Correlation between beta-amyloid deposits revealed by BF-227-PET imaging and brain atrophy detected by voxel-based morphometry-MR imaging: a pilot study

Objective The purpose of this study was to investigate whether beta-amyloid (A beta) deposition was associated with local atrophy of corresponding areas in the brain. Methods [11C]2-[2-(2-Dimethylaminothiazol-5-yl) ethenyl-6-[2-(fluoro)ethoxy]benzoxazole (BF-227)-PET, MRI and neuropsychological tests were carried out on 56 subjects, out of which 21 were patients with Alzheimer’s disease (AD), 20 were patients with mild cognitive impairment (MCI) and 15 were normal controls (NC). The BF-227 uptake in each local brain region was set up with automated anatomical labeling atlas using Wake Forest University PickAtlas software and local standardized uptake value ratios of BF-227 were calculated as the average value of right and left using the MRIcron software. Results Group comparisons of A beta deposition as determined by BF-227 uptake using PET imaging showed no significant differences between MCI and AD. A beta deposition was significantly higher in MCI and AD than in NC. The correlation analysis between local A beta deposition and gray matter atrophy showed that in AD, the A beta deposition in the inferior temporal gyrus was strongly related to the gray matter atrophy in this region. On the contrary, the A beta deposition in the precuneus was associated with the atrophy in the right occipital-temporal region. In the NC, the A beta deposition in the inferior temporal gyrus was associated with the atrophy in the precuneus. Conclusion In the AD, the relationship between the A beta deposition and local atrophy is area-dependent. In NC, A beta deposition in the inferior temporal gyrus correlated to the atrophy in the precuneus.

SONOCHEMICAL SYNTHESIS OF SOME NOVEL 6-IMINOBENZOXAZOLINONES WITH POTENTIAL ANTIBACTERIAL AND ANTIFUNGAL ACTIVITIES

An environmentally benign protocol for the synthesis of a novel series of imine derivatives containing 2-oxo-3H-benzoxazole scaffold was successfully developed. Thus benzoxazolinone-6-carbaldehyde (3) was carried out by the formylation of 3-methyl-2-oxo-3H-benzoxazole (1) using hexamethylenetetramine (HMTA) in polyphosphoric acid (PPA). The designed compounds were prepared by the treatment of compound (3) with primary amines in the presence of methanol as solvent, by using ultrasonic-assisted method under catalyst-free conditions and conventional heat in methanol at reflux in presence of catalytic amount of acetic acid, to afforded the pure desired 6-imino-2-oxo-3H-benzoxazoles (4a-4f) in appreciable yields; their purity was confirmed by melting point as well as thin layer chromatography (TLC). The chemical structures of the new synthesized compounds were elucidated on the basis of the FT-IR, H-1 and C-13-NMR spectroscopic techniques.

These dyes incorporate strongly electrodonating aromatic amino groups on the benzofuranyl side, enabling the appearance of a dual fluorescence emission corresponding to the radiative decay of the excited enol (E*) and keto (K*) tautomers at high and low energy respectively. It was previously demonstrated that dual E*/K* emission could originate from a beneficial decrease of phenolic acidity upon absorption of light leading to a thermodynamic stabilization of the first excited-state. The innovation within these dyes lies in the double functionalization of the aniline moiety with 2-(2-methoxyethoxy)ethyl units which allowed a better solubilization in protic solvents, as compared to their butyl analogs while keeping strong electrodonating capacity. Their intrinsic amphiphilic character leads to a good vectorization in a wide range of solvents from toluene to PBS buffer. The investigation of the photophysical properties of these dyes in solution showed a clear dual emission in apolar solvents with the E* band gradually red-shifting along with the dipole moment of the solvent. Dual emission is also observed in the solid-state when these dyes are doped as 1% wt in PMMA or PS films. Finally, the interactions of one dye with calf-thymus (ct)-DNA and Bovine Serum Albumin (BSA) have been explored and reveal pronounced modifications of the UV-Vis profile of the dye. Additionally, a gradual hypsofluorochromic shift and narrowing of the K* band along with the appearance of the E* fluorescence band upon addition of ct-DNA or BSA is also observed, presumably evidencing an intercalation mode of binding.

Azole-hydrazone derivatives: Design, synthesis, in vitro biological evaluation, dual EGFR/HER2 inhibitory activity, cell cycle analysis and molecular docking study as anticancer agents

In this research, three series of azole-hydrazone derivatives namely, benzimidazole, benzoxazole and benzothiazole were designed and synthesized. Their structures were confirmed by elemental analysis and spectroscopic techniques. Stereochemical configuration of the synthesized compounds (Z/E) was determined. The new derivatives were tested in vitro against both human breast adenocarcinoma (MCF-7) and human hepatic adenocarcinoma (HepG2) cell lines. The most active compounds 3h (IC50 = 0.067 mu M against MCF-7) and 3l (IC50 = 0.027 mu M against HepG2) were further tested for Epidermal Growth Factor Receptor (EGFR) inhibitory activity. The most active 3h on EGFR was then screened for HER2 and VEGFR enzymes. Caspase-3/9 protein level expression were measured for the two compounds 3h and 3l. Cell cycle analysis showed pre G1 apoptosis and cell cycle arrest at G2/M phase. Up-regulation of Bax and down-regulation of Bcl-2 protein expression level confirmed apoptosis. Oxidative coupling of benzaldehyde with o-aminophenol utilizing lead tetraacelate approach was used to realize the synthesis of compounds 1-11. The cytotoxicity of 1-11 was screened against breast cancer cell line MCF-7 and human colon cancer cell line HCT-116 utilizing doxorubicin as a reference drug. Among these compounds, 2-(3-benzyloxyphenyl)benzoxazole-5-acetic acid (5) and 2-(4-methoxyphenyl)benzoxazol-5-acetic acid (10), were found to be promising cytotoxic compounds against the MCF-7 cell line. In addition, this study showed that the presence of an acetic acid group at position 5 of the benzoxazole nucleus enhances the activity. Moreover, we noticed that the presence of an oxygen atom directly linked to the phenyl substituent improves activity as well. These results offer a new benzoxazole based template to design and develop novel antineoplastic agents.

Synthesis and application of benzoxazole derivative-based fluorescent probes for naked eye recognition

A fluorescent probe to detect biothiols (GSH/Hcy/Cys) was synthesized with benzothiazole as the fluorophore and nitromethane as the recognition group. The recognition mechanism of the fluorescent probe was deduced. It was found that a nitroalkene in the molecule was used as the recognition site for this probe and reacted with the mercapto group of the biothiols by electrophilic addition, significantly enhancing the fluorescent signal. Experimental results showed that the fluorescent probe had a low detection limit, good selectivity, strong anti-interference ability, and good naked eye recognition. The probe could detect biothiols (GSH/Hcy/Cys) in 20% organic solvent, with detection limits of 0.33 mu M, 0.70 mu M, and 0.87 mu M, respectively. The compounds show enantiotropic smectic/nematic phases with mesophase ranges are 71-97 degrees C and 87-136 degrees C on heating and cooling processes for nPPF(2)BP, respectively. They give low melting points due to lateral fluoro substituent and flexible terminal 2-propenyloxymethyl chain. It is found that the compounds nPPF(2)BP with shorter alkoxy chain (n = 3, 4) exhibit a wide range of nematic mesophase, which is ascribed to enhanced pi-pi interaction caused by terminal vinyl moiety, whereas further elongation of the terminal alkoxy chain results in supressing nematic phase and increasing smectic mesophase. Compared with methyl terminated analogues, 2-propenyloxymethyl terminated compounds nPPF(2)BP display much lower melting points and wider or comparable mesophase range both in heating and cooling. Densification-induced hollow fiber membranes using crosslinked thermally rearranged (XTR) polymer for CO2 capture

Since thermally rearranged (TR) polymers were known as high gas permeable and processable materials, fabricating high performance hollow fiber (HF) membranes have been tried using them. However, an unexpected drawback emerged which is the gas productivity loss by thermal densification of skin layers during thermal treatment above their glass transition temperature (T-g). In this work, we used a recently reported crosslinked-TR (XTR) polybenzoxazole to develop a new class of high-flux TR hollow fibers by directly exploiting the thermal densification phenomenon. The TR temperature range (320-460 degrees C) and T-g (394 degrees C) were determined by thermal gravimetric analysis (TGA) and dynamic mechanical analysis (DMA). The chain rigidity of the XTR polymer increased during an isotherm treatment at its T-g, suggesting a restricted densification. Surprisingly, the undesired pinhole-defects (pore diameter < 5 nm) on precursor fibers were perfectly healed after thermal treatment (> 400 degrees C), forming an ultrathin defect-free skin layer on thermally-densified XTR hollow fiber membranes. The pore-healed XTR hollow fibers exhibited an outstanding CO2 permeance of similar to 2300 GPU and a CO2/N-2 selectivity of 17.4 with a skin thickness of 103 nm.
