The fluorescence behaviors and properties of three novel photoactive mono-formylated benzoxazole derivatives A-C are found to be affected by different heterocycles and solvents, as reported in a recent experiment (Rodembusch, et al., New J. Chem., 2016, 40, 2785). Unfortunately, the detailed excited-state intramolecular proton transfer (ESIPT) mechanisms of these compounds are lacking. In this study, we used density functional theory (DFT) and time-dependent DFT (TDDFT) methods to study the dynamic ESIPT processes of the three compounds A-C in two different surroundings (polar 1,4-dioxane and nonpolar dichloromethane solvents). The calculated absorption and fluorescence spectra were observed to mutually agree with the experimental data, which indicated that the TDDFT method we adopted was reliable. In addition, based on the analysis of bond lengths, bond angles and IR vibrational spectra in both solvents, it was confirmed that the intramolecular hydrogen bonds (HBs) of these compounds were strengthened in the S-1 state, which could promote the ESIPT reactions. Moreover, the frontier molecular orbitals (MOs) and the maps of the electron density difference between the S-0 and S-1 states displayed intramolecular charge transfer, which provided the probability of ESIPT reactions for the three compounds. Furthermore, based on the constructed potential energy curves, we revealed detailed dynamical ESIPT mechanisms of the compounds A-C. As a consequence, we found that the ESIPT processes were more likely to take place from A (8.48 kcal mol(-1)) B (5.36 kcal mol(-1)) C (0.75 kcal mol(-1)) compounds in the polar 1,4-dioxane solvent, whereas the sequence changed to B (4.01 kcal mol(-1)) A (1.30 kcal mol(-1)) C (1.15 kcal mol(-1)) in the nonpolar dichloromethane solvent. Additionally, it could be determined that the solvent polarity had a tremendous effect on compound A, whereas the effect on compound C was the smallest.

New drugs and molecular targets are urgently needed to address the emergence and spread of drug-resistant tuberculosis. Mycobacterium tuberculosis (Mtb) inosine 5′-monophosphate dehydrogenase 2 (MtbIMPDH2) is a promising yet controversial potential target. The inhibition of MtbIMPDH2 blocks the biosynthesis of guanine nucleotides, but high concentrations of guanine can potentially rescue the bacteria. Herein we describe an expansion of the structure activity relationship (SAR) for the benzoxazole series of MtbIMPDH2 inhibitors and demonstrate that minimum inhibitory concentrations (MIC) of <= 1 mu M can be achieved. The antibacterial activity of the most promising compound, 17b (Q151), is derived from the inhibition of MtbIMPDH2 as demonstrated by conditional knockdown and resistant strains. Importantly, guanine does not change the MIC of 17b, alleviating the concern that guanine salvage can protect Mtb in vivo. Macrophage migration inhibitory factor (MIF) is a key pleiotropic mediator and a promising therapeutic target in cancer as well as in several inflammatory and cardiovascular diseases including pulmonary arterial hypertension (PAH). Here, a novel series of N-(phenylmethyl)-benzoxazol-2-thiones 5-32 designed to target the MIF tautomerase active site was synthesized and evaluated for its effects on cell survival. Investigation of structure-activity relationship (SAR) particularly at the 5-position of the benzoxazole core led to the identification of 31 that potently inhibits cell survival in DU-145 prostate cancer cells and pulmonary endothelial cells derived from patients with idiopathic PAH (iPAH-ECs), two cell lines for which survival is MIF-dependent. Molecular docking studies helped to interpret initial SAR related to MIF tautomerase inhibition and propose preferred binding mode for 31 within the MIF tautomerase active site. Interestingly, daily treatment with 31 started 2 weeks after a subcutaneous monocrotaline injection regressed established pulmonary hypertension in rats.

The compounds nPEDFPBx except nitro-substituted ones have enantiotropic nematic mesophases with mesophase ranges of 16-86 degrees C and 29-108 degrees C on heating and cooling, respectively. The results show that introduction of two lateral fluorine atoms into molecule results in enhanced nematic mesophase stability. Meanwhile, nPEDFPBx displays much higher birefringence (0.507-0.624) than common tolanebased liquid crystals, which is ascribed to its large pi-conjugated molecule composed of benzene, ethynyl, and benzoxazole mesogenic unit. To explore the luminescence properties of silver complexes in the solid state, two Ag(I) complexes, [Ag(4-PBO)(2)(NO3)] {di[2-(4 ‘-pyridyl)-benzoxazole]silver(I) nitrate} (1) and [Ag-2(SPPh3)(4)](ClO4)(2) {[tetra(triphenylphosphine sulfide)disilver(I)] diperchlorate} (2) (4-PBO =2-(4 ‘-pyridyl)-benzoxazole, SPPh3 = triphenylphosphine sulfide), have been synthesized and characterized by elemental analysis, FT-IR and UV-Vis spectroscopy. Single-crystal X-ray diffraction revealed that 1 is three-coordinate by two nitrogen atoms from two 4-PBO ligands and an oxygen atom from the coordinated nitrate anion forming a triangular plane configuration. In 2, the SPPh3 ligands adopt a monodentate coordinated and monoatomic bridging mode to connect two Ag(I) ions, resulting in a three-coordinate symmetrical binuclear structure. In the solid state, the luminescence properties of the ligands 4-PBO, SPPh3 and their complexes were investigated. The results demonstrated that 1 results in the fluorescence quenching of aggregates due to the strong pi-pi stacking effect. The fluorescence enhancement of 2 may be attributed to the aggregation-induced emission (AIE) effect of restricted intramolecular rotations of the peripheral phenyl rings against the central core. In addition, cyclic voltammograms of 1 and 2 indicated an irreversible Ag+/Ag couple.
