A solution of methyl benzo[d]oxazole-7-carboxylate (300 mg, 1.7 mmol) in THF (5 mL) was cooled down to 0C and methyl magnesium bromide 3.2M in 2-Me-THF (3.0 equiv., 5.1 mmol) was added dropwise. The mixture was allowed to warm-up to room temperature and quenched with saturated solution of NH4Cl. The resulting mixture was extracted 3 times with EtOAc and the organic solutions were washed with brine, dried with Na2SO4 and concentrated to dryness. The crude was purified by column chromatography on silica gel using DCM:EtOAc (95:5) as mobile phase to give 2-(benzo[d]oxazol-7-yl)propan-2-ol (110 mg, 37%). UPLC-MS (Acidic Method, 2 min): rt 0.78 min, m/z 178.0 [M+H]+

A solution of succinic anhydride (141.1 g, 1.41 mol) and 70 g (about 0.47 mol) of compound of step 1a (70 g, 0.47 mol) in 2500 ml of anhydrous toluene was refluxed for overnight. After that, HATU (10 g, 0.26 mol) and 4-methylmorpholine (41.36 ml, 0.376 mol) were added and the resulting mixture was refluxed for 2-3 hours. TLC showed that the major spot was product (Rf=0.35, acetone_hexane=1:2). After reaction was completed, the solvent was evaporated and the residue was dissolved in about 2000 ml of CH2Cl2. The solution was washed with aqueous NaHCO3. After pH was adjusted to 7-8 and washed with brine, organic phase was separated and dried over MgSO4. Filtration and removal of solvent gave the title compound (98 g) as fine white needle crystalline ESI MS m/e: 231 (M+H)+.

After3 h, TLC analysis showed complete conversion of starting materials. The reaction mixture was cooled to room temperature, charged water (30 mL) and extracted with EtOAc (2 × 20 mL). The combined organic layer was washed with water (20 mL) followed by brine and concentrated under reduced pressure to obtain gummy liquid. This crude material was triturated with MTBE to afford desired product as solid and which used as such in next step without any further purification.

The reaction mixture was stirred for 10 mm and CS2 (6.32mL, 93.2mmol) was added dropwise at 0 00 and the reaction mixture was further stirred for 10 mm at 0 00 An additional portion of 20.0 M NaOH (1.86mL, 37.3mmol) was added at 0 00 and reaction mixture was again stirred for 10 mm at 0 00 Finally, CH3I (5.84mL, 93.2mmol) was added dropwise at 0 00. The reaction mixture was stirred at rt for 30 mm. The TLC showed reaction to be complete. The mixture was poured into ice-water (lOOmL) and the precipitated solid was filtered , washed with water(5OmL) followed by hexane (3OmL) and dried under reduced pressure to obtain dimethyl benzo[d]oxazol-2- ylcarbonimidodithioate as an off white solid . Yield: 6.92g (78percent). MS (ESl+) for CHNOS m/z 239.03 [M+H] 1H NMR (400 MHz, DMSO-d6): 7.64 (bs, 2H), 7.32 (bs, 2H), 2.67 (bs, 6H).

To a mixture of 3H-benzooxazol-2-one (20 g, 0.15 mol) in DCM (500 mL) was added bromine (8.34 mL, 0.16 mol). After stirring at room temperature for 19.5 h, the orange precipitate that had formed was filtered off and washed with DCM until the orange color was washed out. The filtrate was concentrated to approximately 33% of its original volume and filtered and washed as before. The combined solids weighed 28.36 g and contained ca. 8-9% starting material.

The product from step 2 (0.108 g, 0.6 mmol) and 2- hydroxyacetophenone (0.09 g, 0.66 mmol) were added to a round bottomed flask under nitrogen. Anhydrous pyridine (5 mL was added to it with stirring. The reaction mixture was cooled down using an ice bath. Phosphorus oxychloride (0.15 g, 0.99 mmol) was added and the reaction mixture was stirred at room temperature overnight. Pyridine was removed under vacuum and the residue was acidified with 1 N HCI (pH 4~5) and extracted with ethyl acetate. The solvent was EPO evaporated in vacuo to leave a residue which was purified by silica gel column chromatography (15 g). The eluent was a mixture of ethyl acetate and hexane in 1 :1. Fraction 1 was evaporated affording pure compound (0.0192 g, 11%).
