric imaging in live Hela cells

The detection of Hg2+ in biological systems and its imaging is of highly importance. In this work, a novel ratiometric fluorescence probe is developed based on through-bond energy transfer (TBET) with a 2-(2-hydroxyphenyl)benzoxazole (HBO) as donor and a Rhodamine derivative-Hg conjugate (RDM-Hg) as acceptor. Hg2+ weakens the fluorescence of HBO at 430 nm and meanwhile interacts with Rhodamine B derivative to form a fluorescent conjugate (RDM-Hg) giving rise to emission at 597 nm with a 167 nm red-shift. Further, the difference 282 nm between the donor absorption (315 nm) and the accepter emission (597 nm) for 1 + Hg2+ is comparable to the highest value of the Stokes shift (282 nm) reported earlier for other reported TBET-based cassette. Through-bond energy transfer from HBO to RDM-Hg is triggered by Hg2+ resulting in concentration dependent variation of fluorescence ratio I-597/I-430. A linear calibration of I-597/I-430 versus Hg2+ concentration is obtained within 0-5 mu M, along with the lowest detection limit being found to be as low as 1.31 x 10(-9) mol center dot l(-1) (similar to 0.26 ppb) for Hg2+. This feature is further demonstrated by colorimetric imaging of test strip and intracellular Hg2+. On the other hand, the HBO/RDM TBET sensing system is characterized by a combination of high sensitivity and selectivity. The present study provides an approach for further development of ratiometric probes dedicated to selective in vitro or in vivo sensing some species of biologically interest.

A facile and effective C-H functionalization strategy for the synthesis of 2-mercaptobenzothiazoles and 2-mercaptobenzoxazoles is described. 1,3-Propanedithiol was employed to convert benzothiazoles and benzoxazoles to the corresponding heteroarylthiols in the presence of potassium hydroxide and DMSO. This novel protocol is featured by direct C-H mercaptalization of heteroarenes and a simple reaction system.

Synthesis and Antimicrobial Activity of Bis(azolyl) Urea Derivatives

A variety of bis(azolyl) urea derivatives were prepared by the reaction of methyl benzazoyl carbamates with azolyl amines in the presence of mild base and studied their antimicrobial activity. The presence of electron donating substituents on the aromatic ring enhanced the activity. Methoxy substituted benzothiazolyl thiazolyl urea, benzothiazolyl imidazolyl urea and benzimidazolyl thiazolyl urea displayed prominent antibacterial activity against Bacillus subtilis. Benzothiazolyl imidazolyl urea and methyl substituted benzimidazolyl imidazolyl urea showed good antifungal activity against Aspergillus niger.

New Coumarin-benzoxazole derivatives: Synthesis, photophysical and NLO properties

In this work, a series of novel D-pi-A structure coumarin-benzoxazole derivatives have been synthesized. The third-order NLO properties of the coumarin derivatives in DMSO are investigated by Z-scan measurement, and the photophysical properties of the compounds are studied through experimental and theoretical calculation methods. The results showed that the introducing specific functional groups at multiple active sites of coumarin reduced the pi-pi intermolecular stacking, while the formation of push-pull structure decreased E-gap of entire molecule. The excellent third-order NLO responses based on chromophore 4c with maximum nonlinear absorptive coefficients is 50.64 x 10(-13) esu and the largest second-order NLO hyperpolarizability is 7.12 x 10(-30) esu. Experimental results have also been confirmed through DFT, TDDFT and ZINDO calculations. Our results indicate that the coumarin fluorescent dyes are promising candidate materials for NLO applications.

Benzoxazole Derivative K313 Induces Cell Cycle Arrest, Apoptosis and Autophagy Blockage and Suppresses mTOR/p70S6K Pathway in Nalm-6 and Daudi Cells

Benzoxazole derivative K313 has previously been reported to possess anti-inflammatory effects in lipopolysaccharide-induced RAW264.7 macrophages. To date, there have been no related reports on the anticancer effects of K313. In this study, we found that K313 reduced the viability of human B-cell leukemia (Nalm-6) and lymphoma (Daudi) cells in a dose-dependent manner without affecting healthy peripheral blood mononuclear cells (PBMCs) and induced moderate cell cycle arrest at the G0/G1 phase. Meanwhile, K313 mediated cell apoptosis, which was accompanied by the activation of caspase-9, caspase-3, and poly ADP-ribose polymerase (PARP). Furthermore, cells treated with K313 showed a significant decrease in mitochondrial membrane potential (MMP), which may have been caused by the caspase-8-mediated cleavage of Bid, as detected by Western blot analysis. We also found that K313 led to the downregulation of p-p70S6K protein, which plays an important role in cell survival and cell cycle progression. In addition, treatment of these cells with K313 blocked autophagic flux, as reflected in the accumulation of LC3-II and p62 protein levels in a dose- and time-dependent manner. In conclusion, K313 decreases cell viability without affecting normal healthy PBMCs, induces cell cycle arrest and apoptosis, reduces p-p70S6K protein levels, and mediates strong autophagy inhibition. Therefore, K313 and its derivatives could be developed as potential anticancer drugs or autophagy blockers in the future.

CuNiFe a Magnetic Nano-Catalyst: an Efficient Catalyst for the Selective Synthesis of Benzoxazoles

An efficient protocol for the synthesis of benzoxazoles from o-nitrophenols and benzyl alcohols using CuNiFe as a magnetic nano-catalyst via hydrogenation transfer has been described. The CuNiFe magnetic nano-catalyst was prepared by an operationally simple method, without the use of any toxic reagents. The prepared catalyst was characterized by XRD, FEG-SEM, EDAX, TEM, and FT-IR techniques. No additional additives such as oxidants and reductants were required for this reaction. CuNiFe magnetic nano-catalyst showed good air stability and could be recycled up to six times without any significant loss in its catalytic activity.
