Benzimidazoles and perimidines are subsidiary structures for research and development of new biologically active molecules and have established prominence because of their promising biological activities. A novel coordination mode of kappa(1)-N-Br-pyridylbenz-(imida, oxa or othia)-zole to Pt(II): synthesis, characterization, electrochemical and structural analysis

Cyclic voltammetry studies show an irreversible two-electron process at -0.50 and -0.51 V, which was ascribed to the Pt(II)/Pt(IV) couple, for complexes 2 and 3. Antiinflammatory and Antiproliferative Activity of Naphthoxazole, Fused Hetero-benzoxazole and Bridged Benzobicyclic Photoproducts

Several naphthoxazoles and fused hetero-benzoxazoles inhibited TNF alpha protein expression in LPS stimulated PBMC, indicating possible antiinflammatory role which would be interesting to further investigate. Physico-chemical properties of tested compounds have been also studied using chromatographic lipophilicity measure, chrom logD and logP was calculated as the importance of physico-chemical properties of compounds at early stage of discovery of new drugs is well established. The similarities in structure and activity of some representative compounds affirm the need to further address their antiinflammatory properties.

The identification and pharmacological evaluation of potent, selective and orally available ACC1 inhibitor

In our effort to explore the potential of ACC1-selective inhibitor as in vivo probe molecule, a series of 1,3-benzoxazole derivatives was synthesized. Previously, we reported a series of novel bicyclic and monocyclic ACC1-selective inhibitors. Among them, compound 1a exhibited highly potent cellular activity (acetate uptake IC50 = 0.76 nM) as well as promising in vivo PD efficacy. However, compound 1a caused severe body weight reduction in repeated dose administration in the mouse model. Since 1a showed potent inhibitory activity against mouse ACC1 as well as strong inhibition of mouse ACC2, we further examined a series of 1a analogues in order to reduce undesirable body weight change. The replacement of acetamide moiety with ureido moiety dramatically improved selectivity of mouse ACC1 against ACC2. In addition, analogue 1b displayed favorable bioavailability in mouse cassette dosing PK study, hence in vivo PD studies were also carried out. Oral administration of 1b significantly reduced the concentration of malonyl-CoA in HCT-116 xenograft tumors at doses of more than 30 mg/kg. Furthermore, compound 1b showed significant antitumor efficacy in 786-O xenograft mice at an oral dose of 30 mg/kg (T/C = 0.5%). Accordingly, our novel potent ACC1-selective inhibitor represents a set of useful orally-available research tools, as well as potential therapeutic agents particularly in terms of new cancer therapies.

Effects of non-TR-able codiamines and rearrangement conditions on the chain packing and gas separation performance of thermally rearranged poly (benzoxazole-co-imide) membranes

In present work, the incorporation of nonrearrangable codiamines (mPDA, DMB or TFMB) with different substituted groups into an ortho-hydroxypolyimide (HPI) precursor was applied to modify the gas separation and mechanical performances of resultant thermally rearranged (TR) polybenzoxazole membranes. Evolution of the thermal rearrangement for ortho-hydroxy copolyimide precursors was effectively monitored by the TGA-FTIR, WAXD and PALS measurements. The incorporated non-TR-able codiamines and the thermal treatment protocols have a great effect on the chains packing behavior, free volume, mechanical properties as well as the gas separation behavior of resulted TR-PBOI membranes. These TR-PBOI membranes show high tensile strength of 117-160 MPa and good elongation at break of 7.5-9.0% as thermally treated at 400 degrees C for 2 h. The t-TR400-2 membrane comprising the TFMB codiamine exhibits a synergistic effect of high gas permeability and high gas pairs selectivity, which is mainly attributed to the loose chains packing resulted from the thermal rearrangement and the substituted bulky -CF3 group that resulting a high fractional free volume (FFV approximate to 0.15). The systematic structure/property relationship studies serve as a guide of materials and process development of commercial TR membranes for gas separation applications.

Bimolecular fluorescence quenching of benzoxazole/benzothiazole-based functional dyes

Four dyes that belong to the group of hemicyanines and possess benzoxazole and benzothiazole moieties are synthesised and described. The spectroscopic properties of these compounds in 1-methyl-2-pyrrolidinone (MP) were studied. Synthesised dyes absorb in the UV-Vis region. The emission spectra are broad with a maximum located at approximately 500-520 nm. The deactivation of excited states of the synthesised molecules by various quenchers is also presented. The fluorescence quenching parameters were calculated using the Stern-Volmer equation. It was found, that the fluorescence quenching of the excited dye by quencher molecules occurs. Utilization of hydrophobic ligands for water-insoluble Fe(II) water oxidation catalysts – Immobilization and characterization

Herein, we compare the electrochemical and electrocatalytic properties of two selected, water-insoluble Fe(II) coordination complexes made with the non-symmetric, bidentate ligands, 2-(2′-pyridyl)benzimida zole (PBI) in [Fe(PBI)(3)](OTf)(2) (1, OTf- = trifluoromethyl sulfonate anion) and 2-(2′-pyridyl)benzoxazole (PBO) in [Fe(PBO)(2)(OTf)(2)] (2). Cyclic voltammetry in water/acetonitrile mixture indicates considerable activity for both compounds. However, only 1 acts as homogeneous catalyst. The complexes have been successfully immobilized on indium-tin-oxide (ITO) electrode surface. The hydrophobic ligands allowed for a simple dip-coating and drop-casting of 1 and 2 onto ITO. Both 1/ITO and 2/ITO showed increased activity in electrocatalytic O-2 evolution in borate buffer at pH 8.3. According to scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), X-ray photoelectron spectroscopy (XPS), moreover, re-dissolution tests, the Fe remains in complex with PBI during electrolysis in the drop-casted, nano-porous films of 1/ITO.
