An unexpected BF3.OEt2-catalyzed Michael addition/cyclization/dehydration sequence of ortho-hydroxyenaminones with aromatic or aliphatic aldehyde-derived beta,gamma -unsaturated alpha -ketoesters has been achieved to afford a wide range of fused hydrogenated benzoxazole polycycles in 67-95% yields in a highly diastereoselective manner. When isatin-derived beta,gamma -unsaturated alpha -ketoesters were employed as substrates, completely different reactivities were observed, delivering an array of new atropisomers with an oxindole-substituted N-C axially chiral pyrrole-phenol backbone in up to 97% yield. Chemical synthesis and molecular modeling of novel substituted N-1,3-benzoxazol-2yl benzene sulfonamides as inhibitors of inhA enzyme and Mycobacterium tuberculosis growth

Tuberculosis (TB) is one of the major contagious diseases with high mortality which is caused by Mycobacterium tuberculosis (Mtb) pathogen. Due to the existing antibiotic resistance (MDR-TB) to tuberculosis, the demand for the development of new potential chemotherapy drugs is increasing. Herein, we report synthesis of two novel benzoxazole-based series, namely 2-phenyl benzoxazole sulfonamide and 2-piperidine-benzoxazole sulfonamides. These compounds were evaluated for their antimycobacterial activity against Mycobacterium tuberculosis H37Rv strain, using the microplate alamarBlue assay. Molecular docking studies were carried out to comprehend the binding mode of the compounds. It is evident from molecular docking studies and minimum inhibitory concentration assay (MIC) that 2-phenyl benzoxazole sulfonamide scaffold has a greater potential of antitubercular activity possibly by ENR inhibition (inhA inhibitors). Molecular properties of 5-(1H-Benzo[D]Oxa, thia, imid azole-2-Yl)-2-methyl quinazolin-4-ol fluorescent brighteners: Theoretical and experimental approach

A series of seven new fluorescent brighteners 5-(1H-benzo[d]oxazole, thiazole, imidazole-2-yl)-2-methyl quinazolin-4-ol were synthesized by chemical reaction of 2-Methyl-4-oxo-3,4-dihydroquinazoline-5-carboxylic acid and derivatives of 2-aminophenol/2-aminothiophenol/1,2-phenylenediamine attributes to its computational and experimental comparison. Density Functional Theory (DFT) and Time-Dependent Density Functional Theory (TD-DFT) calculations was performed by PCM model maintaining C-1 symmetry in ground state both in gas and DMF solvent. Halo-Bridged Abnormal NHC Palladium(II) Dimer for Catalytic Dehydrogenative Cross-Coupling Reactions of Heteroarenes

This work describes the dehydrogenative coupling of heteroarenes using a dimeric halo-bridged palladium(II) catalyst bearing an abnormal NHC (aNHC) backbone. A benzoxazole compound as a novel MEK inhibitor for the treatment of RAS/RAF mutant cancer

Mutations in RAS/RAF occur in large portion of malignancies and are associated with aggressive clinical behaviors and poor prognosis. Therefore, we developed a novel benzoxazole compound (KZ-001) as a highly potent and selective MEK 1/2 inhibitor. Our efforts were focused on enhancing the activity of the known MEK inhibitor AZD6244 and overcoming the shortcomings existing in current MEK inhibitors. Here we show that compound KZ-001 exhibits approximately 30-fold greater inhibition against BRAF- and KRAS-mutant tumor cells than that of AZD6244. These results were also demonstrated using in vivo xenograft models. Furthermore, pharmacokinetics (PK) analysis was performed for KZ-001, and this compound showed good orally bioavailability (28%) and exposure (AUC(0-infinity) = 337 +/- 169 ng h/mL). To determine its potential clinical application, the synergistic effect of KZ-001 with other agents was investigated both in vitro and in vivo (xenograft models). KZ-001 exhibited synergistic anti-cancer effect in combination with BRAF inhibitor vemurafenib and a microtubule-stabilizing chemotherapeutic agent docetaxel. In addition, KZ-001 inhibited the MAPK pathway like known MEK inhibitors. In summary, KZ-001, a structurally novel benzoxazole compound, was developed as a MEK inhibitor that has potential for cancer treatment.

One-Pot Multicomponent Reaction of Catechols, Ammonium Acetate, and Aldehydes for the Synthesis of Benzoxazole Derivatives Using the Fe(III)-Salen Complex

The Fe(III)-salen complex has been applied successfully as a catalyst for the novel, simple, efficient, and one-pot multicomponent synthesis of benzoxazole derivatives from catechols, ammonium acetate as the nitrogen source, and aldehydes (nontoxic and cheap alternatives of amines) for the first time. Using this procedure, a wide range of benzoxazoles was successfully synthesized in the presence of a catalyst in EtOH under mild conditions, and all products were obtained in excellent yields. To the best of our knowledge, this method is the first example of the multicomponent synthesis of benzoxazole derivatives using these starting materials. The notable features such as the use of air that is considered as a benign oxidant and EtOH as a green solvent, ease of product separation, readily available and inexpensive aldehydes, and mild conditions make our procedure more efficient and practical for organic synthesis. Moreover, the current protocol is successfully applied to synthesize desirable products on a large scale.

Quantum-chemical modeling, spectroscopic (FT-IR, excited states, UV/Vis, polarization, and Dichroism) studies of two new benzo[d]oxazole derivatives

In the given research, the molecular structures of the two new benzo[d]oxazole derivatives including H-I and H-2: have been studied with the use of density functional theory (DFT/B3LYP and M062X) in dimethylformamide (DMF) for the first time. Cross-linkedpoly(benzoxazole-co-siloxane) networks with high thermal stability and low dielectric constant based on a newortho-amidefunctional benzoxazine

In this study, an amide functionalized bis-benzoxazine (AI-al) has been synthesized using allylamine,ortho-amide functional bis-phenol and paraformaldehyde as raw materials via Mannich condensation. This newly obtained benzoxazine has been used to react with polydimethylsiloxane (PDMS) through hydrosilylation to form poly(benzoxazine-co-amide-co-siloxane) (AI-al-PDMS) featuring siloxane, amide and benzoxazine as repeating units. The chemical structures of both oxazine ring-containing monomer and copolymer are confirmed by NMR and FT-IR spectroscopies. Besides, the thermally activated polymerization behaviors of AI-al and AI-al-PDMS are investigated by DSC, and the subsequent conversion of benzoxazole formation is studied by in situ FT-IR. Moreover, dynamic mechanical analysis and thermogravimetric analysis are used to determine the thermal properties of the cross-linked polymers. The resulting cross-linked poly(benzoxazole-co-siloxane) derived from AI-al-PDMS shows excellent thermal stability (noT(g)can be observed before 400 degrees C; Td5 of 393 degrees C) and low dielectric constants (2.52-2.13 in the frequency range of 1 Hz to 1 MHz), evidencing its great potential applications in electronic packing, aerospace, and other high-performance fields.
