A strategy to design, develop, and evolve the recyclable Mg-Al-hydrotalcite-OtBu (HT-OtBu) by the interaction of KOtBu with the calcined hydrotalcite that transforms a well-defined stoichiometric WE reaction into a catalytic one in an effort to minimize the quantum of effluents released and reduce the cost of the process is described here. HT-OtBu is found to be an efficient solid base for WE reactions for the simple synthesis of alpha,beta-unsaturated esters and nitriles for the first time. The HT-OtBu, composed of various ratios of Mg/Al (i.e., 2, 2.5, and 3) and different contents of tBuO – (Mg/Al:3), and their precursors were prepared and well characterized to draw a correlation between structure and reactivity of the hydrotalcites in WE reactions.

The solubilities of bis(benzoxazolyl-2-methyl) sulfide in ethanol, 1-butanol, 2-propanol, chlorobenzene, toluene, o-xylene, and ethanol + water were measured by a synthetic method. The laser monitoring observation technique was used to determine the disappearance of the solid phase in the solid + liquid mixture. All data were measured in the temperature range from (273.25 to 325.25) K at atmosphere pressure. The effect of solvent composition and temperature on the solubility was discussed. For all solvents studied, the data are well fitted with a semiempirical Apelblat equation.

Selected pharmaceutical excipient prevent isoniazid and rifampicin induced hepatotoxicity

Background &Aims: The incidence of isoniazid (INH)-and rifampicin (RIF)-induced abnormal liver enzyme activity is 27% but only 19% with INH alone. Cytochrome P450 2E1 (CYP2E1) is thought to contribute to the synergistic effects of RIF and INH. Pharmaceutical excipients are inactive ingredients that are added to a pharmaceutical compound. The purpose of this study was to screen excipients for CYP2E1 inhibition and identify whether the screened excipients prevented INH/RIF-induced hepatotoxicity. Methods: Fifty-five known pharmaceutical excipients were screened for CYP2E1 inhibition. The hepatotoxic doses of INH and RIF were 50 and 100 mg/kg/day, respectively. Hepatotoxicity was assessed by the galactose single point (GSP) method (a US Food and Drug Administration (FDA) recommended quantitative liver function test), liver histopathology, malondialdehyde (MDA) assay, and measurement of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activity. We chose the CYP2E1-specific substrate chlorzoxazone to assess CYP2E1 activity in animal and human. Results: Mannitol inhibited CYP2E1 activity by 54% in mice with INH/RIF-induced hepatotoxicity (p < 0.005). Serum AST, ALT and GSP levels were significantly increased 3.8-to 7.8-fold in these mice (p < 0.005), and these levels could be lowered by mannitol. Mannitol significantly alleviated the depletion of hepatic glutathione (GSH) and partially reversed the increase in MDA formation in mice treated with INH/RIF (p < 0.005). Mannitol also decreased CYP2E1 activity by 58% in humans (p < 0.005). Furthermore, an antituberculosis (TB) efficacy assay revealed that mannitol did not affect the anti-TB effects of INH/RIF. Conclusions: Mannitol, an FDA-approved excipient, was found to be a CYP2E1 inhibitor. Mannitol may be a useful adjuvant for drugs that induce hepatotoxicity through CYP2E1, such as INH and RIF. Studying the Inhibitory Effect of Quercetin and Thymoquinone on Human Cytochrome P450 Enzyme Activities

Background: Quercetin (QR) and thymoquinone (TQ) are herbal remedies that are currently extensively used by the general population to prevent and treat various chronic conditions. Therefore, investigating the potential of pharmacokinetic interactions caused by the concomitant use of these herbal remedies and conventional medicine is warranted to ensure patient safety. Purpose of the Study: This study was conducted to determine the inhibitory effect of QR and TQ, two commonly used remedies, on the activities of selected cytochrome P450 (CYP) enzymes that play an important role in drug metabolism and/or toxicology. Materials and Methods: The in vitro studies were conducted using fluorescence-based high throughput assays using human c-DNA baculovirus expressed CYP enzymes. For measuring CYP2E1 activity, a validated High-performance liquid chromatography (HPLC) assay was utilized to measure the formation of 6-hydroxychlorzoxazone. Results: The obtained half-maximum inhibitory concentration values with known positive control inhibitors of this study were comparable to the published values indicating accurate experimental techniques. Although QR did not show any significant effect on CYP1A2 and CYP2E1, it exhibited a strong inhibitory effect against CYP2D6 and a moderate effect against CYP2C19 and CYP3A4. On the other hand, TQ demonstrated a strong and a moderate inhibitory effect against CYP3A4 and CYP2C19, respectively. Conclusions: The findings of this study may indicate that consumption of QR or TQ, in the form of food or dietary supplements, with drugs that are metabolized by CYP2C19, CYP2D6, or CYP3A4 may cause significant herb-drug interactions. Synthesis and antimicrobial evaluation of N-(3-benzoxazol-2-yl-phenyl)-acrylamide

N-(3-benzoxazol-2-yl-phenyl)-acrylamide has been synthesized as an expected product by reacting 3-benzoxazol-2-yl-phenylamine (1) with chlo ropropioanyl chloride. The 3-benzoxazol-2-yl-phenylamine (1) was prepared according to reported methods through condensation reaction of 2-aminophenole with 3-aminobenzoic acid in presence of polyphosphoric acid (PPA). The acrylamide derivative 3 was scree ned for antimicrobial activity. The acrylamide 3 showed moderate inhibitory activity for both C. neoformans and leishmania species but the acrylamide 3 has no antibacterial activity. The course of the reaction was found to be of high yield (87%) and the title compound was spectroscopically confirmed and characterized by IR, 13C/1H-NMR, mass spectrometric techniques, and elemental analysis.

Clear structure-activity relationships and a structural model were developed in the studies which provided many such 20? amides that exhibit substantial and some even remarkable enhancements in potency, many that exhibit further improvements in activity against a Pgp overexpressing resistant cancer cell line, and an important subset of the vinblastine analogues that display little or no differential in activity against a matched pair of vinblastine sensitive and resistant (Pgp overexpressing) cell lines. The improvements in potency directly correlated with target tubulin binding affinity, and the reduction in differential functional activity against the sensitive and Pgp overexpressing resistant cell lines was found to correlate directly with an impact on Pgp-derived efflux.

Guanfu base A (GFA) is a novel heterocyclic antiarrhythmic drug isolated from Aconitum coreanum (Lvl.) rapaics and is currently in a phase IV clinical trial in China. However, no study has investigated the influence of GFA on cytochrome P450 (P450) drug metabolism. We characterized the potency and specificity of GFA CYP2D inhibition based on dextromethorphan O-demethylation, a CYP2D6 probe substrate of activity in human, mouse, rat, dog, and monkey liver microsomes. In addition, (+)-bufuralol 19-hydroxylation was used as a CYP2D6 probe for the recombinant form (rCYP2D6), 2D1 (rCYP2D1), and 2D2 (rCYP2D2) activities. Results show that GFA is a potent noncompetitive inhibitor of CYP2D6, with inhibition constant Ki = 1.20 ¡À 0.33 muMin human liver microsomes (HLMs) and Ki = 0.37 ¡À 0.16 muM for the human recombinant form (rCYP2D6). GFA is also a potent competitive inhibitor of CYP2D in monkey (Ki = 0.38 ¡À 0.12 muM) and dog (Ki = 2.4 ¡À 1.3 muM) microsomes. However, GFA has no inhibitory activity on mouse or rat CYP2Ds. GFA did not exhibit any inhibition activity on human recombinant CYP1A2, 2A6, 2C8, 2C19, 3A4, or 3A5, but showed slight inhibition of 2B6 and 2E1. Preincubation of HLMs and rCYP2D6 resulted in the inactivation of the enzyme, which was attenuated by GFA or quinidine. Beagle dogs treated intravenously with dextromethorphan (2 mg/ml) after pretreatment with GFA injection showed reduced CYP2D metabolic activity, with the Cmax of dextrorphan being one-third that of the saline-treated group and area under the plasma concentration-time curve half that of the saline-treated group. This study suggests that GFA is a specific CYP2D6 inhibitor that might play a role in CYP2D6 medicated drug-drug interaction.
