Three styrylbenzoxazole derivatives with terminal dicyanomethylene group were studied in order to understand how minor modifications brought to the benzoxazole ring influence the photoluminescence (PL) properties. To determine its potential clinical application, the synergistic effect of KZ-001 with other agents was investigated both in vitro and in vivo (xenograft models). KZ-001 exhibited synergistic anti-cancer effect in combination with BRAF inhibitor vemurafenib and a microtubule-stabilizing chemotherapeutic agent docetaxel. In addition, KZ-001 inhibited the MAPK pathway like known MEK inhibitors. In summary, KZ-001, a structurally novel benzoxazole compound, was developed as a MEK inhibitor that has potential for cancer treatment.

Results Group comparisons of A beta deposition as determined by BF-227 uptake using PET imaging showed no significant differences between MCI and AD. A beta deposition was significantly higher in MCI and AD than in NC. The correlation analysis between local A beta deposition and gray matter atrophy showed that in AD, the A beta deposition in the inferior temporal gyrus was strongly related to the gray matter atrophy in this region. On the contrary, the A beta deposition in the precuneus was associated with the atrophy in the right occipital-temporal region. In the NC, the A beta deposition in the inferior temporal gyrus was associated with the atrophy in the precuneus. Conclusion In the AD, the relationship between the A beta deposition and local atrophy is area-dependent. In NC, A beta deposition in the inferior temporal gyrus correlated to the atrophy in the precuneus.

Background: Benzoxazole is the most important class of heterocyclic compound in medicinal chemistry. It has been incorporated in many medicinal compounds making it a versatile heterocyclic compound that possess a wide spectrum of biological activities. Results: The molecular structures of synthesized benzoxazole derivatives were confirmed by physicochemical and spectral means. The synthesized compounds were further evaluated for their in vitro biological potentials i.e. antimicrobial activity against selected microbial species using tube dilution method and antiproliferative activity against human colorectal carcinoma (HCT 116) cancer cell line by Sulforhodamine B assay. Conclusion: In vitro antimicrobial results demonstrated that compounds 4, 5, 7 and 16 showed promising antimicrobial potential. The in vitro anticancer activity indicated that compounds 4 and 16 showed promising anticancer activity against human colorectal cancer cell line (HCT 116) when compared to standard drug and these compounds may serve as lead compound for further development of novel antimicrobial and anticancer agents.

In the present study, we attempted to develop novel class of Mycobacterium tuberculosis (Mtb) inhibitors by exploring the pharmaceutically underexploited enzyme targets which are majorly involved in cell wall biosynthesis of mycobacteria. For this purpose glutamate racemase was selected which racemizes D-glutamate from L-glutamate, a key step in peptidoglycan synthesis. Furthermore, enzyme is neither expressed nor its product, n-glutamate is produced in mammals, and hence inhibiting this enzyme will have no vulnerable effect in host organism. A library of our in-house compounds were screened against glutamate racemase using a biophysical technique; thermal shift assay and further by enzyme inhibition assay to identify Lead 1 molecule. Lead 1 optimization and expansion resulted in twenty four compounds. Among the synthesized compounds twelve compounds shown good enzyme inhibition than Lead 1 (IC50 20.07 +/- 0.29 mu M). Among all the compounds; compound 22 (IC50 1.1 +/- 0.52 mu M) showed potent non-competitive mode of inhibition in enzyme assay. Further showed good susceptibility (in replicating bacteria) of MIC 8.72 mu M and bactericidal time dependant kill on dormant culture. It also exhibited significant activity in Mtb nutrient starvation model (2.5) and Mtb biofilm model (2.4) and in vivo M. marinum infected Zebra fish model studies (3.6) reduction at logarithmic scale.
