At room temperature and with good stirring, sulfuryl chloride (8.8 mL, 109.52 mMole) was added drop by drop over a 20-minute period. As the addition proceeds, additional ethyl acetate (350 mL) was added to keep the mixture stirring. After 1 hour the precipitate was filtered off, washed with ether and air-dried. Yield 10.5 g.

General procedure: To an oven dried 5 mL microwave vessel was addedPd(dppf)Cl2¡¤CH2Cl2 (4 mol%), halide/pseudohalide (1 equiv),boron coupling partner (1 equiv), and Cs2CO3 (3 equiv). Thevessel was then capped and purged with N2 before addition ofCyrene (1 mL, 0.25 M) and H2O (1.8 mL). The reaction mixturewas heated to 50 C and maintained at this temperature withstirring for 5 h before the vessel was vented and decapped. Thesolution was then diluted with Et2O (10 mL) and washed withwater (2 ¡Á 20 mL) and brine (2 ¡Á 20 mL). The organics were thenpassed through a hydrophobic frit and concentrated underreduced pressure to give a residue, which was purified by flashchromatography (silica gel) to afford the title compound.

Thionyl Chloride (540 mL) and Dimethyl formamide (270 mL) were added to a stirred mixture of 5-Chloro-1,3-benzoxazole-2-thiol (540 g, 2.903 moles) and dichloromethane (5 liter) at 5-10 C. and stirred till clear solution was observed. The reaction mixture was then stirred at 10 C. to room temperature for 4 hour. After completion of the reaction, the mixture was poured into cold water (4 liter), neutralized with solid sodium bicarbonate (1440 g) portion wise over a period of 1 hour and extracted with dichloromethane (2*2.5 liter). The combined organic extracts were washed with brine solution, dried over anhydrous sodium sulphate (300 g) and concentrated under reduced pressure to give a crude compound. The crude material was triturated with Hexane (2*2 liter) at -20 C., filtered and and dried under reduced pressure to obtain 2,5-dichloro-1,3-benzoxazole (475 g, 88.9%) as a yellow liquid.

Commercially available 2-fluoro-4-trifluoromethylbenzylamine (0.5 ml, 3.7 mmol) was dissolved in 20ml of AcOEt and at 0C triphosgene (1.12 g, 3.7 mmol) was added to the solution. The mixture was warmed at 80C for 4 hours then evaporated and the residue was dissolved in 5 ml of DMF. The solution of the isocyanate was added drop wise to a solution in DMF (5 ml) of compound lb (360 mg, 2.4 mmol) and the mixture was warmed at 80C for 8 hours. (TLC AcOEt). The solvent was evaporated and the crude was dissolved in AcOEt (30 ml) and washed with water (1 X 20 ml) and brine. The organic phase was dried over sodium sulfate and concentrated under vacuum. The purification of the crude residue by chromatographic column gave 100 mg of a white solid. Yield = 1 1 % ‘HNMR (DMSO, 400 MHz) delta 4.43 (2H, d, J = 6 Hz), 6.99 (3H, m), 7.05 (1H, m), 7.62 (3H, m), 8.53 (1H, bs), 10.98 (1H, bs); [M+1] 370.1 (C 16H1 1F4N3O3 requires 369.27).

The reaction mixture was heated at 60 C for 3 h. After cooling, the reaction mixture was poured into 100 mL water and extracted with ethyl acetate (3 x 70 mL). The combined organic layers were washed with saturated aqueous NaCl and dried over sodium sulfate. The solvent was removed in vacuo, and the residue was purified by flash column chromatography (SiO2) using hexane/ethyl acetate (8:2) as eluent to give 3.6 g (62%) of 2e as a white solid.

The product was recrystallized with dichloromethane (DCM)/n-hexane as an eluent to obtain 8.91 g (yield: 72%) of Intermediate (A). (0346) LC-Mass (calc: 245.12 g/mol, found: M+1=246 g/mol).
