General procedure: To a solution of 1 mmol of thiol in acetonitrile (2mL), was added a solution of 8 mL of phosphate buffer including laccase (40 U) and 4-phenyl urazole (10 mol%). The resulting mixture was stirred under air at room temperature for the time specified. After completion of the reaction (monitored by TLC), NaOH (10%, 10 mL). The combined organic phases were dried over anhydrous Na2SO4, filtered and evaporated under reduced pressure. The crude products were purified by recrystallization or chromatography on silica gel (n-hexane/EtOAc, 5/1). All the products were known and characterized by comparison of their spectral data (1H NMR) and physical properties (melting point) with those of authentic samples.

The mixture was warmed at 80C for 4 hours then evaporated and the residue was dissolved in 5 ml of DMF. The solution of the isocyanate was added dropwise to a solution in DMF (10 ml) of compound 1b (384 mg, 2.56 mmol) and the mixture was warmed at 80C for 8 hours. (TLC AcOEt). The solvent was evaporated and the crude was dissolved in AcOEt (30 ml) and washed with water (1 X 20 ml) and brine. The organic phase was dried over sodium sulfate and concentrated under vacuum. The purification of the crude residue by chromatographic column gave 200 mg of a pale rose solid. Yield = 18% 1HNMR (DMSO, 400 MHz) delta 2.91 (4H, m), 3.76 (4H, m), 4.46 (2H, d, J = 5.6 Hz), 6.97 (3H, m), 7.05 (1H, m), 7.36 (1H, s), 7.46 (1H, d), 7.54 (1H, d), 8.53 (1H, s), 11.00 (1H, bs); [M+1] 437.1 (C20H19F3N4O4 requires 436.4).).

Commercially available 2-chloro-4-trifluoromethylbenzylamine (572 mg, 2.7 mmol) was dissolved in 20ml of AcOEt and at 0C triphosgene (809 mg, 2.7 mmol) was added to the solution. The mixture was warmed at 80C for 4 hours then evaporated and dissolved in 5 ml of DMF. The solution of the isocyanate was added dropwise to a solution in DMF (5 ml) of compound lb (270 mg, 1.8 mmol) and the mixture was warmed at 80C for 8 hours. (TLC AcOEt). The solvent was evaporated and the crude was dissolved in AcOEt (30 ml) and washed with water (1 X 20 ml) and brine. The organic phase was dried over sodium sulfate and concentrated under vacuum. The purification of the crude residue by chromatographic column gave 70 mg of a white solid. Yield = 10% ‘HNMR (DMSO, 400 MHz) delta 4.45 (2H, d, J = 6 Hz), 6.97 (2H, d, J = 4.4 Hz), 7.07 (2H, m), 7.63 (1H, d, J = 8 Hz), 7.74 (2H, d), 7.86 (1H, s), 8.61 (1H, bs), 10.90 (1H, bs); [M+1] 386.6 (C16H11CIF3N3O3 requires 385.7).

After completion of the reaction, the catalyst wasseparated by simple filtration and the mixture was washedwith CH2Cl2 (5 9 2 mL), and next, the product wasextracted with CH2Cl2 (5 mL 9 4). The organic layer wasdried over anhydrous Na2SO4 (1.5 g). After the evaporationof dicoloromethan gave the pure products in 85-98 %yields.

General procedure: Commercially available 5-chloro benzoxazoles or benzothiazoles(5 mmol) and K2CO3 (15 mmol) were added to 50 mL of DMF.The mixture was heated at 60 C and 1-(2-chloroethyl)hexahydro-1H-azepine hydrochloride (6.5 mmol) was added. The mixture was maintained at 60 C under stirring for 3 h, then was cooled to room temperature, poured into water, and extracted with diethyl ether(3 50 mL). The organic layer was washed with brine, dried overan hydrous Na2SO4 and evaporated under reduced pressure to give are sidue, which was purified by flash column chromatography oversilica gel using ethyl acetate/methanol 8:2 v/v as eluent. Immediately after solvent evaporation, free bases were converted into hydrochlorides by dissolving the corresponding amine in diethyl ether and treating with a 1.25MHCl ethanol solution (pH 1e2).
