Triptycene-containing poly(benzoxazole-co-imide) membranes with enhanced mechanical strength for high-performance gas separation

Herein, a series of novel triptycene-containing thermally rearranged poly(benzoxazole-co-imide) membranes (TPI-PBOs) with enhanced mechanical properties and gas separation performance are reported. The effects of chemical structures and poly(benzoxazole-co-imide) composition on the fractional free volume, polymer chain packing, microcavity size and size distribution, mechanical properties, and gas transport properties have been comprehensively investigated. Due to the incorporation of triptycene-containing non-TR-able polyimide segments, the resulting poly(benzoxazole-co-imide) films exhibited good mechanical properties even after being treated at 450 degrees C. The incorporation of bulky and rigid triptycene units led to significantly improved fractional free volume and gas transport properties compared to previously reported poly(benzoxazole-co-imide)s. Several triptycene-containing poly(benzoxazole-co-imide) films displayed excellent gas separation performance for H-2/CH4, H-2/N-2, and CO2/CH4 gas pairs that exceeded the 2008 upper bounds.

Novel SIRT1 activator MHY2233 improves glucose tolerance and reduces hepatic lipid accumulation in db/db mice

The NAD(+)- dependent deacetylase SIRT1, which is associated with the improvement of metabolic syndromes, such as type 2 diabetes, is a well-known longevity-related gene. Several in vitro and in vivo studies have shown the known protective effects of SIRT1 activators, such as resveratrol and SRT1720, on diabetes-or obesity-induced fatty liver and insulin resistance. Here, we newly synthesized 18 benzoxazole hydrochloride derivatives based on the structure of resveratrol and SRT1720. We performed an in vitro SIRT1 activity assay to identify the strongest SIRT1 activator. The assay confirmed MHY2233 to be the strongest SIRT1 activator (1.5-fold more potent than resveratrol), and docking simulation showed that the binding affinity of MHY2233 was higher than that of resveratrol and SRT1720. To investigate its beneficial effects, db/db mice were orally administered MHY2233 for 1 month, and various metabolic parameters were assessed in the serum and liver tissues. MHY2233 markedly ameliorated insulin signaling without affecting body weight in db/db mice. In particular, the mRNA expression of lipogenic genes, such as acetyl CoA carboxylase, fatty acid synthase, and sterol regulatory element-binding protein, which increased in db/db mice, decreased following oral treatment with MHY2233. In conclusion, the novel SIRT1 activator MHY2233 reduced lipid accumulation and improved insulin resistance. The compounds nPF(3)PF(3)Bx show enantiotropic mesophases with mesophase ranges of 0-40 degrees C and 0-63 degrees C on heating and cooling for hydrogen-terminated derivatives (nPF(3)PF(3)BH), 43-93 degrees C and 54-123 degrees C for methyl-terminated ones (nPF(3)PF(3)BM), 60-108 degrees C and 74-152 degrees C for nitro terminated ones (nPF(3)PF(3)BN), respectively. They exhibit photoluminescence emission peaks at 390-392 nm and UV-vis absorption bands with maxima at 327-330 nm, respectively. Identification and Characterization of a New Series of Ghrelin O-Acyl Transferase Inhibitors

Ghrelin O-acyl transferase (GOAT; MBOAT4) catalyzes O-acylation at serine-3 of des-acyl ghrelin. Acyl ghrelin is secreted by stomach X/A-like cells and plays a role in appetite and metabolism. Therefore, GOAT has been expected to be a novel antiobesity target because it is responsible for acyl ghrelin production. Here, we report homogeneous time-resolved fluorescence (HTRF) and enzyme-linked immunosorbent assay (ELISA) methods utilizing human GOAT-expressing microsomes as a novel high-throughput assay system for the discovery of hit compounds and optimization of lead compounds. Hit compounds exemplified by compound A (2-[(2,4-dichlorobenzyl)sulfanyl]-1,3-benzoxazole-5-carboxylic acid) were identified by high-throughput screening using the HTRF assay and confirmed to have GOAT inhibitory activity using the ELISA. Based on the hit compound information, the novel lead compound (compound B, (4-chloro-6-{[2-methyl-6-(trifluoromethyl)pyridin-3-yl]methoxy}-1-benzothiophen-3-yl)acetic acid) was synthesized and exhibited potent GOAT inhibition with oral bioavailability. Both the hit compound and lead compound showed octanoyl-CoA competitive inhibitory activity. Moreover, these two compounds decreased acyl ghrelin production in the stomach of mice after their oral administration. These novel findings demonstrate that GOAT is a druggable target, and its inhibitors are promising antiobesity drugs.
