Also, (E)-2-(5-(benzo[d][1,3]dioxol-5-yl)-3-(naphthalen-1-yl)-4,5 dihydro-1H-pyrazol-1-yl)-4-methyl-5-(substituted phenyldiazenyl)thiazole (11) were prepared through reactions of carbothioamide (5a,b) with hydrazonoyl halides. In addition, thioamides (5a-b) were used as starting materials for preparation of thiazoles (12a-b) and benzylidene thiazoles (13a-b). Most of synthesized compounds show interesting biological properties as antimicrobial and antiproliferative activities, the results of minimum inhibitory concentration showed that pyrazole derivative 7c (MIC: 0.23 mg/mL) showed better results when compared with 11c and 12a (MIC: 0.1-0.125 mg/mL) as obtained from their MIC values. On the other hand, 2-(5-(benzo[d][1,3]dioxol-5-yl)-3-(naphthalen-2-yl)-4,5-dihydro-1H-pyrazol-1-yl)-4-(4-chlorophenyl) thiazole (7c) can be considered as the most promising anti-proliferative agent against HCT-116 cancer cells owing to its notable inhibitory effect on HCT-116 cells with an IC50 value of 6.19 mu M.

Synthesis and properties of a novel highly thermal stable N-propargyl monomer containing benzoxazole ring

A novel highly thermal stable propargyl functional compound containing benzoxazole ring, N, N, N, N-tetra propargyl-5-amino-2-(p-aminophenyl) benzoxazole (TPAPB), was proposed and synthesized using a phase-transfer catalytic method. The cure behavior of TPAPB was investigated by non-isothermal differential scanning calorimetry analysis. The solubility and rheological properties of TPAPB, as well as its broad temperature window from 130 degrees C to 200 degrees C with low viscosity, offered excellent processability for TPAPB to be used as a potential monomer of thermosetting polymer resin. It was found that the glass transition temperature of cured TPAPB was 359 degrees C, and the temperature of 5% weight loss was 418 degrees C in argon with the char residue up to 70% at 700 degrees C. The polymerized resin exhibited high heat resistance and thermal stability, together with its processability, making it good candidate as highly heat-resistant polymer matrix for advanced composite applications.

Design, synthesis, antimicrobial activity and computational studies of novel azo linked substituted benzimidazole, benzoxazole and benzothiazole derivatives

Metal-free selective synthesis of 2-substituted benzimidazoles catalyzed by Bronsted acidic ionic liquid: Convenient access to one-pot synthesis of N-alkylated 1,2-disubstituted benzimidazoles

A novel efficient method for the selective synthesis of 2-substituted benzimidazoles is described through condensation reaction of o-phenylenediamines with a wide rang of aliphatic, aromatic and heteroaromatic aldehyde substrates using Bronsted acidic ionic liquid as a reusable catalyst under metal-free conditions at ambient temperature. Notably, Dodecylimidazolium hydrogen sulfate ([Dodeclm][HSO4]) is the most efficient catalyst for good to excellent yields of the corresponding products (up to 98%). Subsequently, this protocol was successfully applied for the preparation of N-alkylated 1,2-disubstituted benzimidazoles in high to excellent yields via sequential one-pot reaction. Encapsulation of Risperidone by Methylated beta-Cyclodextrins: Physicochemical and Molecular Modeling Studies

Risperidone (RSP) is an atypical antipsychotic drug which acts as a potent antagonist of serotonin-2 (5TH2) and dopamine-2 (D2) receptors in the brain; it is used to treat schizophrenia, behavioral and psychological symptoms of dementia and irritability associated with autism. It is a poorly water soluble benzoxazole derivative with high lipophilicity. Supramolecular adducts between drug substance and two methylated beta-cyclodextrins, namely heptakis(2,6-di-O-methyl)-beta-cyclodextrin (DM-beta-CD) and heptakis(2,3,6-tri-O-methyl)-beta-cyclodextrin (TM-beta-CD) were obtained in order to enhance RSP solubility and improve its biopharmaceutical profile. The inclusion complexes were evaluated by means of thermoanalytical methods (TG-thermogravimetry/DTG-derivative thermogravimetry/HF-heat flow), powder X-ray diffractometry (PXRD), universal-attenuated total reflectance Fourier transform infrared (UATR-FTIR), UV spectroscopy and saturation solubility studies. Job’s method was employed for the determination of the stoichiometry of the inclusion complexes, which was found to be 2:1 for both guest-host systems. Molecular modeling studies were carried out for an in-depth characterization of the interaction between drug substance and cyclodextrins (CDs). The physicochemical properties of the supramolecular systems differ from those of RSP, demonstrating the inclusion complex formation between drug and CDs. The RSP solubility was enhanced as a result of drug encapsulation in the CDs cavity, the higher increase being obtained with DM-beta-CD as host; the guest-host system RSP/DM-beta-CD can thus be a starting point for further research in developing new formulations containing RSP, with enhanced bioavailability.

Synthesis, biological evaluation and docking study of a new series of di-substituted benzoxazole derivatives as selective COX-2 inhibitors and anti-inflammatory agents

A new series of substituted-N-(3,4-dimethoxyphenyl)-benzoxazole derivatives 13a-13p was synthesized and evaluated in vitro for their COX (I and II) inhibitory activity, in vivo anti-inflammatory and ulcerogenic potential. Compounds 13d, 13h, 13k, 13l and 13n exhibited significant COX-2 inhibitory activity and selectivity towards COX-2 over COX-1. These selected compounds were screened for their in vivo anti-inflammatory activity by carrageenan induced rat paw edema method. Among these compounds, 13d was the most promising analogs of the series with percent inhibition of 84.09 and IC50 value of 0.04 mM and 1.02 mu M (COX-2 and COX-1) respectively. Furthermore, ulcerogenic study was performed and tested compounds (13d, 13h, 13k, 13l) demonstrated a significant gastric tolerance than ibuprofen. Molecular docking study was also performed with resolved crystal structure of COX-2 to understand the binding mechanisms of newly synthesized inhibitors in the active site of COX-2 enzyme and the results were found to be concordant with the biological evaluation studies of the compounds. These newly synthesized inhibitors also showed acceptable pharmacokinetic profile in the in silico ADME/T analyses. (C) 2018 Elsevier Ltd. All rights reserved.
