Synthesis and spectral properties of a novel fluorescent probe for Zn2+ based on a benzoxazole fluorophore

A novel turn-on fluorescent probe containing a benzoxazole fluorophore has been synthesised and its response to different metal ions has been investigated by spectrometry. The probe was highly selective and sensitive to Zn2+ and showed a complexation ratio towards Zn2+ of 1:1. It also exhibited a linear response range for Zn2+ from 0.3 to 4.8 mu mol L-1 with a detection limit of 0.36 mu mol L-1.

Tc-99m-labeled Small Molecules for Diagnosis of Alzheimer’s Disease: Past, Recent and Future Perspectives

Background: Alzheimer’s disease (AD) is an age-related progressive neurodegenerative disease. Its prominent hallmarks are extracellular deposition of beta-amyloids (amyloid plaques), intracellular neurofibrillary tangles (NTFs), neurodegeneration and finally loss of cognitive function. Hence, AD diagnosis in the early stage and monitoring of the disease are of great importance. Methods: In this review article, we have reviewed recent efforts for design, synthesis and evaluation of Tc-99m labeled small molecule for AD imaging purposes. Results: These small molecules include derivatives of Congo red, benzothiazole, benzofuran, benzoxazole, naphthalene, biphenyl, chalcone, flavone, aurone, stilbene, curcumin, dibenzylideneacetone, quinoxaline, etc. The different aspects of Tc-99m-labeled small molecules including chemical structure, their affinity toward amyloid plaques, BBB permeation and in vivo/vitro stability will be discussed. Conclusion: The fmdings of this review confirm the importance of Tc-99m-labeled small molecules for AD imaging. Future studies based on the pharmacophore of these designed compounds are needed for improvement of these molecules for clinical application.

Identification of novel 2-benzoxazolinone derivatives with specific inhibitory activity against the HIV-1 nucleocapsid protein

In this report, we present a new benzoxazole derivative endowed with inhibitory activity against the HIV-1 nucleocapsid protein (NC). NC is a 55-residue basic protein with nucleic acid chaperone properties, which has emerged as a novel and potential pharmacological target against HIV-1. In the pursuit of novel NC-inhibitor chemotypes, we performed virtual screening and in vitro biological evaluation of a large library of chemical entities. We found that compounds sharing a benzoxazolinone moiety displayed putative inhibitory properties, which we further investigated by considering a series of chemical analogues. This approach provided valuable information on the structure-activity relationships of these compounds and, in the process, demonstrated that their anti-NC activity could be finely tuned by the addition of specific substituents to the initial benzoxazolinone scaffold. Lipase-catalyzed kinetic resolution of novel antitubercular benzoxazole derivatives

Novel benzoxazole derivatives were synthesized, and their antitubercular activity against sensitive and drug-resistant Mycobacterium tuberculosis strains (M.tuberculosis H(37)Rv, M.tuberculosis sp. 210, M.tuberculosis sp. 192, Mycobacterium scrofulaceum, Mycobacterium intracellulare, Mycobacterium fortuitum, Mycobacterium avium, and Mycobacterium kansasii) was evaluated. The chemical step included preparation of ketones, alcohols, and esters bearing benzoxazole moiety. All racemic mixtures of alcohols and esters were separated in Novozyme SP 435-catalyzed transesterification and hydrolysis, respectively. The transesterification reactions were carried out in various organic solvents (tert-butyl methyl ether, toluene, diethyl ether, and diisopropyl ether), and depending on the solvent, the enantioselectivity of the reactions ranged from 4 to >100. The enzymatic hydrolysis of esters was performed in 2 phase tert-butyl methyl ether/phosphate buffer (pH=7.2) system and provided also enantiomerically enriched products (ee 88-99%). The antitubercular activity assay has shown that synthesized compounds exhibit an interesting antitubercular activity. Racemic mixtures of alcohols, (+/-)-4-(1,3-benzoxazol-2-ylsulfanyl)butan-2-ol ((+/-)-3a), (+/-)-4-[(5-bromo-1,3-benzoxazol-2-yl)sulfanyl]butan-2-ol ((+/-)-3b), and (+/-)-4-[(5,7-dibromo-1,3-benzoxazol-2-yl)sulfanyl]butan-2-ol ((+/-)-3c), displayed as high activity against M.scrofulaceum, M.intracellulare, M.fortuitum, and M.kansasii as commercially available antituberculosis drug-Isoniazid. Moreover, these compounds exhibited twice higher activity toward M.avium (MIC 12.5) compared with Isoniazid (MIC 50).

Azole-hydrazone derivatives: Design, synthesis, in vitro biological evaluation, dual EGFR/HER2 inhibitory activity, cell cycle analysis and molecular docking study as anticancer agents

In this research, three series of azole-hydrazone derivatives namely, benzimidazole, benzoxazole and benzothiazole were designed and synthesized. Their structures were confirmed by elemental analysis and spectroscopic techniques. Stereochemical configuration of the synthesized compounds (Z/E) was determined. The new derivatives were tested in vitro against both human breast adenocarcinoma (MCF-7) and human hepatic adenocarcinoma (HepG2) cell lines. The most active compounds 3h (IC50 = 0.067 mu M against MCF-7) and 3l (IC50 = 0.027 mu M against HepG2) were further tested for Epidermal Growth Factor Receptor (EGFR) inhibitory activity. The most active 3h on EGFR was then screened for HER2 and VEGFR enzymes. Caspase-3/9 protein level expression were measured for the two compounds 3h and 3l. Cell cycle analysis showed pre G1 apoptosis and cell cycle arrest at G2/M phase. Up-regulation of Bax and down-regulation of Bcl-2 protein expression level confirmed apoptosis. Preparation and gas separation performance of thermally rearranged poly (benzoxazole-co-amide) (TR-PBOA) hollow fiber membranes deriving from polyamides

Flexible poly(benzoxazole-co-amide) (PBOA) gas separation hollow fiber membranes were successfully prepared deriving from ether-containing polyamide precursors by thermal densification with or without a supplementary coating treatment. Compared with polyimide precursors, thermally rearrangement in this work required lower treatment temperature but generated better mechanical properties. Both spinning process and thermal treatment conditions were systemically manipulated to intensify the membrane densification aiming at achieving greatly improved selectivity of the obtained TR-PBOA hollow fiber membranes. It was found that draft ratio had little contribution to selectivity enhancement. Separation performance was substantially improved when precursor fibers were dried using ethanol-hexane exchange method and then thermally treated at 350-390 degrees C TR hollow fibers obtained by combined thermal treatment with coating method exhibited the highest selectivity values in this study.

Novel high-performance poly(benzoxazole-co-imide) resins with low dielectric constants and superior thermal stabilities derived from thermal rearrangement of ortho-hydroxy polyimide oligomers

Unsatisfied dielectric property and insufficient thermal stability are major obstacles for the commercialization of polyimide resins in the future ultralarge scale integration (ULSI) or radar-wave-transparent composite applications. The incorporation of ortho-hydroxy diamine into a common phenylethynyl terminated oligoimides, aiming to subsequently form additional rigid benzoxazole units by the thermal rearrangement (TR), were prepared. The effects of TR-able codiamine on the processing ability of oligoimides, molecular packing and properties, including thermal stability, dielectric property and bonding ability to reinforcing fibers, for the resulting poly(imide-co-benzoxazole) (PI-co-PBOs) resins have been examined in detail. These thermally rearranged PI-co-PBO resins exhibit reduced dielectric constants of 2.56-3.4 at f = 0.1 GHz, lower than the cured PI with a dielectric constant of 3.55. Meanwhile, the 5 wt% weight loss temperature (T-d5) of the resultant resins increases from 455 degrees C for the cured PI to 491 degrees C for the PI-co-PBO-60 containing 60 mol% benzoxazole unit, and correspondingly, the tensile modulus of PI-co-PBO-60 reaches 2.47 GPa, which is 76% higher than that of the cured PI. These resultant PI-co-PBOs exhibit the combined excellent properties, indicating them a great potentials as the new low-dielectric constant polymer resins in microelectronic industries or advanced composites.

Low Band Gap Benzoxazole-Linked Covalent Organic Frameworks for Photo-Enhanced Targeted Uranium Recovery

The inherent features of covalent organic frameworks (COFs) make them highly attractive for uranium recovery applications. A key aspect yet to be explored is how to improve the selectivity and efficiency of COFs for recovering uranium from seawater. To achieve this goal, a series of robust and hydrophilic benzoxazole-based COFs is developed (denoted as Tp-DBD, Bd-DBD, and Hb-DBD) as efficient adsorbents for photo-enhanced targeted uranium recovery. Benefiting from the hydroxyl groups and the formation of benzoxazole rings, the hydrophilic Tp-DBD shows outstanding stability and chemical reduction properties. Meanwhile, the synergistic effect of the hydroxyl groups and the benzoxazole rings in the pi-conjugated frameworks significantly decrease the optical band gap, and improve the affinity and capacity to uranium recovery. In seawater, the adsorption capacity of uranium is 19.2x that of vanadium, a main interfering metal in uranium extraction.

Excited-state intramolecular proton transfer of 6-amino-2-(2 ‘-hydroxyphenyl) benzoxazole (6A-HBO) in different solvents

The excited-state intramolecular proton transfer (ESIPT) process of 6-amino-2-(2 ‘-hydroxyphenyl) benzoxazole (6A-HBO) was investigated using density functional theory and time-dependent density functional theory methods with B3LYP and TZVP basis sets. n-Heptane, dichloromethane, methanol, and acetonitrile were chosen as a series of polar solvents in calculations using the IEFPCM model. To obtain a more comprehensive ESIPT mechanism, we constructed the S-0 and S-1 states’ potential energy surfaces (PESs) by incrementally twisting the OH bond and increasing the distance of O-H bond. Based on the analysis of the bond lengths, the IR vibrational spectra, and the frontier molecular orbits (MOs), the intramolecular hydrogen bonding (OH…N) is clearly strengthened, and the charge is redistributed in the S-1 state. The results of calculated absorption spectrum are in accord with the experimental data. The fluorescence spectrum of 6A-HBO-enol showed a normal red shift, but the red shift of the 6A-HBO-keto is larger and increases with the solvent polarity, indicating a charge transfer. Analysis of the PESs indicates a lower potential energy barrier in S-1 state for the proton transfer from the O atom to the N atom, with the excited state potential barrier slightly decreasing with the increase of the solvent polarity.

Copper-Catalyzed Regio- and Enantioselective Addition of Silicon Grignard Reagents to Alkenes Activated by Azaaryl Groups

A new application of silicon Grignard reagents in C(sp(3))-Si bond formation is reported.
