Quantum Mechanical Studies of Three Aromatic Halogen-Substituted Bioactive Sulfonamidobenzoxazole Compounds with Potential Light Harvesting Properties

Three different organic compounds, 2-phenyl-5-(4-trifloromethyl phenyl sulfonamido) benzoxazole (PTPS), 2-(4-chlorobenzyl)-5-(2,4-dinitrophenylsulfonamido)benzoxazole (CNSB) and 2-(4-fluorobenzyl)-5-(2,4-dinitrophenylsulfonamido)benzoxazole (FBPS), were synthesized. To find their energetically stable conformation, geometry optimization was done using density functional theory with the level B3LYP/cc-pVDZ. Electron distribution of the system was studied using molecular electrostatic potential map. Different intermolecular interactions arising from hyperconjugative effect were investigated using the natural bond orbital (NBO) formalism. Nonlinear optical properties were further studied using first-order hyperpolarizability values. The three compounds may be important in the development of novel inhibitor molecules of Topoisomerase II enzyme, as lead compounds. Light harvesting efficiency of PTPS is 0.9342, which shows that it is having potential applications in the design of new DSSC’s.

CuNiFe a Magnetic Nano-Catalyst: an Efficient Catalyst for the Selective Synthesis of Benzoxazoles

An efficient protocol for the synthesis of benzoxazoles from o-nitrophenols and benzyl alcohols using CuNiFe as a magnetic nano-catalyst via hydrogenation transfer has been described. The CuNiFe magnetic nano-catalyst was prepared by an operationally simple method, without the use of any toxic reagents. The prepared catalyst was characterized by XRD, FEG-SEM, EDAX, TEM, and FT-IR techniques. No additional additives such as oxidants and reductants were required for this reaction. CuNiFe magnetic nano-catalyst showed good air stability and could be recycled up to six times without any significant loss in its catalytic activity.

The small molecule AU14022 promotes colorectal cancer cell death via p53-mediated G2/M-phase arrest and mitochondria-mediated apoptosis

The p53 tumor suppressor plays critical roles in cell cycle regulation and apoptotic cell death, with its activation capable of sensitizing cancer cells to radiotherapy or chemotherapy. To identify small molecules that induce apoptosis via increased p53 transcriptional activity, we used a novel in-house library containing 96 small-molecule compounds. Using a cell-based screening method with a p53-responsive luciferase-reporter assay system involving benzoxazole derivatives, we found that AU14022 administration significantly increased p53 transcriptional activity in a concentration-dependent manner. Treatment with AU14022 increased p53 protein expression, p53 Ser15 phosphorylation, p53-mediated expression of downstream target genes, and apoptosis in p53-wild-type HCT116 human colon cancer cells, but not in p53-knockout HCT116 cells. Additionally, p53-wild-type HCT116 cells treated with AU14022 exhibited mitochondrial dysfunction, including modulated expression of B-cell lymphoma-2 family proteins and cytochrome c release. Combination treatment with AU14022 and ionizing radiation (IR) synergistically induced apoptosis as compared with IR or AU14022 treatment alone, with further investigation demonstrating that cell cycle progression was significantly arrested at the G2/M phase following AU14022 treatment. Furthermore, in a mouse p53-wild-type HCT116 colon cancer xenograft model, combined treatment with AU14022 and IR inhibited tumor growth more effectively than radiation alone. Therefore, AU14022 treatment induced apoptosis through p53-mediated cell cycle arrest involving mitochondrial dysfunction, leading to enhanced radiosensitivity in colon cancer cells. These results provide a basis for further assessments of AU14022 as a promising anticancer agent.

Discovery of novel series of 2-substituted benzo[d]oxazol-5-amine derivatives as multi-target directed ligands for the treatment of Alzheimer’s disease

Alzheimer’s disease (AD) is associated with multifactorial neuropathological conditions, which include cholinergic deficit, amyloid-beta plaques formation, loss of neuronal plasticity and neuronal death. Treating such multifactorial conditions with a single target directed approach is considered to be inadequate. Accordingly, multi-target directed ligand (MTDL) strategy has been evolved as an auspicious approach for the treatment of AD. In light of that, a library of 2-substituted benzo[d]oxazol-5-amine derivatives (29-39; 86-107) was designed using the scaffold hopping guided MTDLs strategy, synthesized and evaluated through various in-vitro and in-vivo biological studies. The optimal compound 92 exhibited potent inhibitory activities against AChE (IC50 = 0.052 +/- 0.010 mu M), BuChE (IC50 = 1.085 +/- 0.035 mu M), and significant amyloid-beta aggregation (20 mu M) inhibition. The compound possessed better blood-brain barrier permeability (Pe = 10.80 +/- 0.055 x 10(-6) cm s(-1)) in PAMPA assay and neuro protective properties (40 mu M) on SH-SY5Y neuroblastoma cell lines. Furthermore, in-vivo behavioural studies were performed on Y-maze test (scopolamine-induced amnesia model) and Morris water maze test (A beta(1-42) induced ICV rat model). The compound 92, at a dose of 10 mg/kg oral administration, demonstrated a substantial improvement of the cognitive and special memory impairment. In summary, both in-vitro and in-vivo investigations evidenced that compound 92 was a potential lead for the discovery of safe and effective disease-modifying agents for AD. Catalytic C-H Bond Alkylation of Azoles with Alkyl Halides Mediated by Nickel(II) Complexes of Phenanthridine-Based N boolean AND N-boolean AND N Pincer Ligands

Synthesis and properties of novel 6,7-dihydrocyclopenta[5,6-b]benzofuran-based liquid crystal compounds

A novel liquid crystal core unit 6,7-dihydrocyclopenta[5,6-b]benzofuran was designed and three different kinds of liquid crystals2MUF, 2MP4, 2MYO2were synthesised through a facile route which employed palladium-catalysed cascade Sonogashira coupling/cyclisation as a key step. Initially, density function theory calculation illustrated that a compound based on this new unit possessed almost a linear structure. The mesophase and physical properties of these novel compounds were measured and the impacts of the newly designed core unit on these properties were investigated by comparing with two series of analogous materials which contained classical mesogenic cores 1,4-disubstituted benzene andtrans-cyclohexyl benzene, finding that the introduction of 6,7-dihydrocyclopenta[5,6-b]benzofuran into liquid crystals tended to promote the formation of nematic phase while increasing both the melting points and the clearing points of target molecules. Meanwhile, the newly synthesised liquid crystals presented higher birefringences when compared with their corresponding reference compounds, indicating that 6,7-dihydrocyclopenta[5,6-b]benzofuran can be a hopeful candidate for the construction of high birefringence liquid crystals.
