Using ionic liquids (ILs) as the reaction solvent for the synthesis of prepolymer polyamide of poly(p-phenylene benzoxazole) (PBO) was investigated. The optimum condition of prepolymer preparation was determined in ILs. A series of 1,3-dialkylimidazolium ILs were used to be the reaction media of the polycondensation. The relationship between the molecular weight of prepolymer and the structure of ILs was analysed by changing the structure of the cation and species of anion of ILs. Benzothiazole derivatives were used as models to study the excited-state intramolecular proton transfer (ESIPT) from an experimental and theoretical point of view. The experimental electronic and vibrational results were compared with a comprehensive selection of state-of-the-art computational methods in a workflow approach. The latter were performed based on modern techniques, such as DLPNO-CCSD(T), which gives the reference energies and current methodologies for ESIPT analysis, such as molecular dynamics and charge density difference testing. The theoretical vibrational results were focused on the stretch vibrational-mode of the hydroxyl group, which indicated a large increase in the intramolecular hydrogen bond strength, which facilitates the ESIPT process. Theoretically, the optimization of a large number of molecules shows that pi-stacking plays a fundamental role in benzothiazole stabilization, with a remarkably strong intramolecular hydrogen bond. The potential energy surface of the ESIPT reactive benzothiazole (4HBS) has a clear transition state where ESIPT is easily observed with a large difference in energy between the enol and keto tautomer. Additionally, molecular dynamics showed that the ESIPT process occurs very fast. The tautomer appears around 8.7 fs and the enolic form is regenerated in just 24 fs, closing the Forster cycle. The calculated Stokes shift could be related to the ESIPT process and the experimental solid-state emission spectrum matched almost perfectly with the theoretical one. In contrast, for the non-ESIPT benzothiazole (4HBSN), the agreement between theory and experiment was limited, probably due to intermolecular interaction effects that are not considered in these calculations.

The effects of the heteroatom and position on excited-state intramolecular proton transfer (ESIPT) of 2-[4 ‘-(N-4,6-dichloro-1,3,5-triazi-n-2-yl)2 ‘ hydroxyphenyl]benzoxazole (4THBO) have been investigated via time-dependent density functional theory studies. The heteroatoms refer to O and S atoms, and the position effect refers to the N-4,6-trichloro-1,3,5-triazin-2-yl (TCT) substituents in the para and meta positions. The configuration of the four compounds (4THBO, 4THBT, 5THBO and 5THBT) was optimized and the bond lengths, bond angles and infrared spectra of the atoms participating in the proton transfer in the S0 and S1 states were studied. The occurrence of ultrafast ESIPT in the four compounds was demonstrated. Moreover, the potential energy curves of the S0 and S1 states were constructed, and the effects of the heteroatom substitution and substituent position changes on the ESIPT mechanism of the four 4THBO derivatives were analyzed. The results show that the ESIPT barrier of the S atom substitution in the excited state is lower than that of the O atom-substituted molecule, and the energy barrier of the substituent (TCT) in the meta-position is significantly smaller than that in the para-position. These results indicate that the substitution of the S heteroatom promotes the ESIPT of the 4THBO compound and that the substituent (TCT) in the para position is more prone to proton transfer than that in the meta position. Our work could provide a theoretical basis for further experiments.

However, there are no related reports about its anti-inflammatory effects. In the present study, we investigated the anti-inflammatory properties and the underlying molecular mechanism of K313 in lipopolysaccharide (LPS)-induced RAW264.7 macrophages. K313 dose-dependently (5, 10, and 20M) inhibited LPS-stimulated nitric oxide (NO), interleukin (IL)-6, tumor necrosis factor (TNF)-, and 3-nitrotyrosine (3-NT) production and significantly decreased the gene transcription levels of inducible nitric oxide (iNOS), IL-6, and TNF-. In addition, the results showed that the inflammatory cytokines suppressed by K313 were not regulated by p65 NF-B, ERK1/2, AKT, or p38 MAPK. Instead, K313 increased phosphorylation of glycogen synthase kinase-3 beta (GSK-3) (Ser9) resulting in GSK-3 deactivation. Moreover, in LPS-stimulated RAW264.7 macrophages, K313 and lithium chloride (LiCl) had a synergistic effect on the anti-inflammatory response. These results indicated that K313 exhibited anti-inflammatory properties and revealed the potential mechanism. K313 can increase GSK-3 (Ser9) phosphorylation to decrease GSK-3 activation in LPS-induced RAW264.7 macrophages.

Racemic mixtures of alcohols, (+/-)-4-(1,3-benzoxazol-2-ylsulfanyl)butan-2-ol ((+/-)-3a), (+/-)-4-[(5-bromo-1,3-benzoxazol-2-yl)sulfanyl]butan-2-ol ((+/-)-3b), and (+/-)-4-[(5,7-dibromo-1,3-benzoxazol-2-yl)sulfanyl]butan-2-ol ((+/-)-3c), displayed as high activity against M.scrofulaceum, M.intracellulare, M.fortuitum, and M.kansasii as commercially available antituberculosis drug-Isoniazid. The present letter reports unusual high fluorescence anisotropy of excited state intramolecular proton transfer (ESIPT) prone 2-(2′-hydroxyphenyl) benzoxazole (HBO) in water followed by its imidazole (HBI) and thiazole (HBT) analogues. The low fluorescence anisotropy of the acetate compound of HBO in water reveals the key role of the hydrogen bonded cage-like probe-water cluster for the findings in water.
