Radical translocation processes triggered by nitrogen-centered radicals (NCRs), such as 1,5-hydrogen atom transfers (1,5-HAT), demonstrated by the well-established Hofmann-Loffler-Freytag (HLF) reaction, provide an attractive approach for the controllable and selective functionalization of remote inert C(sp(3))-H bonds. Here we report an amidyl radical-triggered site-selective remote C(sp(3))-H heteroarylation of amides under organic photoredox conditions. This approach provides a mild and highly regioselective reaction affording remote C(sp(3))-H heteroarylated amides at room temperature under transition-metal free, weakly basic, and redox-neutral conditions. Non-prefunctionalized heteroarenes, such as purines, thiazolopyridines, benzoxazole, benzothiazoles, benzothiophene, benzofuran, thiazoles and quinoxalines, can be alkylated directly. Sequential and orthogonal C-H functionalization of different heteroarenes by taking advantage pH value or polarity of radicals has also been achieved. DFT calculations explain and can predict the site-selectivity and reactivity of this reaction. This strategy expands the scope of the Minisci reaction and serves as its alternative and potential complement.

We report an efficient palladium-catalyzed approach to the synthesis of benzoxazole derivatives via sequential heteroarylation/acylation reaction of iodobenzenes. The antioxidant activity of the synthesized compounds was evaluated for their antioxidant activities, such as reducing power and ABTS (2,2 ‘-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt) radical scavenging. The results of ABTS radical scavenging activities of some of the synthesized molecules showed higher activities than standard Trolox, SC50(mu M) = 213.04 +/- 18.12. One benzoxazole-coumarin (6f), two benzoxazole-isothiocyanate (7b, 7c), and two benzoxazole-triazole (8b, 8c) derivatives showed higher activities (SC50(mu M) values, 82.07 +/- 10.34, 120.19 +/- 7.30, 104.58 +/- 10.55, 153.26 +/- 7.14, and 144.82 +/- 10.68, respectively) than standard Trolox, (SC50(mu M) = 213.04 +/- 18.12).

We provide a critical perspective of the burgeoning literature on microporous polymers prepared using thermal rearrangement (TR) processes based on the learning derived from analogous chemistry involving small-molecular-weight compounds. TR polymers have shown interesting permeability-selectivity relationships in gas separation and, thus, have generated wide interest as potential membrane materials for industrial applications. The intractable nature of the products obtained by TR processes has precluded rigorous structural elucidation of the polymers. Based on small-molecule chemistry, we conclude that structures are likely to be more complex than generally depicted in the published literature. Interestingly, a simpler chemistry, namely thermal dehydrocyclization (TCD), leads to products identical to those derived from TR, but at significantly lower temperatures. However, TCD chemistry does not involve a skeletal rearrangement of the kind purported in TR during the conversion of imide to oxazole ring resulting in spatially confined heterocyclic ring polymers. Yet, they show similar fractional free-volume elements as exhibited by TR polymers. This is intriguing and points to a need for more careful examination of the factors responsible for microporosity in such materials. TR chemistry as currently practiced appears limited to only benzoxazole-type structures. The ability to precisely control and reproducibly produce materials with well-defined structure and properties will be a key to large-scale manufacture and industrial applications of such materials. A novel gas separation membrane was fabricated from a commercially available polymer of phenolphthalein-based cardo poly (arylene ether ketone) (PEK-C). Thermal treatment was employed to improve the gas permeability and anti-plasticization property of PEK-C polymeric membrane via inducing interchain crosslinking. The changes of chemical structure, thermal crosslinking reaction, interchain distance, gas separation performance and anti-plasticization properties of PEK-C membranes were investigated by the FT-IR, XPS, TG-MS, XRD and gas permeation tests. Results show that the gas separation performance and anti-plasticization properties of PEK-C membranes are significantly enhanced after the thermal crosslinking, which is induced by the decomposition of the lactone rings in cardo moieties and crosslinking with the formation of biphenyl linkages. The dramatic enhancement in gas permeability for the crosslinked membrane is attributed to the enlargement of the interchain distance and free volume cavity, and the great improvement on the anti-plasticization property is due to the formation of a rigid crosslinked network structure. Compared to PEK-C polymeric membrane, the CO2 permeability of the crosslinked membrane increased by more than 110 times with an adequate CO2/CH4 selectivity, especially for the separation of CO2/CH4 mixed gas (50:50 mol%). The CO2 plasticization pressure substantially increased from 2 atm to the highest tested pressure of 30 atm. The gas separation performance of the crosslinked membrane surpassed the 2008 upper bound for CO2/CH4, exhibiting that the thermal crosslinking membrane derived from PEK-C membrane material is an attractive candidate for the natural gas purification.

The search for novel targeted inhibitors active on glioblastoma multiforme is crucial to develop new treatments for this unmet clinical need. Herein, we report the results from a screening campaign against glioma cell lines using a proprietary library of 100 structurally-related pyrazolopyrimidines. Data analysis identified a family of compounds featuring a 2-amino-1,3-benzoxazole moiety (eCF309 to eCF334) for their antiproliferative properties in the nM range. These results were validated in patient-derived glioma cells. Available kinase inhibition profile pointed to blockade of the PI3K/mTOR pathway as being responsible for the potent activity of the hits. Combination studies demonstrated synergistic activity by inhibiting both PI3Ks and mTOR with selective inhibitors. Based on the structure activity relationships identified in this study, five new derivatives were synthesized and tested, which exhibited potent activity against glioma cells but not superior to the dual PI3K/mTOR inhibitor and lead compound of the screening eCF324.
