New non-protienogenic fluorescent amino acids: Benzoxazol-5-yl-alanine derivatives containing acetylene unit. Synthesis, spectral and photophysical properties

New derivatives of non-proteinogenic amino acids benzoxazol-5-yl-alanine containing substituted acetylene derivative were synthesized according to Sonogashira coupling reaction. All of the obtained compounds are fluorescent. They are characterized by high or moderate molar absorption coefficients, large Stokes shifts, high fluorescence quantum yields and very high brightness. All of these parameters as well as the positions of absorption and emission bands depend on the type and size of substituent and the solvent polarity. Their high brightness enables working with low concentrations, simple and easy detection of spectral absorption and fluorescence analyzes. Moreover, amino acid part of studied compounds allow to use them as covalently attached to a peptide or protein fluorescent probes in biological system studies.

Synthesis and Antimicrobial Activity of 2-Trifluoroacetonylbenzoxazole Ligands and Their Metal Complexes

Three 2-fluoroacetonylbenzoxazole ligands 1a-c and their new Zn(II) complexes 2a-c have been synthesized. In addition, syntheses of new metal [Mg(II), Ni(II), Cu(II), Pd(II), and Ag(I)] complexes from la have been also described. The molecular and crystal structures of six metal complexes 2b and 2d-h were determined by single-crystal X-ray diffraction analyses. Their antibacterial activities against six Gram-positive and six Gram-negative bacteria were evaluated by minimum inhibitory concentrations (MIC), which were compared with those of appropriate antibiotics and silver nitrate. The results indicate that some metal compounds have more antibacterial effects in comparison with free ligands and have preferred antibacterial activities that may have potential pharmaceutical applications. Noticeably, the Ag(I) complex 2h exhibited low MIC value of 0.7 mu m against Pseudomonas aeruginosa, which was even superior to the reference drug, Norfloxacin with that of 1.5 mu m. Against P. aeruginosa, 2h is bacteriostatic, exerts the cell surface damage observed by scanning electron microscopy (SEM) and is less likely to develop resistance. The new 2h has been found to display effective antimicrobial activity against a series of bacteria.

Two luminescent pseudo-polymorphic cuprous complexes with different optical properties: Synthesis, characterization and TD-DFT calculations

Two luminescent pseudo-polymorphs based on a new cuprous complex, [Cu(2-iQBO)(POP)]PF6 (2-iQBO = 2-(1′-isoquinolyl)benzoxazole, POP = bis[2-(diphenylphosphino)phenyl]ether), have been synthesized and characterized by UV-Vis, elemental analysis and single-crystal X-ray diffraction analyses. The pseudo-polymorphs 1 and 2 are characterized as a dichloromethane solvate and an acetonitrile solvate, respectively. Single-crystal X-ray diffraction data reveal that 1 and 2 are both mononuclear cuprous complexes with a tetrahedral CuN2P2 coordination configuration, consisting of two P atoms from the POP ligand and two N atoms from the 2-iQBO ligand. The pseudo-polymorphic complexes 1 and 2 can be distinguished by their crystal color, UV-Vis spectra and PL spectra. Time-dependent density functional theory (TD-DFT) calculations reveal that the P-Cu-P angle is a key structural factor influencing the absorption and emission properties. Based on the TD-DFT calculations, the lowest singlet excited state corresponding to the UV-Vis absorption edge can be definitely assigned as a metal-to-ligand charge transfer (MLCT) mixed with a ligand-to-ligand charge transfer (L’LCT), and the lowest triplet excited states should be both assigned as a mixed (3)(ML + L’L)CT character. Molecularly imprinted covalent organic polymers for the selective extraction of benzoxazole fluorescent whitening agents from food samples

Molecularly imprinted covalent organic polymers were constructed by an imine-linking reaction between 1,3,5-triformylphloroglucinol and 2,6-diaminopyridine and used for the selective solid-phase extraction of benzoxazole fluorescent whitening agents from food samples. Binding experiments showed that imprinting sites on molecularly imprinted polymers had higher selectivity for targets compared with those of the corresponding non-imprinted polymers. Parameters affecting the solid-phase extraction procedure were examined. Under optimal conditions, actual samples were treated and the eluent was analyzed with high-performance liquid chromatography with diode-array detection. The results showed that the established method has a wide linearity, satisfactory detection limits and quantification limits, and acceptable recoveries. Thus, this developed method possesses the practical potential for the selective determination of benzoxazole fluorescent whitening agents in complex food samples.

Prediction of circulating human metabolites of pemafibrate, a novel antidyslipidemic drug, using chimeric mice with humanized liver

Pharmacokinetics and metabolism of recently launched antidyslipidemic drug pemafibrate ((2R)-2-[3-({1,3-benzoxazol-2-yl[3-(4-methoxyphenoxy)propyl]amino}methyl)phenoxy]butanoic acid) was investigated in chimeric mice with humanized liver in the present study. The plasma unbound fractions of [C-14]pemafibrate in mice (0.0046-0.0048) were higher than those in monkeys and humans (0.0015-0.0022). In chimeric mice with humanized liver intravenously treated with pemafibrate at 1.0 mg/kg body weight, the pharmacokinetic parameters (CLtotal, V-ss and AUC(0-inf)) of unbound pemafibrate in chimeric mice with humanized liver were more similar to those reported in monkeys and humans than those in control mice. High concentrations of N-dealkylated form (M4) and benzoxazole 6-hydroxylated form (M6) of pemafibrate in plasma were observed as the main circulating metabolites in chimeric mice with humanized liver treated with pemafibrate. Moreover, the concentrations of other specified metabolites of pemafibrate were much higher in chimeric mice with humanized liver than in control mice. These results suggest that there are species differences in the pharmacokinetics of pemafibrate in vivo between mice tested and humans reported. Moreover, chimeric mice with humanized liver seem to be a beneficial animal model for further studies to predict the circulating human metabolites of pemafibrate and their pharmacokinetics.

Synthesis of 4-Alkenyl Benzoxazoles via Pd-catalyzed ortho C-H Functionalization of 2-Amidophenols

A one-pot direct transformation to remotely C-H alkene functionalized 2-aryl benzoxazoles from the reaction of amidophenol and electronically deficient olefin was reported. Design, synthesis and biological potential of heterocyclic benzoxazole scaffolds as promising antimicrobial and anticancer agents

Background: Benzoxazole is the most important class of heterocyclic compound in medicinal chemistry. It has been incorporated in many medicinal compounds making it a versatile heterocyclic compound that possess a wide spectrum of biological activities. Results: The molecular structures of synthesized benzoxazole derivatives were confirmed by physicochemical and spectral means. The synthesized compounds were further evaluated for their in vitro biological potentials i.e. antimicrobial activity against selected microbial species using tube dilution method and antiproliferative activity against human colorectal carcinoma (HCT 116) cancer cell line by Sulforhodamine B assay. Conclusion: In vitro antimicrobial results demonstrated that compounds 4, 5, 7 and 16 showed promising antimicrobial potential. The in vitro anticancer activity indicated that compounds 4 and 16 showed promising anticancer activity against human colorectal cancer cell line (HCT 116) when compared to standard drug and these compounds may serve as lead compound for further development of novel antimicrobial and anticancer agents.
