The reaction of 2,6-bis(benzoxazolyl)-4-tert-butylphenol (HL) with [(BuxSnCl4-x)-Bu-n] (x = 0, 1) in 1:1 stoichiometry yielded the tin coordination complexes [(HL)(SnBuxCl4-x)-Bu-n] [x = 0 (1); x = 1 (2)]. Deprotonation of HL was performed using reagents having groups with high basicity such as (BuLi)-Bu-n or [Sn{N(SiMe3)(2)}(2)]. These basic reagents prompted the coordination of the ligand in its anionic form, yielding the complexes [(thf)(2)Li(L)SnCl4] (3) and [(L)Sn{N(SiMe3)(2)}] (4), respectively. The molecular structure of HL displayed an intramolecular hydrogen bond OH-N and a planar arrangement of the bis(benzoxazolyl)phenolic system. In the molecular structures of both complexes containing HL an intramolecular hydrogen bond of NH-O type was also present. The coordination of the ligand in either neutral or anionic form is described by a kappa O,kappa N chelate mode toward Sn. All complexes displayed bis(benzoxazolyl)phenolic moieties close to planar; the least planar system was observed in 4 that was also studied by DFT methods. [GRAPHICS] .

The effects of TR-able codiamine on the processing ability of oligoimides, molecular packing and properties, including thermal stability, dielectric property and bonding ability to reinforcing fibers, for the resulting poly(imide-co-benzoxazole) (PI-co-PBOs) resins have been examined in detail. These thermally rearranged PI-co-PBO resins exhibit reduced dielectric constants of 2.56-3.4 at f = 0.1 GHz, lower than the cured PI with a dielectric constant of 3.55. Meanwhile, the 5 wt% weight loss temperature (T-d5) of the resultant resins increases from 455 degrees C for the cured PI to 491 degrees C for the PI-co-PBO-60 containing 60 mol% benzoxazole unit, and correspondingly, the tensile modulus of PI-co-PBO-60 reaches 2.47 GPa, which is 76% higher than that of the cured PI. These resultant PI-co-PBOs exhibit the combined excellent properties, indicating them a great potentials as the new low-dielectric constant polymer resins in microelectronic industries or advanced composites.

A novel series of benzoxazolyl linked benzylidene based rhodanine and their cyclic analogs were synthesized, characterized and evaluated for their alpha-amyloglucosidase inhibitory activity. Out of eight target compounds, two compounds (4b and 5b) displayed potent inhibitory activity against alpha-amyloglucosidase with IC50 values in the range of 0.24 +/- 0.01-0.94 +/- 0.01 A mu M as compared to standard drug acarbose. Mixed-matrix membrane (MMM) is an effective way to overcome trade-off limitations of conventional polymeric membranes. Herein, a novel organic porous nanosphere (TC-cPSB), which is prepared by the polycondensation of 9,9-bis(4-aminophenyl) fluorene (BAFL) and terephthalaldehyde (TPAL) followed by thermal crosslinking, is chosen to engineer the polymer/filler interface. Benefiting from strong intermolecular interaction (pi-pi stacking and hydrogen bonding), the TC-cPSB nanosphere can well disperse in TR-PBO matrix with a defect-free interface. With an increase in TC-cPSB loading, well-designed MMMs exhibit a significant anti-trade-off’ phenomenon whereby gas permeability and selectivity increase simultaneously, following the trend predicted by the Maxwell model. Compared with TR-PBO membrane, the MMM containing 15 wt% of nanosphere shows an increase of 282% and 217.6% in H-2/CO2 selectivity and H-2 permeability, respectively, which is far beyond 2008 Robeson upper bound.

To develop novel CNS penetrant HDAC inhibitors, a new series of HDAC inhibitors having benzoheterocycle were designed, synthesized, and biologically evaluated. Among the synthesized compounds, benzothiazole derivative 9b exhibited a remarkable anti-proliferative activity (GI(50) = 2.01 mu M) against SH-SY5Y cancer cell line in a dose and time-dependent manner, better than the reference drug SAHA (GI(50) = 2.90 mu M). Moreover, compound 9b effectively promoted the accumulation of acetylated Histone H3 and alpha-tubulin through inhibition of HDAC1 and HDAC6 enzymes, respectively. HDAC enzyme assay also confirmed that compound 9b efficiently inhibited HDAC1 and HDAC6 isoforms with IC50 values of 84.9 nM and 95.9 nM. Furthermore, compound 9b inhibited colony formation capacity of SH-SY5Y cells, which is considered a hallmark of cell carcinogenesis and metastatic potential. The theoretical prediction, in vitro PAMPA-BBB assay, and in vivo brain pharmacokinetic studies confirmed that compound 9b had much higher BBB permeability than SAHA. In silico docking study demonstrated that compound 9b fitted in the substrate binding pocket of HDAC1 and HDAC6. Taken together, compound 9b provided a novel scaffold for developing CNS penetrant HDAC inhibitors and therapeutic potential for CNS-related diseases.

The NAD(+)- dependent deacetylase SIRT1, which is associated with the improvement of metabolic syndromes, such as type 2 diabetes, is a well-known longevity-related gene. Several in vitro and in vivo studies have shown the known protective effects of SIRT1 activators, such as resveratrol and SRT1720, on diabetes-or obesity-induced fatty liver and insulin resistance. Here, we newly synthesized 18 benzoxazole hydrochloride derivatives based on the structure of resveratrol and SRT1720. We performed an in vitro SIRT1 activity assay to identify the strongest SIRT1 activator. The assay confirmed MHY2233 to be the strongest SIRT1 activator (1.5-fold more potent than resveratrol), and docking simulation showed that the binding affinity of MHY2233 was higher than that of resveratrol and SRT1720. To investigate its beneficial effects, db/db mice were orally administered MHY2233 for 1 month, and various metabolic parameters were assessed in the serum and liver tissues. MHY2233 markedly ameliorated insulin signaling without affecting body weight in db/db mice. In particular, the mRNA expression of lipogenic genes, such as acetyl CoA carboxylase, fatty acid synthase, and sterol regulatory element-binding protein, which increased in db/db mice, decreased following oral treatment with MHY2233. In conclusion, the novel SIRT1 activator MHY2233 reduced lipid accumulation and improved insulin resistance.
