Identification of novel 2-benzoxazolinone derivatives with specific inhibitory activity against the HIV-1 nucleocapsid protein

In this report, we present a new benzoxazole derivative endowed with inhibitory activity against the HIV-1 nucleocapsid protein (NC). NC is a 55-residue basic protein with nucleic acid chaperone properties, which has emerged as a novel and potential pharmacological target against HIV-1. In the pursuit of novel NC-inhibitor chemotypes, we performed virtual screening and in vitro biological evaluation of a large library of chemical entities. We found that compounds sharing a benzoxazolinone moiety displayed putative inhibitory properties, which we further investigated by considering a series of chemical analogues. This approach provided valuable information on the structure-activity relationships of these compounds and, in the process, demonstrated that their anti-NC activity could be finely tuned by the addition of specific substituents to the initial benzoxazolinone scaffold. Tc-99m-labeled Small Molecules for Diagnosis of Alzheimer’s Disease: Past, Recent and Future Perspectives

Background: Alzheimer’s disease (AD) is an age-related progressive neurodegenerative disease. Its prominent hallmarks are extracellular deposition of beta-amyloids (amyloid plaques), intracellular neurofibrillary tangles (NTFs), neurodegeneration and finally loss of cognitive function. Hence, AD diagnosis in the early stage and monitoring of the disease are of great importance. Methods: In this review article, we have reviewed recent efforts for design, synthesis and evaluation of Tc-99m labeled small molecule for AD imaging purposes. Results: These small molecules include derivatives of Congo red, benzothiazole, benzofuran, benzoxazole, naphthalene, biphenyl, chalcone, flavone, aurone, stilbene, curcumin, dibenzylideneacetone, quinoxaline, etc. The different aspects of Tc-99m-labeled small molecules including chemical structure, their affinity toward amyloid plaques, BBB permeation and in vivo/vitro stability will be discussed. Conclusion: The fmdings of this review confirm the importance of Tc-99m-labeled small molecules for AD imaging. Future studies based on the pharmacophore of these designed compounds are needed for improvement of these molecules for clinical application.

NH3(CH2)(5)NH3BiCl5: an efficient and recyclable catalyst for the synthesis of benzoxazole, benzimidazole and benzothiazole heterocycles

In this work, the condensation of aromatic aldehydes with different two-substituted aniline catalyzed by NH3(CH2)(5)NH3BiCl5 as heterogeneous and recyclable catalyst was reported. It was demonstrated that NH3(CH2)(5)NH3BiCl5 can act as an efficient and active catalyst and is reusable for six runs without a significant loss of their catalytic activity. Simple preparation of the catalyst, high catalytic activity and good reusability are noteworthy advantages of this catalytic system in the synthesis of benzoxazole, benzimidazole and benzothiazole heterocycles at room temperature under solvent-free conditions.

An energetic study of 5-fluoro-2-methylbenzoxazole (FMBO) and of 5-fluoro-2-methylbenzothiazole (FMBT), in condensed and gaseous states, has been performed using calorimetric techniques and computational calculations. The standard (p degrees = 0.1 MPa) molar enthalpies of formation of FMBO and FMBT, in the liquid phase, at T = 298.15 K, were derived from the corresponding standard molar energies of combustion, measured by rotating-bomb combustion calorimetry. At T = 298.15 K, the standard (p degrees = 0.1 MPa) molar enthalpy of vaporization, for each compound, was determined, by a direct method, using the vacuum drop microcalorimetric technique. For each compound, from this last value and from the enthalpy of formation of the liquid compounds, the corresponding standard (p degrees = 0.1 MPa) enthalpy of formation in the gaseous phase has been calculated. Additionally, the gas-phase standard molar enthalpies of formation of these two compounds were estimated computationally at the G3(MP2)//B3LYP level of theory, as well as their gas-phase basicities and proton affinities. Chromogenic properties of 2-(2-carbomethoxy-3,4-dichloro-6-hydroxyphenyl)benzoxazole and its Zn (II) and Cd(II) complexes

The chromogenic properties of 2-(2-carbomethoxy-3,4-dichloro-6-hydroxyphenyl)benzoxazole (1) were studied. Solvatochromism of 1 was observed at keto-enol equilibrium due to the ground state intramolecular proton transfer (GSIPT). It was found that solvents with a good hydrogen bond acceptor (beta) ability were able to stabilise the keto form of 1. The excited-state intra molecular proton transfer (ESIPT) in molecules of 1 resulted in intense fluorescence with an anomalous Stokes shift of up to 8493 cm 1 and quantum yields of 0.08-0.19. The keto-form of 1 exhibits negative T-type photochromism with a thermally reversible photoconversion to the enol form. The complexes of ligand 1 with Zn(II) and Cd(II) were synthesised. According to X-ray structural analysis, the zinc complex is the neutral complex, which is formed by two oxazole-N and two phenol O donors of ligand 1 which crystallise in a centrosymmetric group with the zinc ion located on the inversion centres. The synthesised complexes possess effective blue fluorescence and resistance to photodegradation.

A new cuprous coordinating polymer constructed by bridging 2,2-(1,4-butanediyl)bis-1,3-benzoxazole ligand: synthesis, structure, and properties

A new cuprous coordination polymer (CP) with 2,2-(1,4-butanediyl)bis-1,3-benzoxazole (BBO) and triphenylphosphine sulfide (SPPh3) ligands, {[Cu(BBO)(SPPh3)]ClO4}(n) (1), has been characterized by elemental analysis, UV-Vis, infrared spectra and single-crystal X-ray diffraction. The structural analysis revealed that 1 exhibits a one-dimensional CP and is in a three-coordinate slightly distorted trigonal planar environment in which the total angle around the copper(I) atom is 359.2 degrees. Solid luminescence properties investigation showed that 1 had one emission peak at 428 nm (lambda(ex) = 355 nm, blue luminescence), which may be attributed to MLCT [(d(10)(Cu)->pi*]. The cyclic voltammogram of 1 represents irreversible Cu2+/Cu+ pairs. The antioxidant activity experiment shows that 1 has the potential ability to scavenge hydroxyl and superoxide radical in vitro studies. [GRAPHICS] .

Modification of poly(benzimidazole-amide) nanocomposites by the incorporation of amine-functionalized ZnO nanoparticles: Thermal and morphological characterization

Fabrication, characterization, and properties of novel poly(benzimidazole-amide)/functionalized ZnO nanocomposites (PBIA/APS-ZnO NCs) were investigated. At first, an aromatic PBA containing 3 imidazole units per repeat unit was synthesized by direct polycondensation of 1,3-bis(5-carboxylic acid-2-benzimidazole)benzene (BCAB) with 5-(2-benzimidazole)-1,3-phenylenediamine (DAMI) with good yield as a polymeric matrix. The periphery of zinc oxide nanoparticles (ZnO NPs) was modified with 3-aminopropyltriethoxysilane (APS) to have a better dispersion NPs and enhancing interactions between nanoparticles and PBIA matrix. Different percentages of functionalized NPs (0, 4, 8, and 12wt.%) were then embedded in PBA matrix through ultrasonic irradiation technique. Fourier transform infrared and thermo-gravimetric analysis (TGA) confirmed that APS was successfully attached on the ZnO NP surface. The obtained NCs were characterized by means of Fourier transform infrared, X-ray diffraction, scanning electron microscopy, and TGA. The TGA of the PBIA/APS-ZnO NCs showed the enhancement in the thermal stability in comparison with the neat PBIA and that this increase is higher when the NP content increases. Scanning electron microscopy analyses of NCs revealed that the dispersion of APS-ZnO NPs was uniformly done in the PBIA matrix.

Two Cu(I) complexes constructed by different N-heterocyclic benzoxazole ligands: Syntheses, structures and fluorescent properties

Reaction of 2,2-(1,4-butanediyl)bis-1,3-benzoxazole (BBO) or 2-(2′-pyridyl)benzoxazole (PBO) ligands with [Cu(CH3CN)(2)(PPh3)(2)](BE4) afforded a copper(I) coordination polymer{[Cu(BBO)(PPh3)](BE4)}(n) (1) and a mononuclear copper(I) complex [Cu(PBO)(PPh3)(2)](BF4)center dot 2CH(2)Cl(2) (2) (where PPh3= triphenyl-phosphine), which have been characterized by elemental analysis, IR spectra and single-crystal X-ray diffraction. The structural analysis revealed that in complex 1, copper(I) ions are three-coordinated and the geometric structure around the central copper(I) atom can be described as planar trigonal configuration. Complex 1 exhibits an one-dimensional coordination polymer by two BBO bridging adjacent copper(I) ions, forming a single-stranded meso-helical chain structure. Mononuclear complex 2 is four-coordinated and slightly distorted triangular-pyramid geometry. Solid luminescence properties investigation show that complex 1 has two emission peaks, which attributed to the pi*-pi and pi*-n transitions. However complex 2 only has one emission peak, which may be attributed to MLCT [d(10)(Cu) -> pi*]. This indicates that different types of N-heterocyclic ligands have an important influence on the structure and luminescent properties of copper(I) complexes. (C) 2019 Elsevier B.V. A Magnetic Heterogeneous Biocatalyst Composed of Immobilized Laccase and 2,2,6,6-Tetramethylpiperidine-1-oxyl (TEMPO) for Green One-Pot Cascade Synthesis of 2-Substituted Benzimidazole and Benzoxazole Derivatives under Mild Reaction Conditions

The design of reusable high-performance heterogeneous catalysts via the immobilization of chemical and biochemical species on magnetic nanoparticles increases the efficiency of catalytic systems by facilitating easy, fast, and clean separation processes. Laccase and 4-amino-2,2,6,6-tetramethylpiperidine-1-oxyl were separately immobilized on amine functionalized iron (II, III) oxide nanoparticles with covalent bonding using glutaraldehyde as a coupling reagent. The prepared catalyst was used to synthesize 12 benzoxazole and benzimidazole derivatives. The one-pot, two-step enzymatic aerobic oxidation reaction included the condensation of insitu-produced salicylaldehyde derivatives with aromatic amines, followed by an enzymatic dehydrogenation process. Optimal reaction conditions consisted of a citrate buffer (10mM, pH4.5) at 40 degrees C for an incubation time of 10 h and a heterogeneous catalyst containing immobilized laccase (80 mg, 100 U) and immobilized 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) (40 mg, 2 mol%). The catalyst retained more than 85% of its initial activity after 10 runs. In addition to the potential for reuse without significant losses in performance, eco-friendly attributes of this catalytic system include its high catalytic activity and the ease with which it can be recovered from the reaction mixture using an external magnet.
